Soft red winter wheat (Triticum aestivum L.) is both an economically and nutritionally important crop in the global food supply. Despite its global importance, wheat is continually threatened by fungal diseases. Fusarium head blight (FHB), caused by Fusarium graminearum, is a devastating fungal disease affecting wheat worldwide, resulting in low yields, reduced test weight and mycotoxin production. The mycotoxin deoxynivalenol (DON) is toxic to humans, results in feed rejection in livestock, and is heavily monitored in the food supply. Efforts to control downy mildew disease continue to be carried out in collaboration with various parties in different ways, including genetic improvement of the varieties planted. Mosaic is a potentially devastating disease of sugarcane that had severe economic impact on Louisiana’s sugarcane industry in the 1920’s and has caused periodic problems ever since. The disease is vectored by migrating aphids that transmit Sugarcane mosaic virus (SCMV) and Sorghum mosaic virus (SrMV) in a non-persistent manner. Mosaic in Louisiana is currently caused by strains of SrMV. Mosaic has been controlled primarily by developing resistant cultivars. Recently, mosaic was detected in some near-commercial clones in the cultivar selection program. Spinach (Spinacia oleracea) is a self-pollinated, dioecious winter crop. Prevalent challenges to the production of spinach include disease pressure imposed by downy mildew, which is caused by Peronospora effusa (=P. farinosa f. sp. spinaciae [Pfs]). A total of 19 new races of P. effusa have emerged, imposing serious challenges to the disease management in spinach production. Accordingly, this study was designed to explore the genetic components for establishing the basis of durable disease resistance development against the downy mildew pathogen (P. effusa 13) in spinach, through the use of various genome engineering approaches.

Aspergillus flavus is a soil-borne fungal pathogen that infects maize and produces aflatoxins. In the current study, portions of the alkaline protease (alk) and the O-methyl transferase (omtA) genes, which are key in A. flavus virulence and aflatoxin biosynthesis, respectively, were targeted for suppression through an RNAi (RNA interference) approach known as Host-Induced Gene Silencing (HIGS). Separate RNAi vectors were designed to carry regions of the alk and omtA gene fragments (Alk-RNAi and OmtA-RNAi) and introduced into B104 maize zygotic embryos. Eight and six transformation events were positive for the alk and omtA transgene, respectively. …

Bacterial spot, caused by the bacterium Xanthomonas arboricola pv. pruni (Xap), is a threat to the peach [Prunus persica (L.) Batsch], Japanese and European plum (P. salicina L. and P. domestica L.), and tart and sweet cherry (P. cerasus L. and P. avium L.) industries. Markers for fruit resistance to bacterial spot have been developed however, markers associated with foliar resistance have yet to be developed. Black walnut anthracnose, caused by Gnomonia leptostyla, is the most widespread and destructive disease affecting black walnut trees (Juglans nigra). Breeding cultivars for a higher resistance to anthracnose is a natural and efficient strategy for improving the health and production quality of black walnut trees. The two goals of this study were to reveal that the ‘Sparrow’ cultivar of black walnut contains a significantly higher resistance to anthracnose than the ‘Football’ cultivar when separated from environmental factors, and to expand the ‘Football’ × ‘Sparrow’ F1 mapping population to evaluate how the trait of resistance is inherited in the progeny. Linkage maps and quantitative trait loci (QTL) analysis have become essential tools for the positional cloning of agronomically important genes and for marker-assisted breeding. In this study, two North American grape species, Vitis rupestris and Vitis riparia, and their 294 F1 progeny were used to construct parental linkage maps and to perform QTL analysis for downy mildew resistance. Single nucleotide polymorphism (SNP) discovery was accomplished using genotyping-by-sequencing (GBS) and resulted in 348,888 SNPs. Of these, 11,063 informative SNP markers (3.17% of the original SNP dataset) were derived after filtering for various quality parameters and missing data. A two-way …

Glyceollin phytoalexins are the pathogen-elicited defense metabolites that belong to the isoflavonoid family of molecules of soybean (Glycine max L. Merr). Phytophthora root and stem rot of soybean caused by Phytophthora sojae is a destructive disease throughout the soybean-growing regions worldwide causing devastating economic damages (globally $1-2 billion and over $250 million in the USA) every year. The identification and quantification of the resistance is difficult, and very little is known about the genetics and markers linked to the charcoal rot (CR) resistance genes. This is a serious fungal disease that can result in a loss of 50% or more of a strawberry harvest when grown in infested soil. Soybean mosaic virus (SMV) causes the most serious viral disease in soybean worldwide. Seven SMV strains, G1 - G7, and three independent multi-allelic loci for SMV resistance, Rsv1, Rsv3, and Rsv4, have been identified. In the initial study, 299 soybean germplasm lines were genotyped for Rsv4 region, inoculated with SMV-G1 and G7 strains, and classified into several resistance groups. The Glyma.02g121400 locus was sequenced from ten soybean accessions, and alignment of the sequences revealed three SNPs displaying 100% polymorphic consistency when a soybean genotype carrying the Rsv4 gene was present. Yellow spot (syn. tan spot), caused by Pyrenophora tritici-repentis, is an important foliar disease of wheat in Australia that causes losses exceeding 50 % when conditions are favourable for disease development. Although good progress has been made internationally to understand yellow spot resistance, relatively few resistance genes have been identified and mapped in Australian germplasm and only one (tsn1 on chromosome 5BL) is in general and known use in Australian breeding programs. Sources of combined resistance to spotted alfalfa and pea aphids were found in Ranger alfalfa and its parental strains, foreign plant introductions, clones and their progenies. Ranger alfalfa appeared to be a good source for selection of plants with resistance to both aphids.
