The primary goal of the Arkansas Cotton Variety Test is to provide unbiased data regarding the agronomic performance of cotton varieties and advanced breeding lines in the major cotton-growing areas of Arkansas. This information helps seed companies establish marketing strategies and assists producers in choosing varieties to plant. These annual evaluations will then facilitate the inclusion of new, improved genetic material in Arkansas cotton production. Sphingolipids are a vital component of plant cellular endomembranes and carry out multiple functional and regulatory roles. Different sphingolipid species confer rigidity to the membrane structure, facilitate trafficking of secretory proteins, and initiate programmed cell death. Although the regulation of the sphingolipid pathway is yet to be uncovered, increasing evidence has pointed to orosomucoid proteins (ORMs) playing a major regulatory role and potentially interacting with a number of components in the pathway, including both enzymes and sphingolipids. Although genome-sequence assemblies are available for a growing number of plant species, gene-expression responses to stimuli have been cataloged for only a subset of these species. Many genes show altered transcription patterns in response to abiotic stresses. However, orthologous genes in related species often exhibit different responses to a given stress. Accordingly, data on the regulation of gene expression in one species are not reliable predictors of orthologous gene responses in a related species. Upland cotton (Gossypium hirsutum L.) is an economically important multi-purpose crop cultivated globally for fibre, seed oil and protein. Cottonseed oil also is naturally rich in vitamin E components (collectively known as tocochromanols), with a- and c-tocopherols comprising nearly all of the vitamin E components. By contrast, cottonseeds have little or no tocotrienols, tocochromanols with a wide range of health benefits. Here, we generated transgenic cotton lines expressing the barley (Hordeum vulgare) homogentisate geranylgeranyl transferase coding sequence under the control of the Brassica napus seed-specific promoter, napin. Aspergillus flavus is a soil-borne fungal pathogen that infects maize and produces aflatoxins. In the current study, portions of the alkaline protease (alk) and the O-methyl transferase (omtA) genes, which are key in A. flavus virulence and aflatoxin biosynthesis, respectively, were targeted for suppression through an RNAi (RNA interference) approach known as Host-Induced Gene Silencing (HIGS). Separate RNAi vectors were designed to carry regions of the alk and omtA gene fragments (Alk-RNAi and OmtA-RNAi) and introduced into B104 maize zygotic embryos. The uses of vegetable oils are determined by functional properties arising from their chemical composition. Soybean oil was previously used in margarines and baked foods after partial hydrogenation to achieve heat and oxidative stability. This process, however, generates trans fats that are now excluded from food use because of cardiovascular health risks. Also present in soybean oil are the anti-oxidant tocopherols, with α- tocopherol (vitamin E) typically present as a minor component compared to γ-tocopherol. Genetic improvement of the fatty acid profile and tocopherol profile is an attractive solution to increase the functional and health qualities of soybean oil. The …

Latinx fresh market vegetable farmers in Minnesota face numerous barriers to market access. Many Latinx farmers live in rural communities where farmers market sales are low, and opportunities for wholesale are limited. The growers who make up Shared Ground Farmers’ Cooperative have developed an innovative cooperative marketing system to aggregate produce and sell it in Twin Cities markets, but many of the individual farmers involved also wish to find ways to keep the produce they grow in their own communities. Objectives: The purpose of this data set is to capture the complete diurnal (i.e., daily) transcriptome of fully expanded third leaves from the C4 panacoid grasses sorghum (Sorghum bicolor), maize (Zea mays), and foxtail millet (Setaria italica) with RNA-seq transcriptome profiling. These data are the cornerstone of a larger project that examined the conservation and divergence of gene expression networks within these crop plants. The lipid matrix in cell membranes is a dynamic, bidimensional array of amphipathic molecules exhibiting mesomorphism, which contributes to the membrane fluidity changes in response to temperature fluctuation. As sessile organisms, plants must rapidly and accurately respond to environmental thermal variations. However, mechanisms underlying temperature perception in plants are poorly understood. We studied the thermal plasticity of membrane fluidity using three fluorescent probes across a temperature range of -5 to 41 °C in isolated microsomal fraction (MF), vacuolar membrane (VM), and plasma membrane (PM) vesicles from Arabidopsis plants. Aragoa, comprising 19 high-altitude North Andean species, is one of three genera in the Plantagineae (Plantaginaceae, Lamiales), along with Littorella and Plantago. Based primarily on plastid data and nuclear ITS, Aragoa is sister to a clade of Littorella + Plantago, but Plantagineae relationships have yet to be assessed using multigene datasets from the nuclear and mitochondrial genomes. Here, complete mitogenomes were assembled for two species of Aragoa (A. abietina and A. cleefii). Background: Biological mutagens (such as transposon) with sequences inserted, play a crucial role to link observed phenotype and genotype in reverse genetic studies. For this reason, accurate and efficient software tools for identifying insertion sites based on the analysis of sequencing reads are desired. Results: We developed a bioinformatics tool, a Finder, to identify genome-wide Insertions in Mutagenesis (named as “InMut-Finder”), based on target sequences and flanking sequences from long reads, such as Oxford Nanopore Sequencing. Selection for yield at high planting density has reshaped the leaf canopy of maize, improving photosynthetic productivity in high density settings. Further optimization of canopy architecture may be possible. However, measuring leaf angles, the widely studied component trait of leaf canopy architecture, by hand is a labor and time intensive process. Here, we use multiple, calibrated, 2D images to reconstruct the 3D geometry of individual sorghum plants using a voxel carving based algorithm. There are three enzymes that play a role in protein prenylation: protein farnesyltransferase (PFT), protein geranylgeranyl-transferase-I (PGGT), and Rab geranylgeranyltransferase (Rab-GGT). Pollen cross-contamination has been a major problem for maize breeders. Various mechanical methods applied to avoid the contaminations are ineffective. The genetic factors related to maize fertilization can be used to develop an effective method to prevent pollen contamination. Pollen rejection ability controlled by Teosinte crossing barrier 1 (Tcb1) is such a genetic system. Silks possessing dominant Tcb1-s reject pollen possessing the recessive allele (tcb1). Successful fertilization occurs when Tcb1-s pollen falls upon tcb1 silks. The efficacy of dominant Tcb1-s was, however, reduced when repeatedly backcross with maize inbred lines, which suggests that there are modifiers to Tcb1-s. To find …

Soybean leaf-associated gemycircularvirus-1 (SlaGemV-1) is a novel mycovirus discovered through the metagenomic sequencing of soybean leaves which is capable of inducing hypovirulence in the highly pathogenic fungus Sclerotinia sclerotiorum. RNASeq analysis techniques were used to determine the transcriptional changes caused by the infection of virus in S. sclerotiorum, as well as the transcriptional changes in Glycine max caused by the colonization of hypovirulent, SlaGemV-1-infected S. sclerotiorum. RNASeq results indicate that viral infection leading to hypovirulence may attenuate expression of genes relating to cell wall synthesis, microtubule formation, and metabolism of steroids and natural antibiotics. Stagonospora nodorum blotch (SNB) is an economically important wheat disease caused by the necrotrophic fungus Parastagonospora nodorum. SNB resistance in wheat is controlled by several quantitative trait loci (QTLs). Thus, the identifying of novel resistance/susceptibility QTLs is crucial for continuous improvement of the SNB resistance. Kernel size is a vital milling characteristic for Avena sativa L. (oats), with larger kernel size being desirable for milling facilities. This project evaluated the effect of genotype, environment, and their genotype by environment interaction (G by E) on kernel width, length, and area for a set of 27 oat cultivars grown at multiple locations in South Dakota over three years. Findings indicated significant differences among oat varieties, and that environment had a substantial effect on kernel size. However, we did not see a strong influence of genotype by environment interaction on kernel size. The lack of impact for genotype …

Crop designing for resilience traits to produce satisfactorily under adverse conditions and scenarios is a priority for the future of global food security. In this thesis, two different approaches addressing major stress factors for tomato cropping are presented. We first aimed to domesticate tomato de novo as a high-salinity resistant crop using the wild relative, S. cheesmaniae via gene editing. The tallgrass prairies of North America are one of the most threatened ecosystems in the world, making efficient species identification essential for understanding and managing diversity. Here, we assess DNA barcoding with high‐throughput sequencing as a method for rapid plant species identification.

Fusarium head blight (FHB) of wheat (Triticum aestivum L.) results in discolored grain contaminated with deoxynivalenol (DON). DON accumulation, an indicator of FHB resistance, can be quantified and used as the basis for direct phenotypic selection, but testing is expensive. The primary objective of this study was to evaluate an optical seed sorter as an alternative to DON testing for FHB resistance breeding. Genomic selection (GS) is a form of marker-assisted selection (MAS) that simultaneously estimates all locus, haplotype or marker effects across the entire genome to calculate genomic estimated breeding values (GEBVs). Since its inception, it has had the attention of breeders keen on finding tools to accelerate genetic gain and reduce phenotyping costs in the breeding program. Premise of research. Numerous biotic and abiotic factors can contribute to local selection and lead to geographic structure and genetic divergence between populations. The southwestern United States contains many distinctive plant communities, ranging from woodlands to desert scrub, that are shaped by species adapting to local variation in elevation, precipitation, seasonality, and soils. Given this variation, species occurring across diverse habitats are expected to harbor high genetic diversity and exhibit significant genetic differences associated with environmental variation.

Establishing genetically diverse communities that can adapt to dynamic selective pressures is crucial in ecological restoration. However, the genetic structure of native plant species used for restoration is often poorly understood. Pseudoroegneria spicata, is a keystone species of the Inland Northwestern US that has become a staple in commercially available restoration seed mixes. It is abundant in remnant prairies of the endangered Palouse Prairie ecoregion, which is characterized by rolling hills of deep loess soil. Peningkatan produksi cabai (Capsicum sp.) dapat dilakukan dengan melakukan diversifikasi jenis baru dari hasil persilangan antar spesies. Tujuan penelitian ialah untuk mempelajari kompatibilitas pada penyerbukan sendiri dan penyerbukan silang buatan interspesies. Penelitian dilaksanakan pada Januari 2020 – Juni 2020. Bahan yang digunakan ialah 4 spesies cabai. Persilangan disusun berdasarkan rancangan perkawinan dialil lengkap. Terdapat 16 kombinasi persilangan. Perbedaan karakter kuantitatif penyerbukan sendiri dan silang diuji menggunakan uji t 5%. Penyerbukan sendiri pada setiap spesies memiliki kompatibilitas penuh dengan persentase pembentukan buah yaitu 100%.
