The main aim of this study was to investigate the effects of Portulaca oleracea extract on tumor formation in colon cancer stem cells and chemotherapy sensitivity. In addition, this study analyzed the genetic changes within the Notch signal transduction pathway associated with the effects of the extracts. Serum-free cultures of colon cancer cells (HT-29) and HT-29 stem cells were treated with the chemotherapeutic drug 5-FU to assess sensitivity. UV-A and -B blocking agents are essential components for the formulation of topical cream for psoriasis phototherapy. In this research, many natural compounds were discovered as potent UV-A and UV-B blocking agents, but small number could absorb narrowband UV-B. Quercetin hydrate, oxyresveratrol and curcumin were good UV-A blocking agents, while catechin, hesperetin, lutein and naringenin were proper for UV-B blocking agents. Berberine chloride hydrate, demethoxycurcumin and mansonone G were UV-A and -B blocking agents. From random combination, the mixture of mansonone G 0.010 % w/v and quercetin hydrate 0.0025 % w/v was disclosed to be a right choice for this …

โรคกระดูกพรุนเป็นปัญหาสุขภาพที่สำคัญอย่างหนึ่งของสังคมผู้สูงอายุ สาเหตุหลักของภาวะดังกล่าวเกิดจากความไม่สมดุลของเซลล์ที่ทำหน้าทีสร้างกระดูก (osteoblast) และเซลล์ที่ทำหน้าที่สลายกระดูก (osteoclast) การลดลงของมวลกระดูกในผู้สูงอายุนำมาซึ่งความเสี่ยงในการแตกหักของกระดูก จุดประสงค์ของการศึกษาครั้งนี้ คือ การคัดกรองสารออกฤทธิ์ทางชีวภาพที่มีฤทธิ์ในการต้านการเปลี่ยนสภาพของเซลล์สลายกระดูกและกลไกการออกฤทธิ์ของสารนั้นๆ ในกลุ่มสารทดสอบอนุพันธ์สาร (3S)-1-(3,4-dihydroxyphenyl)-3-hydroxy-7-phenyl-(6E)-6-heptene (DHPH) และ กรดไซเปอเรโนอิก (Cyperenoic acid) ซึ่งสกัดได้จากส่วนเหง้า Curcuma comosa Rox. และราก Croton crassifolius Geiseler. ตามลำดับ มีฤทธิ์แรงในการต้านการเกิดเซลล์สลายกระดูก ในระดับ in vitro ใช้เซลล์จากไขกระดูกที่ถูกกระตุ้นด้วย receptor activator of nuclear factor-κB ligand (RANKL) DHPH มีฤทธิ์ในการยับยั้งการเปลี่ยนสภาพไปเป็นเซลล์สลายกระดูก โดยยับยั้งการเกิด TRAP-positive multinucleated cells (TRAP+ MNCs) ที่ระดับความเข้มข้นยับยั้งกึ่งหนึ่ง (IC50) 325±1.37 นาโนโมลาร์ การยับยั้งดังกล่าวสอดคล้องกับการลดต่ำลงของสัญญาณภายใต้การกระตุ้นของ RANKL โดยรวมถึงวิถี Mitogen-activated protein kinases (MAPKs) เช่น ERK (p44/42) และ p38 แต่ไม่มีผลต่อวิถี NF-κB โดยผลของสารดังกล่าวยังส่งผลกระทบต่อระดับของ transcription factor เช่น nuclear factor of activation T cells (NFATc1) และ c-Fos ซึ่งมีความสำคัญมากในการเปลี่ยนสภาพเป็นเซลล์สลายกระดูก DHPH ลดการกัดกร่อนกระดูก (bone resorption) บนแผ่นกระดูก (bone slices) นอกจากนั้นสาร DHPH ยังสามารถเพิ่มกิจกรรมของเอนไซม์ alkaline phosphatase (ALP) ที่เกี่ยวข้องกับการเปลี่ยนสภาพของเซลล์สร้างกระดูกในเซลล์ชนิด MC3T3-E1 ในกรณีของกรดไซเปอเรโนอิกนั้น พบว่าสามารถลดการเกิด TRAP+ MNCs ที่ระดับความเข้มข้น IC50 ที่ 36.69±1.02 …

The molecular mechanisms by which the endothelial barrier becomes compromised during lipopolysaccharide (LPS) mediated acute lung injury (ALI) are still unresolved. We have previously reported that the disruption of the endothelial barrier is due, at least in part, to the uncoupling of endothelial nitric oxide synthase (eNOS) and increased peroxynitrite-mediated nitration of RhoA. The purpose of this study was to elucidate the molecular mechanisms by which LPS induces eNOS uncoupling during ALI. Drought stress is considered one of the most devastating abiotic stress factors that limit crop productivity for modern agriculture worldwide. There is a large range of physiological and biochemical responses induced by drought stress. Catharanthus roseus (Madagascar periwinkle) is the exclusive source of an array of terpenoid indole alkaloids (TIAs) that are used in the treatments of hypertension and certain types of cancer. TIA biosynthesis is under stringent spatiotemporal control and is induced by jasmonate (JA) and fungal elicitors. Tryptamine, derived from the indole branch, and secologanin from the iridoid branch are condensed to form the first TIA, strictosidine. Biosynthesis of TIA is regulated at the transcriptional level and several transcription factors (TFs) regulating the expression of genes encoding key enzymes in the pathway have been isolated and characterized. This paper uses the composition and abundance of microbial species to analyze soil recovery in disturbed land. Surface mining disturbs ecological communities throughout the world. As organizations seek to reclaim these disturbed lands, a proper analysis of recovery is needed. In previous studies, recovery of disturbed land was limited to surface examinations, which do not characterize the possible unseen devastating effects of the subsoil. Soil microorganisms are extremely sensitive to environmental changes such as strip mining. It is proposed that these microorganisms may serve as better indicators of recovery post disturbance. The CRISPR/Cas9 nuclease system is a powerful and flexible tool for genome editing, and novel applications of this system are being developed rapidly. Here, we used CRISPR/Cas9 to target the FAD2 gene in Arabidopsis thaliana and in the closely related emerging oil seed plant, Camelina sativa, with the goal of improving seed oil composition. We successfully obtained Camelina seeds in which oleic acid content was increased from 16% to over 50% of the fatty acid composition. These increases were associated with significant decreases in the less desirable polyunsaturated fatty acids, linoleic acid (i.e. a decrease from ~16% to <4%) and linolenic acid (a decrease from ~35% to <10%). These changes result in oils that are superior on multiple levels: they are healthier, more oxidatively stable and better suited for production of certain commercial chemicals, including biofuels. The World Health Organization designates lidocaine as an essential medicine in healthcare, greatly increasing the probability of human exposure. Its use has been associated with ROS generation and neurotoxicity. Physiological and metabolomic alterations, and genetics leading to the clinically observed adverse effects have not been temporally characterized. To study alterations that may lead to these undesirable effects, Saccharomyces cerevisiae grown on aerobic carbon sources to stationary phase was assessed over 6 h. Human DJ-1 is a highly conserved and yet functionally enigmatic protein associated with a heritable form of Parkinson’s disease. It has been suggested to be a redox-dependent regulatory scaffold, binding to proteins to modulate their function. Here we present the X-ray crystal structure of the Toxoplasma orthologue Toxoplasma gondii DJ-1 (TgDJ-1) at 2.1-A resolution and show that it directly associates with calcium-dependent protein kinase 1 (CDPK1). The TgDJ-1 structure identifies an orthologously conserved arginine dyad that acts as a phospho-gatekeeper motif to control complex formation. The importance of rhizodeposit C and associated microbial communities in deep soil C stabilization is relatively unknown. Phenotypic variability in plant root biomass could impact C cycling through belowground plant allocation, rooting architecture, and microbial community abundance and composition. We used a pulse-chase 13C labeling experiment with compound-specific stable-isotope probing to investigate the importance of rhizodeposit C to deep soil microbial biomass under two switchgrass ecotypes (Panicum virgatum L., Kanlow and Summer) with contrasting root morphology. Liver fibrosis is awound-healing response to chronic liver injury such as alcoholic/nonalcoholic fatty liver disease and viral hepatitis with no FDA-approved treatments. Liver fibrosis results in a continual accumulation of extracellular matrix (ECM) proteins and paves the way for replacement of parenchyma with nonfunctional scar tissue. The fibrotic condition results in drastic changes in the local mechanical, chemical, and biological microenvironment of the tissue. Liver parenchyma is supported by an efficient network of vasculature lined by liver sinusoidal endothelial cells (LSECs). Copper is an essential element for proper organismal development and is involved in a range of processes, including oxidative phosphorylation, neuropeptide biogenesis, and connective tissue maturation. The copper transporter (Ctr) family of integral membrane proteins is ubiquitously found in eukaryotes and mediates the high-affinity transport of Cu_ across both the plasma membrane and endomembranes. Deriving biofuels and other lipoid products from algae is a promising future technology directly addressing global issues of atmospheric CO2 balance. To better understand the metabolism of triglyceride synthesis in algae, we examined their metabolic origins in the model species, Coccomyxa subellipsoidea C169, using stable isotopic labeling. The ability to directly determine endogenous kinase activity in tissue homogenates provides valuable insights into signaling aberrations that underlie disease phenotypes. When activity data is collected across a panel of kinases, a unique “signaling fingerprint” is generated that allows for discrimination between diseased and normal tissue. Here we describe the use of peptide-based kinase activity sensors to fingerprint the signaling changes associated with disease states. This approach leverages the phosphorylation-sensitive sulfonamido-oxine (Sox) fluorophore to provide a direct readout of kinase enzymatic activity in unfractionated tissue homogenates from animal models or clinical samples. Background: Copepods play a critical role in marine ecosystems but have been poorly investigated in phylogenetic studies. Morphological evidence supports the monophyly of copepods, whereas interordinal relationships continue to be debated. In particular, the phylogenetic position of the order Harpacticoida is still ambiguous and inconsistent among studies. Until now, a small number of molecular studies have been done using only a limited number or even partial genes and thus there is so far no consensus at the order-level.

Chemosensory-related gene (CRG) families have been studied extensively in insects, but their evolutionary history across the Arthropoda had remained relatively unexplored. Here, we address current hypotheses and prior conclusions on CRG family evolution using a more comprehensive data set. In particular, odorant receptors were hypothesized to have proliferated during terrestrial colonization by insects (hexapods), but their association with other pancrustacean clades and with independent terrestrial colonizations in other arthropod subphyla have been unclear. A unique adaptation of many internal parasites of mammals is their ability to stay in the intestine for extended periods of time and resist the normal peristaltic movements and forces that push and expel material. To better understand parasite adhesion behavior and replicate their attachment method in medical devices, an experiment was designed and performed using the rat tapeworm, Hymenolepis diminuta. Catharanthus roseus (Madagascar periwinkle), is a well-known medicinal plant that produces a vast array of terpenoid indole alkaloids (TIAs), including two anticancer compounds vinblastine and vincristine. Industrial scale production of TIAs is hampered by the difficulties of total chemical synthesis of these compounds and the fragmented knowledge on TIA pathway. Using microalgae as a biofuel feedstock has played more attention over a decade. The cultivation of microalgae requires high water use and high initial investment that can make the process still not economically appealing. This study attempted to examine the potential for using brackish shrimp aquaculture effluents as culture medium for algal biomass production. วัตถุประสงค์ของงานวิจัยนี้คือเพื่อหาภาวะที่เหมาะสมในการผลิตบีตากลูแคนจาก Aureobasidium thailandense NRRL 58543 และศึกษาผลกระทบของน้ำหนักโมเลกุลบีตากลูแคนต่อคุณสมบัติพรีไบโอติก และประยุกต์ใช้ในการผลิตเยลลี ปริมาณการผลิตบีตากลูแคนสูงสุด (37.73±0.15 กรัมต่อลิตร) กับความขาวที่ยอมรับได้ (ดัชนีความขาว ≥ 48) ได้รับหลังจากที่เลี้ยง A. thailandense NRRL 58543 ในอาหารสูตร production medium โดยมีโซเดียมไนเตรท ร้อยละ 0.06 (โดยน้ำหนักต่อปริมาตร) เป็นแหล่งไนโตรเจน เสริมด้วยน้ำมันดอกทานตะวันร้อยละ 8.17 (โดยปริมาตรต่อปริมาตร) มีค่า pH เริ่มต้น 6.5 ภายใต้สภาวะการเขย่าที่ 150 รอบต่อนาที อุณหภูมิ 25 องศาเซลเซียส เป็นเวลา 4 วัน บีตากลูแคนที่ได้ถูกนำมาย่อยด้วยเอนไซม์บีตากลูแคนเนสทางการค้า ได้เป็นบีตากลูแคนไฮโดรไลเซทซึ่งประกอบด้วยน้ำตาลกลูโคสและโอลิโกแซ็กคาไรด์ มีน้ำหนักโมเลกุลเฉลี่ย 77,720 ดาลตัน ผลได้ของการย่อยบีตากลูแคนไฮโดรไลเซทด้วยน้ำย่อยเทียมเป็นเวลา 4 ชั่วโมง คือร้อยละ 26.51 ขณะที่บีตากลูแคนดั้งเดิมทนต่อการย่อยได้ นอกจากนั้นยังพบว่าบีตากลูแคนดั้งเดิมที่ 2 มิลลิกรัม คาร์บอน/มิลลิลิตร สามารถกระตุ้นการเติบโตของ Lactobacillus casei และ L. brevis ได้มากกว่า 1.8 เท่า เมื่อเทียบกับบีตากลูแคนไฮโดรไลเซท ภายหลังจากที่บ่มเป็นเวลา 48 ชั่วโมง บีตากลูแคนดั้งเดิมถูกนำมาใช้เป็นส่วนผสมสำหรับการผลิตเยลลี เมื่อนำบีตากลูแคนดั้งเดิมที่ความเข้มข้นสูงสุดร้อยละ 0.5 (โดยน้ำหนักต่อปริมาตร) ผสมร่วมกับเยลลี พบว่าเป็นที่ยอมรับทางประสาทสัมผัสที่ประกอบด้วย สี ความยืดหยุ่น ความเหนียว และความชอบโดยรวม จากผู้บริโภคจำนวน 30 คน เยลลีพรีไบโอติกที่ผลิตได้นี้สามารถเก็บรักษาที่อุณหภูมิห้องได้นาน 7 วัน

ความสม่ำเสมอของขนาดซากไก่เป็นปัจจัยหนึ่งของการเกิดปัญหาไก่ไส้แตก ซึ่งปัญหานี้มักพบในขั้นตอนการนำเอาอวัยวะภายในออก เมื่อเกิดปัญหาไก่ไส้แตกพนักงานในโรงงานผลิตไก่ต้องเสียเวลาทำความสะอาด และเสียเวลา จึงทำให้ต้นทุนการผลิตเนื้อไก่สูงขึ้น โดยปรกติขนาดของซากไก่ถูกควบคุม ด้วยน้ำหนักและอายุของไก่ แต่ในความเป็นจริง ขนาดไก่ มีความสัมพันธ์กับน้ำหนักและอายุ เพียงเล็กน้อย งานวิจัยนี้จึงนำเสนอวิธีการตรวจสอบความสม่ำเสมอของซากไก่ ด้วยวิธีการประมวลผลภาพดิจิทัล โดยงานวิจัยนี้แบ่งออกเป็นสองส่วน ได้แก่ ส่วนแรกนำเสนอวิธีการตัดแบ่งส่วนซากไก่ซึ่งเกิดจากการประมวลผลร่วมกันสามวิธีการ ได้แก่ การใช้แบ่งส่วนภาพด้วยสีการใช้ฟิลเตอร์โครงสร้างส่วนประกอบ และการใช้สัณฐานวิทยาของภาพ โดย งานวิจัยนี้ใช้ 5 ตัวชี้วัด เพื่อเปรียบเทียบประสิทธิภาพของกระบวนการ และงานวิจัยส่วนที่สองเป็นการนำเสนอวิธีการตรวจและสังเกตความสม่ำเสมอของขนาดซากไก่ หลังจากที่ภาพไก่ถูกตัดแยกด้วยกระบวนการแยกภาพซากไก่จากการวิจัยส่วนแรก ซึ่งงานวิจัยนี้ได้ทดสอบวิธีการนำเสอน 4 วิธีการ ซึ่งพบว่า วิธีการที่อยู่บนพื้นฐานของลายเซ็นภาพดิจิทัลทีมีการกำหนด ช่วงขอบเขตการยอมรับ เป็นวิธีการที่เหมาะสมที่สุด สำหรับการตรวจสอบความสม่ำเสมอของภาพซากไก่

โปรตีนสกัดเข้มข้นในใบ (leaf protein concentrate, LPC) เป็นโปรตีนสำคัญที่ใช้เป็นส่วนผสมของอาหารมังสวิรัติ และอาหารเสริม ทั้งในมนุษย์และสัตว์ โดยส่วนใหญ่ LPC ผลิตจากผักและธัญพืช เช่น ถั่วลิสง มันสำปะหลัง และผักบุ้ง ขณะที่ปริมาณรายงานการผลิตโปรตีนจากพืชน้ำมีน้อยโดยเฉพาะจากแหน (duckweed) แหนเป็นพืชน้ำที่โตเร็วและมีปริมาณไนโตรเจนอินทรีย์สูง ซึ่งงานวิจัยครั้งนี้ศีกษาปริมาณโปรตีนทั้งหมด (total protein content, TPC) ในแหนที่พบในประเทศไทย 4 ชนิด ได้แก่ ไข่น้ำ (Wolffia globosa) แหนเป็ดเล็ก (Lemna minor) แหนเป็ดใหญ่ (Spirodela polyrrhiza) และแหนแดง (Azolla pinnata) ด้วยวิธี Kjeldahl พบว่าไข่น้ำมีปริมาณโปรตีนสูงที่สุด ซึ่งเท่ากับร้อยละ 27.45±0.02 (โดยน้ำหนัก) จึงคัดเลือกมาศึกษาสภาวะที่เหมาะสมในการสกัดโปรตีนเข้มข้นจากไข่น้ำโดยการปรับ pH ระหว่าง 3-6 และอุณหภูมิระหว่าง 37-80 องศาเซลเซียส ซึ่งสภาวะที่เหมาะสมในการผลิตโปรตีนเข้มข้นจากไข่น้ำมากที่สุดที่ pH 4 และอุณหภูมิ 80 องศาเซลเซียส โดยผลิตโปรตีนได้สูงสุดร้อยละ 69.96±0.10 (โดยน้ำหนัก) จากนั้นวิเคราะห์องค์ประกอบทางเคมีของโปรตีนเข้มข้นจากไข่น้ำพบว่ามีปริมาณโปรตีนร้อยละ 69.96 (โดยน้ำหนัก), ไขมันร้อยละ 8.96 (โดยน้ำหนัก), คาร์โบไฮเดรตร้อยละ 8.84 (โดยน้ำหนัก), ความชื้นร้อยละ 5.66 (โดยน้ำหนัก), เส้นใยร้อยละ 4.06 (โดยน้ำหนัก) และเถ้าร้อยละ 2.49 (โดยน้ำหนัก) เมื่อวิเคราะห์โลหะหนักในตัวอย่างด้วยวิธี Air-C2H2 Flame Atomic Absorption Spectrometry พบว่า LPC ที่ผลิตได้มีปริมาณ แมงกานีส 2.37 มิลลิกรัม/ลิตร, เหล็ก 2.29 มิลลิกรัม/ลิตร และสังกะสี 2.16 มิลลิกรัม/ลิตร จากการศึกษาชนิดและปริมาณกรดอะมิโนที่ในตะกอนโปรตีน พบว่ามีปริมาณกรดกลูตามิคสูงที่สุดคือร้อยละ 8.07 …

However, liquid biofuel production from lignocellulose is economically challenging due to the difficulties associated with separating the cellulosic sugars from lignin. In addition, the current thermochemical methods to deconstruct lignocellulose typically involve hazardous chemicals that can negatively impact the environment. BACKGROUND: Crohn's disease (CD) is an inflammatory bowel disease (IBD) and has been associated with Mycobacterium avium subspecies paratuberculosis (MAP). MAP has been detected in stool, tissue and blood samples from patients with CD. Gamma interferon (γ-IFN) is an inflammatory cytokine that plays a crucial role in killing intracellular pathogens like MAP, and its receptor (IFNGR1) mutations cause immunodeficiency and severe disseminated mycobacterial infections. The liver is tasked with managing the concentration of various metabolites in the blood, and of particular importance is the uptake of free fatty-acid (FFA), as elevated concentrations of FFA are toxic to cells. FFAs are transported across the cell membrane by CD36 and distributed by LFABP to the endoplasmic reticulum (ER), where they are esterified to glycerol, yielding more chemically inert triglyceride (TAG), which is essential to the process of VLDL assembly. Epstein-Barr Nuclear Antigen 1 (EBNA1), a crucial protein encoded by the Epstein-Barr virus (EBV), plays an important role in the replication and transcriptional regulator of EBV virus latent gene expression. Therefore, it is associated with several diseases especially human malignancies. The previous studies have demonstrated that chalcone derivatives can potentially inhibit the EBV virus. In this study, a series of designed chalcones were in silico screened toward EBNA1 using molecular docking. The docking results show that the interaction energies of 3a, 3b, 3d, 4g and 4h are better than those of the known anti-EBV agents, 1335528, 2122620 and 4-hydroxyderricin. The …

Oxidized carbon nanospheres (OCSs) and graphene oxide sheets (GOShs) were prepared by oxidation of graphite. Oxidized carbon nanotubes (OCTs) was synthesized from single-walled carbon nanotubes. All three shapes of oxidized carbon nanoparticles (OCNs) were used for investigation of their intracellular trafficking in human embryonic kidney cells (HEK293T), mouse macrophage cells (RAW264.7) and human liver cancer cells (HepG2). The results revealed all three shapes of OCNs were internalized into through HEK293T and RAW264.7 cells without endosomal trapping, including localization of all the three OCNs in HEK293T's lysosome. In case of RAW264.7 cells, OCTs and GOShs were localized in lysosomes, only OCSs …
