Arthrospira platensis (Spirulina) is a filamentous cyanobacterium valued for its nutritional profile, particularly the pigment–protein phycocyanin, with potent antioxidant and anti-inflammatory properties. Despite its industrial significance, large-scale commercialization remains constrained by high cultivation costs and inefficient extraction methods. This study investigated the use of food-based industrial wastewater as an alternative nutrient medium for Spirulina cultivation, aiming to reduce production costs, enhance phycocyanin yield, and support sustainable wastewater remediation. The research is aimed to study the bioremediation of heavy metal landfill leachate contaminated soil using consortia of indigenous filamentous fungi and indigenous plants. Thirteen identified fungal isolates were used to formulate the consortia for mycoremediation. The mycoremediation was carried out in three phases, namely laboratory trial 1 using five consortia, laboratory trial 2 with varied concentrations of inoculum, and field application. The fungi-assisted phytoremediation was carried out in two phases: laboratory trials using Alocasia calidora, Prosopis juliflora, and Jatropha sp. and field trial using A. calidora. A fully interconnected 3-dimensional scaffold with pre-determined dimensions and porosity can be attained by design-dependent rapid prototyping techniques which possess precise control over the internal and external architecture of the scaffolds thus they can overcome the problems associated with the process-dependent classical approaches. Optimisation Of Biomass And Lipid Production Of A Tropical Thraustochytrid Aurantiochytrium Sp. Umacc-T023 In Submerged-Liquid Fermentation For Large-Scale Biodiesel Production, Sohedein Mohamad Nor Azzimi

A tropical thraustochytrid, Aurantiochytrium sp. UMACC-T023, was optimised for high biomass and lipid production in submerged-liquid fermentation (SLF). Biomass and lipid production were optimised based on glucose, polypeptone, and yeast extract concentration using response surface methodology (RSM). In RSM, the applied central composite design (CCD) showed that the optimisation model was significant for all variables studied. Yeast extract and polypeptone were associated with significant effects in UMACC-T023 biomass production with p < 0.05. The model was validated by employing the optimised media composition in shake flasks and in 1.4-L stirred-tank bioreactors. The optimised media composition for both biomass and lipid production was 10 g L-1 glucose, 15.08 g L-1 polypeptone, and 13.56 g L-1 yeast extract. Biomass production in the bioreactor increased 2.12-fold compared with the shake flask culture utilising the same optimised media composition. This result demonstrated that the optimized media composition can be utilised for large-scale lipid production of UMACC-T023 for potential biodiesel feedstock as it also fulfilled the international standards for biodiesel.

An efficient protocol was developed for rapid propagation and regeneration of the Dioscorea alata L. locally known as ‘ubi badak’ because of their rhinoceros-shaped tubers. In the present study, direct and indirect regeneration were induced to produce plantlets of Dioscorea alata L. For direct regeneration explants (leave, stem and node) were cultured on MS media supplemented with various combinations and concentrations of BAP and NAA to induce microshoots and roots formation. Ciguatera fish poisoning (CFP) is a type of illness that causes human intoxication after consuming contaminated coral fishes. The neurotoxins, ciguatoxins, are produced by species of Gambierdiscus, one of the harmful benthic dinoflagellates. Identification of harmful dinoflagellate species is important to gain further insights into the global dispersion. In this study, morphological characteristics of species in the genus Gambierdiscus and their habitat preferences were investigated. The selectable marker system based on the Escherichia coli phosphomannose isomerase (pmi) gene was adapted for genetic transformation of oil palm. This system makes use of the pmi gene that encodes phosphomannose isomerase, which converts mannose-6-phosphate to fructose-6-phosphate and uses mannose as the selection agent. The aim of this study was to isolate protein/peptide and secondary metabolites from plant and fermented sources. The extracts were used to test for antimicrobial and antioxidant properties. In the current study protein/peptide extracts of Allium sativum showed promising results with a 15mm inhibition zone against Escherichia coli, 28mm against Staphylococcus aureus, 16.3mm against Bacillus cereus and 9mm against Pseudomonas aeruginosa. The minimum bacteriocidal concentration (MBC) results showed that Allium sativum protein/peptide extract is only bacteriostatic and not bacteriocidal. Three derivatives of indole-3-carboxaldehyde with indole-3-acetic hydrazide and their copper(II) and nickel(II) complexes were synthesized. The Schiff base compounds were varied on the substituent where each compound holds a hydrogen, chlorine and bromine atom on the 5th position of indole-3-carboxaldehyde. The structures of the ligands and their complexes were elucidated by various characterization methods. All compounds were screened for their biological effects on neuroprotective screening (in vitro), acute toxicity test (in vivo), ethanol-induced gastric ulcer (anti-ulcer) and glucose tolerance test (antidiabetic). In this study, a simple method using Murashige and Skoog basal medium (1962) supplemented with 6-BA alone was developed for direct multiple shoots induction from shoot base culture of C. willisii. Shoots proliferation and elongation were achieved on a single medium without the need of subculturing. The optimum concentration of 6-BA for direct shoots regeneration of C. willisii was investigated. It was found to be 6 mg/ L. Optimum media was then used for regenerate and propagate of transformed C. willisii. Serotyping is the most fundamental approach in epidemiological surveillance and outbreak investigations for Salmonella. Many multiplex PCR assays have been reported for serotyping Salmonella based on O and H antigens. The aim of this study was to apply and improve the approach in differentiation and serotyping Salmonella enterica strains by combination of sequential multiplex PCRs targeting the O, H and Vi antigens of Salmonella, and this achieved by working on 126 strains. In this study, primary and secondary screenings were conducted to evaluate the antibacterial and anti-yeast potential of the eleven selected marine-derived Streptomyces spp., particularly against P. shigelloides. The strains were categorized into three colour groups for morphological grouping according to the colour of aerial mycelium; namely grey, yellow green, and white colour groups. Diffusible pigments were produced by eight of the strains tested. Strain(s) from grey and white with red diffusible pigments colour groups displayed strong antibacterial activity.
