Screening Of Spore Forming D-Lactic Acid Producing Bacteria And Its Optimization, Budsabathip Prasirtsak

In this study, isolation, screening, fermentation optimization, and determination of the expression of key enzymes involving in D-lactic acid production were conducted for selection of the novel D-lactic acid isolate with the high yield, productivity, and optical purity. From the screening experiment, the novel isolate, NK26-11T, was obtained. The isolate NK26-11T was a Gram-stain-positive, catalase-positive, facultatively anaerobic, spore-forming, rod-shaped bacterium isolated from soil sample in Thailand. This isolate homofermentatively fermented glucose for D-lactic acid and grew at the wide range of temperature between 20 and 45 °C and the pH of 5-8.5. Brackish water reverse osmosis (BWRO) and seawater reverse osmosis (SWRO) membranes were used in the 2-stage reverse osmosis (RO) unit to recover, pre-purify, and pre-concentrate lactic acid. Calcium lactate (CaLAC), sodium lactate (NaLAC), and ammonium lactate (NH4LAC) were used as model feed solutions. The operating pressure showed a pronounced effect on lactate passage through the first BWRO unit, and the Donnan exclusion effect and hydrogen bonding were responsible for cation rejection. Bioactive Peptide Derived From Trypsin Hydrolysis Of Unicellular Cyanobacterium Synechococcus Sp., Rutairat Suttisuwan

A free radicals can promote denaturing reactions in many cellular components, which create oxidative stress and it results to damage in body tissue. The free radicals can causing many diseases including heart diseases, Alzheimer, inflammation and cancer. This research aimed to investigate the effect of bioactive peptides derived from Synechococcus sp. cultured for 21 days on antioxidant, anti-inflammation and anticancer activities. The Synechococcus sp. protein hydrolysate was prepared by trypsin digest and purified by ultrafiltration with molecular weight cut off membranes of 10, 5 and 3 kDa. The MW <3 kDa fraction showed the highest NO, ABTS and DPPH radical scavenging activities with IC50 values of 34.51±9.8 µg protein/mL, IC50 11.54±0.3 µg protein/mL and IC50 13.63±0.15 µg protein/mL, respectively. The F2 fraction from gel filtration chromatography showed the strongest NO and ABTS radical scavenging activities with IC50 values of 7.68±0.64 µg protein/mL and 9.74±0.2 µg protein/mL, respectively. The F2 was purified by RP-HPLC to yield four fractions. The 30 – 40 min sub-fraction (F2-4) was selected for further analysis by mass spectroscopy. Five isolated peptides with amino acid sequences of AILQSYSAGKTK; 1,265.69 Da, ALNKTHLIQTK; 1,265.74 Da, LLVHAPVK; 875.55 Da, IPDAHPVK; 875.48 Da and VVVLRDGAVQQLGTPR; 1,706.97 Da were identified. The F2-4 had higher DPPH and NO radical scavenging activity compared to the synthetic peptide. Moreover, AILQSYSAGKTK had the highest ABTS radical scavenging activity. Furthermore, the F2 fraction protected oxidation-induced DNA damage in pBR322, pKS and pUC19 cells. F2 fraction was selected to study anti-inflammatory and anticancer properties. Anti-inflammatory effect, F2 fraction showed no cytotoxicity toward RAW264.7 macrophage cells. RT-PCR and qRT-PCR results showed that F2 reduced gene expression of pro-inflammatory cytokines iNOS, TNF-α, COX-1, COX-2 and IL-6. For anticancer activity, F2 fraction showed high anticancer activities in the human colon cancer cells (SW620) according to cytotoxic activity (MTT assay), with IC50 values of 106.58±21.46 µg protein/mL. The F2 fraction activated the apoptotic pathway in SW620 cells after treatment for 24, 48 and 72 hours. The highest activities of caspases 3, 8 and 9 were observed after treatment for 72 hours. These findings suggested that unicellular cyanobacterium Synechococcus sp. may be used to develop for natural anti-inflammation and natural anticancer drugs.

Fifteen compounds including two new anthraquinones (118-119), two naturally occurring new compounds (116-117) and eleven known compounds were successfully isolated from the CH2Cl2 extracts of Dracaena cochinchinensis and Eleutherine americana i.e. dihydrochalcones (5 and 23), pterostilbene (27), homoisoflavanones (15 and 82-84), naphthalenes (85 and 116), pyranonaphthoquinone (86) and anthraquinones (99-100 and 117-119). This study has evaluated biological activities of isolated compounds from Tinospora baenzigeri stems collecting from two different areas, and from mangrove-derived endophytic fungi isolated from trees inhabiting in Songkhla province. Six different parts of Manilkara zapota which consisted of barks, flowers, fruits, leaves, roots and seeds were investigated for total phenolic content, total flavonoid content, antioxidant and antityrosinase activities. Methanol crude extract of flowers showed the highest total phenolic content (368.73 ± 0.65 mg GAE/g), while methanol crude extracts of seeds and roots showed high total flavonoid content (90.21 ± 0.57 and 89.03 ± 1.00 mg QE/g, respectively). Methanol crude extract of seeds showed the strongest DPPH (IC50 282.05 ± 0.60 μg/mL) and ABTS (IC50 205.11 ± 0.89 μg/mL) radical scavenging activities and showed the highest FRAP value of 296.46 …

น้ำนมวัวเป็นแหล่งโภชนาการที่สำคัญของมนุษย์ ประกอบไปด้วย โปรตีน วิตามิน แร่ธาตุ และแคลเซียม อย่างไรก็ตามเคซีนซึ่งเป็นโปรตีนหลักที่อยู่ในน้ำนมวัวนั้น ก่อให้เกิดการแพ้ในผู้บริโภคบางรายได้ ซึ่งมีอาการตั้งแต่เบาจนถึงหนัก เช่น หายใจไม่ออก อาเจียน เป็นลมพิษ หรือตาย เป็นต้น ดังนั้นในอาหารที่มีส่วนประกอบของน้ำนมวัว จะต้องมีการระบุแจ้งบนฉลากตามที่กฎหมายได้กำหนดไว้ แต่ในอาหารหลายชนิดไม่มีการระบุดังกล่าว ดังนั้นวิธีตรวจเคซีนอย่างง่ายสำหรับผู้บริโภคที่แพ้ต่อเคซีนจึงเป็นสิ่งจำเป็น ในการศึกษาครั้งนี้มีวัตถุประสงค์เพื่อพัฒนาการตรวจด้วยอิมมูโนวิทยา แบบการไหลในแนวระนาบ (lateral flow immunoassay) ในรูปแบบแถบทดสอบสำหรับการตรวจวัด เคซีนจากวัว โดยในการเตรียมแถบทดสอบจะใช้วิธีการเติมสารต่างๆ ลงบนแถบทดสอบ 2 วิธีคือ การหยดสารละลายลงบนขอบกระจกสไลด์แล้วประทับลงบนเมมเบรน และการจ่ายสารละลายด้วยเครื่องดูดจ่ายสารละลายแบบอัตโนมัติ จากการหาภาวะที่เหมาะสมในการเตรียมแถบทดสอบโดยใช้วิธีการประทับด้วยกระจกสไลด์ในการเติมสารนั้น พบว่า ต้องตรึงเคซีนที่เส้นทดสอบและตรึง IgG-Fc ที่เส้นควบคุมด้วยความเข้มข้น 1 และ 0.25 มิลลิกรัมต่อมิลลิลิตร ตามลำดับ โดยมีค่าคัดออก (cut-off value) อยู่ที่ 20 ไมโครกรัมต่อมิลลิลิตร และพบว่าความเข้มของสีที่ปรากฏบนแถบทดสอบมีความไม่แน่นอน ในการหาภาวะที่เหมาะสมในการเตรียมแถบทดสอบโดยใช้เครื่องดูดจ่ายสารละลายแบบอัตโนมัติในการเติมสาร พบว่าต้องใช้ เคซีน 0.2 ไมโครกรัม และ IgG-Fc 0.0125 ไมโครกรัม ตามลำดับ และใช้อนุภาคทองระดับนาโนที่ติดกับแอนติบอดีที่ความเข้มข้น 300 ไมโครกรัมต่อมิลลิลิตร เติมลงบนส่วนแผ่นคอนจูเกต โดยมีค่าคัดออกอยู่ที่ 20 ไมโครกรัมต่อมิลลิลิตร พบว่าความเข้มของสีที่ปรากฏบนแถบทดสอบมีความแน่นอนมากกว่าวิธีแรก เมื่อนำแถบทดสอบที่เตรียมได้ไปตรวจหาเคซีนจากวัวที่เติมลงในตัวอย่างน้ำนมควาย น้ำนมแพะ และน้ำนมถั่วเหลือง พบว่ามีค่าคัดออกอยู่ที่ 22.5, 9 และ15 มิลลิกรัมต่อมิลลิลิตรตามลำดับ จากการทดลองทั้งหมดสรุปได้ว่า สามารถนำแถบทดสอบไปใช้ตรวจในตัวอย่าง นมควาย นมแพะ และนมถั่วเหลือง ที่มีการปนเปื้อนเคซีนของนมวัวได้

การคัดกรองและลักษณะสมบัติของไลเพสจากแบคทีเรียชอบเค็มปานกลาง, ศุภฤกษ์ เยี่ยมสมบัติ

ในการคัดแยกแบคทีเรียชอบเค็มปานกลางจำนวนทั้งหมด 77 สายพันธุ์ จากตัวอย่างกะปิทั้งหมด 8 ตัวอย่าง ที่เก็บจากตลาดสดมหาชัย จังหวัดสมุทรสาคร จากการคัดกรองสร้างเอนไซม์ไลเพสบนอาหารแข็งที่มีสับเสตรทเป็น Tween 20, 40, 60, 80 และ tributyrin พบว่ามีแบคทีเรียจำนวน 61 สายพันธุ์สามารถผลิตเอนไซม์ไลเพสได้ และมีแบคทีเรียจำนวน 23 สายพันธุ์ สามารถผลิตเอนไซม์ไลเพสได้เมื่อนำมาคัดกรองการผลิตเอนไซม์ไลเพสในอาหารเหลวที่มีสับเสตรทเป็น p-Nitrophenyl butyrate (p-NPB) จึงได้คัดเลือกตัวแทนแบคทีเรียชอบเค็มปานกลางที่มีแอกทิวิตีการผลิตเอนไซม์ไลเพสจำนวน 12 ไอโซเลท และไอโซเลทที่ไม่มีแอกทิวิตีจำนวน 26 ไอโซเลท ในอาหารเหลวมาศึกษาลักษณะทางฟีโนไทป์และวิเคราะห์ลำดับนิวคลีโอไทด์ของยีน 16S rRNA พบว่าสามารถจัดกลุ่มแบคทีเรียตัวแทนที่ย้อมติดสีแกรมบวกทั้งหมด 38 ไอโซเลทได้เป็นทั้งหมด 9 สกุล ได้แก่ สกุล Oceanobaillus (2 สายพันธุ์), Virgibacillus (2 สายพันธุ์), Halobacillus (2 สายพันธุ์), Thalassobacillus (2 สายพันธุ์), Bacillus (5 สายพันธุ์), Staphylococcus (9 สายพันธุ์), Salinicoccus (12 สายพันธุ์), Nesterenkonia (2 สายพันธุ์) และ Allobacillus (2 สายพันธุ์) จากการศึกษาลักษณะอนุกรมวิธานแบบโพลีฟาสิกของ Allobacillus sp. 2 สายพันธุ์ ได้แก่ SKP4-8 และ SKP8-2 จัดเป็นแบคทีเรียสปีชีส์ใหม่ ซึ่งมีผลการวิเคราะห์ลำดับนิวคลีโอไทด์บริเวณยีน 16S rRNA ของสายพันธุ์ SKP4-8 และ SKP8-2 ใกล้เคียงกับสายพันธุ์ Allobacillus halotolerans B3AT เป็น 99.14 และ 98.80 เปอร์เซ็นต์ ตามลำดับ และทั้ง 2 สายพันธุ์ …

Antifungal Genome Mining And Genetics In Filamentous Actinomycete Bacteria Isolated From Local Soils, Jacob Alex, Kevin Lou, Bilal Makhdoom

Actinomycetes are gram positive, filamentous bacteria that produce useful antibiotics, antitumor agents, and agricultural products. A series of enrichments were undertaken to isolate actinomycetes from local soils, varying enrichment media, antibacterials, and soil treatments (including heat and CaCO3). Isolates were characterized by 16S rDNA sequencing, phenotypic and morphological observations, and antibiotic production. The genetic tractability of select isolates was analyzed using a panel of integrating vectors derived from ɸC31, ɸBT1, and OzzyJ phage using intergeneric conjugation. Further, a semi-degenerate multiplex PCR assay to detect ɸBT1 genomic integrants was designed and tested for the first time. Finally, PCR screens were used …

Bawang putih merupakan komoditas penting di Indonesia. Indonesia merupakan negara terbesar pengimpor bawang putih. Permasalahan yang dihadapi dalam teknik produksi benih secara konvensional menjadi salah satu faktor pembatas tidak optimalnya pengembangan dan penyebaran benih tersebut. In all multicellular organisms, the fundamental processes of cell proliferation and cell death are crucial for growth regulation during organogenesis. Strict regulation of cell death is important to maintain tissue homeostasis by affecting processes like regulation of cell number, and elimination of unwanted/unfit cells. The developing Drosophila eye is a versatile model to study patterning and growth, where complex signaling pathways regulate growth and cell survival. However, the molecular mechanisms underlying regulation of these processes is not fully understood. Myo-inositol phosphates (phytates) are important biological molecules produced largely by plants to store phosphorus. Phytate is very abundant in many different soils making up a large portion of all soil phosphorus. This review assesses current phytase science from the perspective of its substrate, phytate, by examining the intricate relationship between the phytate-hydrolyzing enzymes and phytate as their substrate. Specifically, we examine available data on phytate’s structural features, distribution in nature and functional roles. The vast majority of clinical human listeriosis cases are caused by serotype 1/2a, 1/2b, 1/2c, and 4b isolates of Listeria monocytogenes. The ability of L. monocytogenes to establish a systemic listeriosis infection within a host organism relies on a combination of genes that are involved in cell recognition, internalization, evasion of host defenses, and in vitro survival and growth. Wound Healing Post-Mesh Repair ― An Observational Study, Lebbe M.N, Kularajasingham J, Dioso P

This study identified the best surgical mesh repair techniques for inguinal hernia and prevalence of wound healing post-mesh repair. The cross-sectional study design used cluster sampling for data collection. Of the 120 respondents, 48.3% preferred anterior tension-free mesh repair and 49.2% Lichtenstein mesh repair, both identified as the common surgical techniques in eastern Sri Lanka. About 82.5% of the respondents (n = 99) healed while 17% (n = 21) had recurrence of hernia after one month. Nevertheless, 2.5% of the total respondents said that the hernia repaired after one month but less than two months; and 97.5% of the interviewees …

This clinical audit identified the compliance rate of ultrasound users to the standard operating procedures (SOP); and identified the factors that led to a compliance rate of ultrasound human users. The review was conducted for six days in a private radiologic academy in Indonesia. Only 23 subjects selected randomly were audited. Of the 23 participants, there were 11 males and 12 females. Of the 11 males, seven were compliant, and three were non-compliant. Of the 12 females, eight were compliant, and four were non-compliant. This research identified evidence of knowledge and caring attitude among Sri Lankan nurses caring for dying patients; and identified the factors affecting nursing care for the dying in the eastern province of Sri Lanka. A quantitative cohort study design was used having questionnaires as the means of collecting the data. The purposive way of sampling selected 120 staff nurses employed in a government hospital as respondents who falls under the inclusion and exclusion criteria.There was significant evidence of caring attitude towards the dying (p = 0.005534) and knowledge on the end-of-life nursing care (p = 0.0004703) for the dying patients. …

The main goal of this study is to elaborate and evaluate the physicochemical properties of the synthetic biphasic calcium phosphate (BCP) powder: an associate compound of hydroxyapatite (HA): Ca10(PO4)6(OH)2 and beta-tricalcium phosphate (β-TCP): Ca3(PO4)2. The new compound BCP has two advantages: high bioactivity (HA) and fast biodegradation (β-TCP). The obtained powder of BCP was prepared by the precipitate method. XRD analysis confirmed the synthetic material contained both HA and β-TCP crystalline phases. Spermatogenesis is the process by which mature functional spermatozoa are formed, and is initiated in the stem cell niche of the testes. During the post-meiotic stage of spermatogenesis, spermiogenesis, transition proteins facilitate the transformation of chromatin from a histone-based nucleosome structure to a protamine-based nucleosome structure. This study is aimed at analyzing genomic, transcript, and protein sequences of transition proteins in 13 sequenced Drosophila species. The Drosophila melanogaster spermatid specific transition protein-like protein (Tpl94D) was used as the reference sequence in this study.
