Characterization Of Wound-Inducible Genes Encoding Enzymes For Terpenoid Biosynthesis In Medicago Truncatula, Mandy M. Cox

In addition to having numerous applications and pharmaceuticals, terpenoids are an important class of defensive compounds that can accumulate in plants after pathogen infection or injury by insects. Sequences of DNA encoding putative terpene synthases and an oxidosqualene synthase, isolated from insect-damaged Medicago truncatula leaves, were selected from an expressed sequence tag (EST) data base. The cDNA clones were used as radio-labeled probes to analyze gene expression in leaves treated with factors known to trigger a defense response in plants. Transcript levels for all of the genes examined increased in response to artificial wounding, insect herbivory, and methyl jasmonate (meJA) …

Search of international DNA database, Genbank, produced eight barley cDNA EFlα sequences with unique accession numbers. The length of these sequences varied from an incomplete 502 bp to a full length 1646 bp encoding a 447 amino acid protein. The knowledge that there are four EF-lα gene family members in barley led us to query whether all four genes were represented in the group of eight database submitted sequences. Single-nucleotide polymorphisms (SNPs) provide an abundant source of DNA polymorphisms in a number of eukaryotic species. Information on the frequency, nature, and distribution of SNPs in plant genomes is limited. Thus, our objectives were (1) to determine SNP frequency in coding and noncoding soybean (Glycine max L. Merr.) DNA sequence amplified from genomic DNA using PCR primers designed to complete genes, cDNAs, and random genomic sequence; (2) to characterize haplotype variation in these sequences; and (3) to provide initial estimates of linkage disequilibrium (LD) in soybean. Comparative Studies Of Evolutionary Responses To Light Environments In Arabidopsis, Massimo Pigliucci, Heidi Pollard, Mitchell B. Cruzan

In this article, we compare the reaction norms to foliage shade (changes in light quality, spatially fine-grained environmental variation) and photoperiod (day length, spatially coarse-grained environmental variation) in several haplotypes of Arabidopsis thaliana from Scandinavia. We found that both across-environment means and phenotypic plasticities evolved continuously and very rapidly within this group. Both character means and trait plasticities were highly integrated, in part as predicted by the adaptive plasticity hypothesis for response to foliage shade (the so-called shade-avoidance syndrome). Corn and grain sorghum performance tests are conducted each year in Arkansas by the University of Arkansas Division of Agriculture. The tests provide information to companies marketing seed within the state and aid the Arkansas Cooperative Extension Service in formulating recommendations for producers. The 2002 corn performance tests contained 59 entries and were conducted at the Northeast Research and Extension Center (NEREC) at Keiser, the Cotton Branch Station (CBS) near Marianna, the Bell Farming Company (BFC) near Des Arc, the Southeast Research and Extension Center - Rohwer Division (SEREC-RD) near Rohwer, and the John Williams Farm (JWF) in Lafayette County. …

Cotton yields in Arkansas increased steadily during the eighties, but in recent years there has been a leveling off. Of more significance, however, is that extreme year-to-year variability in yields has occurred in the last decade, which is a major point of concern with cotton producers. It has been suggested that this may be related to extreme weather conditions during the boll development period in July and August. Average maximum temperatures in the 2001 season were a few degrees above normal. Pseudomonas syringae uses a type III protein secretion system encoded by the Hrp pathogenicity island (Pai) to translocate effector proteins into plant cells. One of these effector proteins is HopPsyA. A small open reading frame (ORF), named shcA, precedes the hopPsyA gene in the Hrp Pai of P. s. syringae 61. The predicted amino acid sequence of shcA shares general characteristics with chaperones used in type III protein secretion systems of animal pathogens. A functionally non-polar deletion of shcA in P. s. syringae 61 resulted in the loss of detectable HopPsyA in supernatant fractions, consistent with ShcA acting as a …

In order for agricultural biotechnology (agri-biotech) to play a larger role in the development of sustainable agricultural systems, intellectual property (IP) rights management must be addressed. These issues are not limited to developing countries. With increased globalization, the management of agri-biotech IP rights affects both developing and industrialized countries. In industrialized countries, for example, IP rights risk management entails protection of inventions via strong patent portfolios. For developing countries, IP rights risk management includes the acquisition of rights requisite for the use of inventions essential to the basic welfare of the population. The ability of Pseudomonas syringae pv. tomato DC3000 to parasitize tomato and Arabidopsis thaliana depends on genes activated by the HrpL alternative sigma factor. To support various functional genomic analyses of DC3000, and specifically, to identify genes involved in pathogenesis, we developed a draft sequence of DC3000 and used an iterative process involving computational and gene expression techniques to identify virulence-implicated genes downstream of HrpLresponsive promoters. Hypersensitive response and pathogenicity (Hrp) promoters are known to control genes encoding the Hrp (type III protein secretion) machinery and a few type III effector proteins in DC3000. This document has been prepared from a cauliflower variety evaluation trial conducted at the Manjimup Horticultural Research Institute, South West Highway, Manjimup WA 6258. The results reflect the growth characteristics, yield, quality and density of the cauliflower varieties as recorded at this site during the conditions of the growing period (May 2002 to September 2002).

This report is the thirty-fourth in a series produced from the evaluation of cauliflower and broccoli varieties, conducted at Manjimup Horticultural Research Institute. Cauliflower only has been evaluated in this and in subsequent trials.

Seed dormancy is a major impediment to germination testing in seed testing laboratories. Dormant seeds are alive but do not germinate under conditions favorable for germination of non-dormant seeds of the same species. Dormancy in many p􀊆ant species can be broken using a single ·treatment or a combination of treatments. Treatments commonly used are light or dark, various temperature regimes, prolonged exposure to cold treatments (pre-chilling), and the use of growth hormones or other chemicals. Fertigation is the technique of using soluble fertilisers to supply essential nutrients to vegetables through sprinkler or trickle irrigation systems, or by means of a boom spray. It is ideally suited to most horticultural areas in Western Australia, especially the sandy soils of the Swan Coastal Plain that have a poor capacity to retain nutrients.

Before and after planting, nutrients are often applied to vegetables on soils of the Swan Coastal Plain by applications of poultry manure* or solid fertilisers. After planting, fertigation is a safe and effective method of applying top-dressings of nutrients to crops, especially after the rows …

Clone size of plants of Andropogon gerardii from Konza Prairie Biological Station, Manhattan, Kansas was estimated from spatial patterns of genetic variation, using proteins detected by starch gel electrophoresis and DNA content (ploidy) measured by flow cytometry. Unique multi-locus protein banding patterns and differences in ploidy were used to exclude plants as members of the same clone. Individual clones averaged about 2 m in diameter and areas of prairie of 100 m2 were calculated to contain an average of 31.8 genetic individuals.

Tissue culture and induced mutation techniques were used to generate variation for the improvement of Pisang Bcrangan cv. 'Intan' (AAA). Best field perform ing banana in vitroplants were multiplied and evaluated for better field performance, which included improved agronomic traits such as earliness in fruiting, short plant stature, high yielding capacity with desirable fruit quality and tolerance to Fusarium wilt. A suitable micropropagation medium for Pisang Berangan was established through modification of MS basic medium, which resulted in significant increase in shoot multiplication. The sand prairie is a unique ecosystem in the state of Illinois, being the habitat for many interesting plants and animals not found elsewhere in the state. One such plant is the state-endangered Stylisma pickeringii (Torr. ex M.A. Curtis) Gray var. pattersoni (Fern. & Schub.) Myint (Convolvulaceae), also known as Patterson's bindweed or Patterson's dawn flower. It currently is found in only three Illinois counties—Cass, Henderson, and Mason. Field resistance to cyst nematode (SCN) race 3 (Heterodera glycines I.) in soybean [Glycine max (L.) Merr.] cv ‘Forrest’ is conditioned by two QTLs: the underlying genes are presumed to include Rhg1 on linkage group G and Rhg4 on linkage group A2. A population of recombinant inbred lines (RILs) and two populations of near-isogenic lines (NILs) derived from a cross of Forrest × Essex were used to map the loci affecting resistance to SCN. During this time there has been no comprehensive survey of the industry to determine its size and production base. Isolated populations have potential to become new species that should have less genetic variation than their ancestors. Small populations are more likely to lose genetic variation, which is, thus, expected to be greater in ancestors. Aliciella caespitosa and A. tenuis, two endemic species, may be derived from small populations of A. subnuda, a widespread species. Chloroplast DNA sequences were used to test this hypothesis. Allozyme data were used to compare genetic variation and numbers of alleles. Chloroplast data do not support the proposed relationships between A. subnuda and the other two species. The Canola industry has rapidly grown with a 10 fold increase in area sown in Western Australia over the four years from 1996-99.

The driving force behind this increase in area can be attributed to the successful adoption of weed control technology in Triazine Tolerant Canola.

The purpose of this study was to characterize the variation pattern that exists in the wild Musa acuminata (AA) populations. widely distributed in Malaysia. Fourteen populations of Musa acuminata, which included subspecies malaccensis, siamea and truncata were collected from different locations including Selangor, Pahang, Perak, Kelantan, Kedah and Terengganu for studies on variation within and between populations, based on morphological characters, biochemical, ploidy and genomic DNA content. Techniques of numerical taxonomy were employed to determine the variation pattern and these included use of the clustering method and principal component analysis.
