The hybridization of DNA by sequential electrostatic and hydrogen-bonding immobilization of single-stranded complementary oligonucleotides at the air-water interface with cationic Langmuir monolayers is demonstrated. The complexation of the single-stranded DNA molecules with octadecylamine (ODA) Langmuir monolayers was followed in time by monitoring the pressure-area isotherms. A large (and slow) expansion of the ODA monolayer was observed during each stage of complexation in the following sequence: primary single-stranded DNA followed by complementary single-stranded DNA followed by the intercalator, ethidium bromide. Sphaeriid clams were collected from an ancient, continuously cultivated taro pond complex at Ke‘anae Peninsula, Maui. Both species are known for adaptations for temporary pond habitats and could have been initially introduced by the earliest Hawaiian settlers transported with moist taro root stock from other areas of Oceania.

Gene therapy by direct delivery of plasmid DNA has several advantages over viral gene transfer, but plasmid delivery is less efficient. In vivo electroporation has been used to enhance delivery of chemotherapeutic agents to tumors in both animal and human studies. Recently, this delivery technique has been extended to large molecules such as plasmid DNA. Here, the successful delivery of plasmids encoding reporter genes to rat hepatocellular carcinomas by in vivo electroporation is demonstrated.

Worldwide, the area planted in genetically modified (GM) crops has increased dramatically in recent years. Between 1996 and 1999, it rose from 1.6 X 106 ha to more than 35 X 106 ha (James 1998, May 1999). This rapid increase has provoked an explosion of concern, particularly in Europe, over the health and environmental impacts of these crops. Despite claims of safety and warnings against popular panic, public concern over GM crops has resulted in changes in their marketing, labeling, planting, and trade. 2,3-Butanediol is an important chemical, which is used in the manufacture of products such as synthetic rubber, printing inks, perfumes, fumigants, moistening and softening agents, explosives and plasticizers. It has been known as a bacterial fermentation product since early part of the 20th century. Klebsiella oxytoca Is an organism, which can convert a variety of sugars to 2,3-butanediol. Available literature shows that principal monosaccharides of cellulosic substances as well as major hemicellulosic compounds have been used as carbon source. In this study, the yield of biomass and polyhydroxyalkanoate (PHA) of Pseudomonas oleovorans grown in batch and fed-batch cultivations were compared. When oleic acid (QA) (0.5% w/v) was used as carbon source, a significant increase in biomass.and PHA content were observed through fedbatch cultivation. The biomass and PHA content obtained from batch cultivation were 1.45±0.19 g/I and 2.50±0.77 % cell dry weight (CDW) respectively, while in the fed-batch cultivation, the biomass and PHA content increased significantly to 3.35±0.78 g/I and 32.72±0.30 %CDW respectively. An inexpensive carrier matrix. vermiculite was used for immobilizing urease. A cost effective and simple method of coupling was developed using vermiculite. Immobilization of urease on Amberlite MB-land Nylon-6 tube were also studied. Activities of free and immobilized urease were studied and compared: Among all methods used for immobilization of urease. the method of coupling the enzyme to vermiculite gave the highest retained activity of 82% (Method I) and 89% (Method 2) at pH 6.0 and 5.5 respectively. Urease immobilized by Method I retained 69% and 30% of its original activity and Method 2 (using glutaraldehyde as coupling agent) retained …

Salmonella typhi S8 is resistant to ampicillin (Ac), chloramphenicol (Cm), cotrimoxazole (Ctm), streptomycin (Sm), and tetracycline (Tc). The Ac, Cm, Ctm. and Sm resistance traits have been shown to be mediated by a multiple antibiotic resistance transposon located on a large conjugative plasmid. Transposition of the transposon encoding Ac^R (ampicillin resistance), Cm^R Ctm^R and Sm^R designated TnX8, was done by Wong (1999) to a recipient replicon, plasmid pUB307. The Ac^R Cm^R, Ctm^R and Sm^R genes were subcloned into plasmid pKan by digesting the recombinant plasmid, des ignated pCL8 (pUB307::TnX8), with San. Human herpes virus 6 (HHV6) is an ubiquitous, lymphoproliferative P-herpesvirus, which infects almost all individuals during infancy and persists lifelong. Two variants of HHV6 (A and B) have been described. The present study is on HHV6B, which causes exanthem subitum, also known as roseola infantum, a common febrile childhood illness. Two methods of detection for anti-HHV6B in body fluids were used, being indirect immunofluorescence assay (IFA) and peptidylated pin-ELISA. For IFA, the target cells were HHV68-infected human cord blood mononuclear cells (HCBMC). Ninety eight infant sera were tested by IFA and all were found to be IgG-HHV6 positive. Seventeen breast …

Captopril is a highly specific competitive inhibitor of the angiotensin-convening enzyme (ACE). This enzyme converts angiotensin I, a relatively inactive decapeptide, to angiotensin II , a potent endogenous vasoconstrictor substance. Captopril is widely used for the treatment of hypertension and congestive heart failure. This drug, which contains a sulphydryl group binds readily to albumin and to other plasma proteins. Captopril is unstable in blood and plasma ex vivo because of the reactivity of sulphydryl group which results in oxidation and rapid formation of disulphides. Therefore a fixative or a stabiliser must be added to each blood sample immediately following collection. …

HilA activates the expression of Salmonella enterica serovar Typhimurium invasion genes. To learn more
about regulation of hilA, we isolated Tn5 mutants exhibiting reduced hilA and/or invasion gene expression. In
addition to expected mutations, we identified Tn5 insertions in pstS, fadD, flhD, flhC, and fliA. Analysis of the
pstS mutant indicates that hilA and invasion genes are repressed by the response regulator PhoB in the absence
of the Pst high-affinity inorganic phosphate uptake system. This system is required for negative control of the
PhoR-PhoB two-component regulatory system, suggesting that hilA expression may …

We showed previously that maturation of the human
frataxin precursor (p-fxn) involves two cleavages by the
mitochondrial processing peptidase (MPP). This observation
was not confirmed by another group, however,
who reported only one cleavage. Here, we demonstrate
conclusively that MPP cleaves p-fxn in two sequential
steps, yielding a 18,826-Da intermediate (i-fxn) and a
17,255-Da mature (m-fxn) form, the latter corresponding
to endogenous frataxin in human tissues. The two cleavages
occur between residues 41–42 and 55–56, and both
match the MPP consensus sequence RX ↓ (X/S). Recombinant
