Oncology-focused biotechnology firms present unique valuation challenges due to high R&D costs, long development timelines, and uncertain clinical outcomes. This thesis addresses such challenges by integrating the clinical development lifecycle with financial valuation models to provide predictive insights into company value. A structured framework is developed that primarily focuses on stage-specific probabilities of success (PoS) for drug candidates and their impact on firm valuation, embedded in techniques such sum-of-the-parts (SOTP) analysis. Intradermal injections of silicone-based fillers for the face and body may cause chronic inflammation and fibrotic reactions in some individuals. Chickens from the UCD 200/206 White Leghorn (WL) line spontaneously develop a fibrotic autoimmune disease known as scleroderma/systemic sclerosis (SSc). Using the SSc-prone UCD and healthy WL chickens, a longitudinal study was conducted to examine the cellular inflammatory responses to intradermal (i.d.) injections of sterile SILIKON-1000. A surrogate is commonly used for process validations. The industry often uses the target log cycle reduction for the test (LCRTest) microorganism (surrogate) to be equal to the desired log cycle reduction for the target (LCRTarget) microorganism (pathogen). When the surrogate is too conservative with far greater resistance than the pathogen, the food may be overprocessed with quality and cost consequences. In aseptic processing, the Institute for Thermal Processing Specialists recommends using relative resistance (DTarget)/(DTest) to calculate LCRTest (product of LCRTarget and relative resistance). High-fiber, low-cost agricultural byproducts offer a sustainable alternative for mitigating the competition for crops between humans and livestock. Pigs predominantly utilize dietary fibers through the process of microbial fermentation within the gut. This study explored the gut microbiota and the capacity for carbohydrate degradation in 30 individual Lantang pigs, a breed indigenous to China. Through metagenomic analysis, a total of 671 metagenome-assembled genomes (MAGs) were assembled and assigned into 14 bacterial and 1 archaeal phylum, including 97 species from uncultured microbes. Introduction: Implementation of gene editing in agriculture and medicine hinges on public acceptance. The objectives of this study were to explore U.S. public opinion about gene editing in agricultural and medical fields and to provide more insight into the relationship between opinions about the safety of gene editing and the potential impact of evidence to improve opinions about safety.

A virus filtration step is integral to the manufacturing of biopharmaceuticals to ensure viral safety. A virus filter is a single-use device that uses a size-based separation process and has a unique structure with minimal defects, so that contaminating virus particles cannot pass through the membrane pores, while therapeutic molecules can. Duchenne muscular dystrophy (DMD) is an X-linked muscle wasting disease caused by mutations in the Dmd gene resulting in non-functional dystrophin. Adeno-associated virus (AAV)-mediated delivery of classical CRISPR systems resulting in double-stranded breaks (DSBs) has yielded promise as a curative DMD therapeutic. However, the consequences of AAV-CRISPR require more thorough characterization. PCR-enriched short-read amplicon sequencing at cut sites is the most common method of sequencing CRISPR editing, but PCR bias results in unreliable quantification of on-target edits. In-vitro meat (IVM) is an alternative that could reduce negative impacts associated with livestock production. The goal of this study was to examine consumers’ preferences for IVM. In an effort to expand the pool of bacterium useful for biotechnology applications, Pseudomonas fluorescens, a common gram negative microbe, was examined for its ability to function in a recombinant setting. P. fluorescens is ubiquitous in nature and was initially identified as a soil bacterium found in dirt and is typically associated with plant material. Gene stacking is a method used in biotechnology by which multiple genes can be placed at a single genomic site, thereby simplifying plant breeding. In this approach, DNA nucleases are used for excising selectable marker genes (SMG), which are the unneeded components of transgenic plants. The goal of this project is to evaluate the effectiveness of the nuclease I-SceI in excising DNA in plants. Growing algae as a source for bio-fuels has become an area of interest due to concerns about global warming and the reliability and ecology of the production of fossil fuels. Dried algae harvested from a pilot water quality improvement technology at the Rockaway Wastewater Treatment Facility in New York were examined as a source of carbohydrates and lipids for the production of bio-fuels in bio-reactors. Propylene glycol dinitrate (PGDN) is an important component used in the formulation of energetic materials. A renewable, environmentally-friendly and effective production technique for the production of 1,2-propanediol, an important precursor to PGDN, is desired. While fermentation of 1,2-propanediol has been achieved, it suffers from major limitations such as expensive feed sugars and relatively low yields. It is hypothesized that algae sugars could serve as a feedstock for the microbial synthesis of 1,2-propanediol, thereby reducing production costs and enhancing sustainability.

Interest in the production of recombinant proteins consisting of collagen binding domain (CBD) fused to a bioactive material has increased due to the targeting/attachment capabilities of CBD. For example, CBD fusions can be applied to the reversing of bone density loss and the repair of the eardrum, specifically, by choosing an appropriate fusion partner (parathyroid hormone or epidermal growth factor). The production of CBD fusions was examined using batch and fed-batch culturing of Escherichia coli to express the fusion proteins, and affinity chromatography to isolate the final product.

In this dissertation, the relationship between the geometry of ion-beam sculpted solid-state nanopores and their ability to analyze single DNA molecules using resistive pulse sensing is investigated. To accomplish this, the three dimensional shape of the nanopore is determined using energy filtered and tomographic transmission electron microscopy. It is shown that this information enables the prediction of the ionic current passing through a voltage biased nanopore and improves the prediction of the magnitude of current drop signals when the nanopore interacts with single DNA molecules. A biosensor is an analytical device that uses biomaterials as elements of the sensing system and converts a biological response into an electrical signal. Biodiesel is a bio-based alternative, biodegradable, renewable, nontoxic diesel fuel made from a chemical reaction between alcohol (usually methanol or ethanol) and plant oil or animal fat. Tests were conducted during the summer of 2009 on a John Deere 3203 diesel tractor to determine differences in specific fuel consumption (sfc), power take-off (PTO) torque (Nm), and PTO power (kW), between ultralow sulfur No. 2 Diesel (D2), 20% biodiesel (B20), 50% biodiesel (B50), and100% biodiesel (B100). Four 1-hr tests were conducted with D2, while three 1-hr tests were conducted with B20, B50, and B100. The results indicated that there was no significant (p < 0.05) difference between D2 and B20 for power or torque. Fueling with B50 resulted in significantly lower power and torque than fueling with D2 or B20, but significantly higher power and torque than fueling with B100. There were significant differences between each fuel in sfc; as the biodiesel blend increased, sfc also increased. Based on these data, B20 appears to be the optimal biodiesel blend for this and similar compact utility tractors since fueling with B20 resulted in no significant loss in power or torque (compared to D2) and only a slight increase in fuel consumption.

The development of bio-fuel synthesis technologies has led to increased interest in woody crops grown specifically for energy production. These woody feedstocks typically involve fast-growing species (e.g., Salix spp., Populus spp.) planted at high densities using short rotations and intensive cultural practices like weed control and fertilization. Under ideal conditions, this type of system can produce 20 dry Mg/ha/yr, which is substantially higher than the 2.5-4 dry Mg/ha/yr produced by pine plantations in the southern U.S. Many of these plantings are projected to be established on lower quality agricultural lands.
