The Philippine Agricultural Scientist

Pangi (Pangium edule Reinw) seed is known to be a poisonous material. However, it becomes safe for consumption after processing. This study determined the effect of village-level processing by the Obu Manuvu community of Marilog District, Davao City, Philippines on the antinutrient and nutritional composition of Pangi seeds. Processing methods involved washing with running water for about 15 s, boiling in water for 30 min, and soaking in flowing water in a nearby river overnight. Results showed that after processing, the levels of antinutrients such as cyanogenic glycosides and oxalates were reduced significantly (p < 0.05) by 99.15% and 78.23%, respectively. Further, the tannin content was also significantly reduced by 96.15%. The crude fat content significantly increased (p < 0.05) by about 54%. Crude ash and fiber decreased, while protein did not change significantly (p < 0.05). Minerals such as calcium and zinc increased (p < 0.05) by 221.43% and 64.39%, respectively while the iron and manganese levels remained unaffected. Findings of the study suggested that the processing method by the Obu Manuvu effectively reduces the level of antinutrients, thereby improving the edibility and safety of Pangi seeds. Further, the processing method improves the level of some nutrients, specifically crude fat, calcium, and zinc.

Genetic strategies of insect pest control are receiving increased interest due to recent advances in genetic engineering. By introducing alleles that make males functionally sterile, genetically modified (GM) insects can be used in area-wide pest management programs similar to the Sterile Insect Technique (SIT). Genetic control strategies carry several potential benefits, including improved efficacy and specificity, lower operation costs, and reduced dependence on chemical insecticides, but their use is still controversial despite extensive technical documentation indicating their innocuity to humans and the environment. Extreme weather conditions associated with climate change affect many aspects of plant and animal life, including the response to infectious diseases. Production of salicylic acid (SA), a central plant defence hormone, is particularly vulnerable to suppression by short periods of hot weather above the normal plant growth temperature range via an unknown mechanism. Here we show that suppression of SA production in Arabidopsis thaliana at 28 °C is independent of PHYTOCHROME B (phyB) and EARLY FLOWERING 3 (ELF3), which regulate thermo-responsive plant growth and development. Somatic embryogenesis is the de novo development of asexual embryos because of the plasticity of the plant cell. In tissue culture, the biochemical and genetic mechanisms of dedifferentiated callus tissues can be reprogrammed to transdifferentiate into developed, polarized embryos, which can ultimately regenerate into whole plants. Although this rarely occurs in nature, scientists have exploited this process for decades to regenerate whole plants following gene transformation or for micropropagation. While some species are amenable to in vitro regeneration, upland cotton is particularly recalcitrant, with regenerative potential being confined to only several genotypes. In-vitro meat (IVM) is an alternative that could reduce negative impacts associated with livestock production. The goal of this study was to examine consumers’ preferences for IVM. This study provides a mixed-methods approach in analyzing a potential closed-loop system between renewable biogas production from anaerobic digestion, vermiculture production, aquaculture production, and organic wastes with a particular focus on stillage wastes. Crude oil desalting operations produce an effluent stream which is challenging to treat due to its salt, heavy metal and hydrocarbon content. Consequently, desalter effluent (DE) is usually diluted into other effluent streams and sent to conventional wastewater treatment plants which may lead to upsets the plant operation.

In this study, a novel microbial approach was applied which investigated DE treatment using halotolerant yeast Debaryomyces hansenii (LAF-3 10 U) or the environmentally robust micro-algae Parachlorella kessleri strain CPCC 266. Agriculture and climate change are interconnected processes, with agriculture implicated in rising green house gas emissions, deforestation, soil and water pollution, and reductions in biodiversity. Conversely, changes within ecology (including a warming climate), alter growing conditions for farmers. Farmers face changes in both temperature and precipitation, as well as an increase in adverse weather events that significantly threaten productivity and livelihoods.

However, very little is known about application of STTM technique in tree species. In this study, STTM was applied to knock down microRNA396 (miR396) in transgenic poplar trees for the first time. Expressing dsRNA with sequence complementarity to a vital gene of feeding pests is a strategy that has shown potential. The cattle industry worldwide is ravaged by bovine respiratory disease (BRD), a bacterial disease caused by Mannheimia haemolytica. Recent efforts to design vaccines against M. haemolytica focus on a virulence factor, leukotoxin, in addition to surface lipoproteins. Plant-based protein production is a safe and inexpensive alternative to traditional methods. Edible vaccines deliver antigens to pharyngeal tissues, which can provide local immunization against M. haemolytica prior to its progression into the lungs. The greenhouse whitefly Trialeurodes vaporariorum is an agricultural pest that damages crops by feeding on plant sap and by vectoring plant viruses. Control of whiteflies has been managed through application of insecticides, but this strategy is not entirely effective and alternative control strategies are needed. In this thesis, I evaluated the efficacy of RNA interference as a means to control whiteflies on greenhouse-grown tomatoes. I found that root uptake of dsRNA synthesized from the v-ATPase subunit A gene caused significant gene knock-down and mortality in feeding whiteflies. Strawberry is a well-known source of natural antioxidants with excellent free radical scav- enging capacity. This study determined the effects of chitosan application in field condition on plant growth, fruit yield and antioxidant activities in strawberry fruit. Foliar applications of chitosan on strawberry significantly increased plant growth and fruit yield (up to 42% higher) compared to untreated control. Increased fruit yield was attributed to higher plant growth, individual fruit weight and total fruit weight/plant due to the chitosan application.
