The bacterial plant pathogen Pseudomonas syringae requires a type III protein secretion system (TTSS) to cause disease. The P. syringae TTSS is encoded by the hrp-hrc gene cluster. One of the genes within this cluster, hrpJ, encodes a protein with weak similarity to YopN, a type III secreted protein from the animal pathogenic Yersinia species. Here, we show that HrpJ is secreted in culture and translocated into plant cells by the P. syringae pv. tomato DC3000 TTSS. A DC3000 hrpJ mutant, UNL140, was greatly reduced in its ability to cause disease symptoms and multiply in Arabidopsis thaliana …

Reduction of the ozone layer results in the increase in ultraviolet radiation reaching the earth's surface, especially the ultraviolet-B (UV-B). The increase of radiation may induce structural and physiological changes in plants, influencing their growth and development. This paper evaluates the effects of ambient UV-B radiation upon to the leaf morphology of Arabidopsis thaliana developed under controlled conditions. The goal of this observational study is to identify productive hybrid hazelnut plants suitable for commercial production. The objectives of this studywere to 1) examine a plantation of hybrid hazelnut plants grown in Southeast Nebraska for the presence of eastern filbert blight (EFB); 2) identify high yielding hybrid hazelnut plants; and 3) characterize plant and nut features of selected high yielding plants.
Data were collected from a 3.64 hectare (9 acre) planting of 5,200 hybrid hazelnut plant located at Arbor Day Farm in Nebraska City, Otoe County, Nebraska. Plants were grown on 1.5 m x 4.6 m (5 foot x …

Sixty-eight 'short day' selections from the Australian Temperate breeding program were compared in a trial planting on a commercial property at Wanneroo from a mid May planting. Thirty-three of the selections were derived from crosses made in 2003 (new Stage 4 selections) and twenty-nine were from crosses made between 1998 and 2002 (Intermediate Stages 5 and 6). In addition, three advanced selections were compared in large plots of 1,000-1 ,500 plants together with three commercial control cultivars, Gaviota, Camino Real and Kiewa. These selections were 'fast tracked' for future commercialisation by being selected for bulking up in September 2004.

The transient assay system based on mesophyll or cultured cell-derived protoplasts has been exploited in several plant species and has become a powerful tool for rapid gene functional analysis and biochemical manipulations. However, the system has not been widely used in rice owing to the difficulties in large-scale isolation of viable rice protoplasts from leaves or suspension-cultured cells. Here, we describe a significantly improved method to isolate a large number of protoplasts from stem and sheath tissues of both young and mature plants. Ca/othamnus sp. Whicher (Myrtaceae) is a narrow range endemic shrub restricted to ironstone soils near the town of Busselton in south-western Australia. Due to extensive land clearing for agriculture and mineral exploration, the species is fragmented over the majority of its range. In the present study, microsatellite markers were used to characterise levels of genetic diversity and describe levels of differentiation and gene flow among seven small, isolated road verge populations. Allelic diversity within the taxon over the six microsatellites was high (A = 17.6 ± 1.6). Diversity within populations was considerably lower (A = 5.19 ± 1.27), …

Efficient regeneration of in vitro cultures of Oryza sativa L.is essential for successful manipulation of recombinant DNA technologies. Arkansas rice varieties perform better on modified Gamborg (B5)media, than on Murashige and Skoog (MS) orN6 media, which are more frequently reported in the literature. While 2,4-dichlorophenoxyacetic acid (2,4-D) is commonly used for regeneration treatments of rice, Picloram (Pic) provides a possible alternative as a synthetic auxin source. The ability to determine the viability of a pollen grain, and optimal conditions for maximum germination and storage are important for hybridisation. Studies were conducted from January to October 2005 to determine the effect of root-cutting length on adventitious shoot yield and the management practices necessary to produce nursery-quality primocane-fruiting blackberry plants. The first portion of the study measured the average number of shoots produced from 7.6 cm- and 15.2 cm-long root cuttings of APF-44 blackberry—a primocane-fruiting genotype from the University of Arkansas breeding program. Cuttings were forced in a shallow bin containing a soilless potting medium. The genus Zephyranthes and the related genus Habranthus in the family Amaryllidaceae are commonly grown ornamental bulbs having new world homologies. Inter-specific and inter-generic hybridizations are complicated by the fact that some of the species are apomictic, cross incompatible, or have widely variable 2n chromosome numbers. A simple, inexpensive method of pollen storage was tested to evaluate the production of hybrid seed. Intact anthers harvested after pollen release and stored at 4°C were used for pollination.
