In grapevine, the influence of the plant’s elemental composition (the ionome) on fruit and wine quality is well established, but its genetic underpinnings have received limited scientific attention. In this study, I analyzed the leaf ionome of 131 interspecific F1 hybrid progeny plants from a cross between a Vitis rupestris Scheele (♀) and a Vitis riparia Michx (♂) parents, which were replicated in four different environments: Southwest Missouri, Central Missouri, South Dakota, and New York. I sampled leaves at three different times during the growing season at all four locations and had the concentration of 20 elements measured using ICP-MS. …

The North American grapes species Vitis rupestris Scheele and Vitis riparia Michx have been the pillars of rootstock breeding for many decades. Though a large body of viticultural knowledge has been accumulated on their impact on grafted scions, the genetic basis of their root system architecture (RSA) has received limited scientific attention. In this study, I generated and analyzed adventitious root systems from dormant cuttings of 22 V. riparia and 19 V. rupestris accessions, as well as 162 interspecific F1 hybrid progeny from a cross between V. rupestris (♀) and V. riparia (♂). Bacterial panicle blight (BPB) and sheath blight (SB) are major diseases of rice causing significant yield reduction in conducive conditions. Few cultivars have shown reliable resistance to these diseases. In this study, a recombinant inbred line (RIL) population derived from the U.S. cultivars Jupiter (moderately resistant) and Trenasse (susceptible) was investigated to identify genomic regions associated with BPB and SB resistance. Phenotypes in BPB and SB, as well as days to heading (DH), of the RIL population were evaluated in the field. DH was found to be correlated to BPB and SB diseases. Stagonospora nodorum blotch (SNB) is an economically important wheat disease caused by the necrotrophic fungus Parastagonospora nodorum. SNB resistance in wheat is controlled by several quantitative trait loci (QTLs). Thus, the identifying of novel resistance/susceptibility QTLs is crucial for continuous improvement of the SNB resistance. Peach: Peaches [Prunus persica (L.) Batsch] are routinely chilled to increase shelf-life. Exposure to temperatures of 5° C for two weeks can induce chilling injury (CI) symptoms, including flesh mealiness (or wooliness) and a lack of juiciness. Phenotypic data were collected on seven biparental F1 peach populations maintained at the University of Arkansas Fruit Research Station. A genome wide association study (GWAS) was performed using TASSEL 5 which identified four quantitative trait loci (QTLs) associated with expressible juice, four QTLs for mealiness, five QTLs for soluble solids, and three QTLs for fruit weight. Bacterial spot, caused by the bacterium Xanthomonas arboricola pv. pruni (Xap), is a threat to the peach [Prunus persica (L.) Batsch], Japanese and European plum (P. salicina L. and P. domestica L.), and tart and sweet cherry (P. cerasus L. and P. avium L.) industries. Flowering time and seed dormancy are two adaptive traits for flowering plants. Fusarium head blight (FHB), caused by the fungus Fusarium spp., is a disease that attacks several grass species, including wheat. Wheat variety AGS 2060 has remained moderately susceptible to FHB and is regularly found in pedigrees of resistant breeding lines. Quantitative trait loci (QTL) mapping was conducted to find sources of resistance to FHB in AGS 2060. Linkage maps and quantitative trait loci (QTL) analysis have become essential tools for the positional cloning of agronomically important genes and for marker-assisted breeding. In this study, two North American grape species, Vitis rupestris and Vitis riparia, and their 294 F1 progeny were used to construct parental linkage maps and to perform QTL analysis for downy mildew resistance. Single nucleotide polymorphism (SNP) discovery was accomplished using genotyping-by-sequencing (GBS) and resulted in 348,888 SNPs. Drought is a major challenge in rice production system worldwide. We conducted a phenotypic screening of the USA rice genotypes for drought tolerance and assessed genetic diversity using SSR markers. Identification of QTLs for drought tolerance during both vegetative and reproductive stage was done using genotyping by sequencing (GBS) based saturated SNP linkage map. The rice genotypes Jes, Leah, Roy J, Jazzman, and Madison were found to be drought tolerant. The main ingredients of chocolate are usually cocoa powder, cocoa butter, and sugar. Both the powder and the butter are extracted from the beans of the cacao tree (Theobroma cacao L.). The cocoa butter represents the fat in the beans and possesses a unique fatty acid profile that results in chocolate’s characteristic texture and mouthfeel. Here, we used a linkage mapping population and phenotypic data of 3,292 samples from 420 progeny which led to the identification of 27 quantitative trait loci (QTLs) for fatty acid composition and six QTLs for fat content. Global temperature increases will affect Fusarium head blight (FHB) levels in wheat (Triticum aestivum L.). A pressing question is whether current sources of resistance will be effective in a warmer environment. We evaluated phenotypic response to disease in 238 soft winter wheat breeding lines and cultivars grown in 2015–2016 and 2016–2017 under control and warmed (+3 °C) conditions. Warming was achieved with heating cables buried 3 cm in the rhizosphere. Seed dormancy (SD) is an adaptive trait of both ecological and agricultural importance. Cereal crops have been selected for reduced SD to promote germination, which also caused the pre-harvest sprouting (PHS) problem in crop production. The previous research identified a set of quantitative trait loci (QTL) for SD, including qSD1- 2 and qSD8, in the conspecific weedy and cultivated rice (Oryza sativa). qSD1-2 carries a dormancy-enhancing allele from the cultivated rice and has a pleiotropic effect on plant height. qSD8 carries a dormancy-enhancing allele from the weedy rice and tightly links to qAL8 for awn length. Rice (Oryza sativa L.) kernel fissuring is a major concern of both rice producers and millers. Fissures are small cracks in rice kernels that increase breakage of kernels when milled, and decrease the value of processed rice. This study employed molecular gene tagging methods to fine-map a fissure resistance (FR) locus found in ‘Cybonnet’, a semidwarf tropical japonica cultivar, as well as transfer this trait to rice genotypes of taller, non-semidwarf plant height that are better adapted to some rice production systems. Seed carbohydrate content is an important aspect in breeding for food-grade soybeans commercialized in the soyfood market. Sucrose and stachyose are the primary carbohydrates in soybean seed. Sucrose affects the quality and taste of various soyfoods such as tofu, soymilk, and natto; however, consumption of soy-based products with high stachyose concentrations can cause diarrhea and flatulence. Sucrose is a desirable sugar in soybean seed that affects the quality and taste of soyfoods, while stachyose is a non-digestible sugar that induces flatulence in non-ruminant animals. Sweet sorghum, a botanical variety of sorghum is a potential source of bioenergy because high sugar levels accumulate in its stalks. The objectives of this study were to explore the global diversity of sweet sorghum germplasm, and map the genomic regions that are associated with bioenergy traits. In assessing diversity, 142 sweet sorghum accessions were evaluated with three marker types (SSR, SRAP, and morphological markers) to determine the degree of relatedness among the accessions. The traits measured (anthesis date [AD], plant height [PH], biomass yield [BY], and moisture content [MC]) were all significantly different (P< 0.05) among accessions. Morphological marker clustered the accessions into five groups based on PH, MC and AD. The three traits accounted for 92.5% of the variation. There were four and five groups based on SRAP and SSR data respectively classifying accessions mainly on their origin or breeding history. The observed difference between SSR and SRAP based clusters could be attributed to the difference in marker type. SSRs amplify any region of the genome whereas SRAP amplify the open reading frames and promoter regions. Comparing the three marker-type clusters, the markers complimented each other in grouping accessions and would be valuable in assisting breeders to select appropriate lines for crossing. In evaluating QTLs that are associated with bioenergy traits, 165 recombinant inbred lines (RILs) were planted at four environments in Nebraska. A genetic linkage map constructed spanned a length of 1541.3 cM, and generated 18 linkage groups that aligned to the 10 sorghum chromosomes. Fourteen QTLs (6 for brix, 3 for BY, 2 each for AD and MC, and 1 for PH) were mapped. QTLs for the traits that were significantly correlated, colocalized in two clusters on linkage group Sbi01b. Both parents contributed beneficial alleles for most of traits measured, supporting the transgressive segregation in this population. Additional work is needed on exploiting the usefulness of chromosome 1 in breeding sorghum for bioenergy.
