Itch is a common skin symptom, with complex aetiology and pathogenesis. It is mediated by 2 pathways, the histaminergic and non-histaminergic pathways. Chronic itch is understood to be processed by the latter and is difficult to treat with traditional pruritus therapies. The Janus kinase and signal transducer and activator of transcription pathway is a signalling mechanism that regulates gene expression through various cytokines. Janus kinase inhibitors, which have been tested and used for several autoimmune diseases, have also been shown to be effective for itch through clinical trials and case reports. Background: Immune-checkpoint inhibitors (ICIs) are an effective therapeutic strategy, improving the survival of patients with lung cancer compared with conventional treatments. However, novel predictive biomarkers are needed to stratify which patients derive clinical benefit because the currently used and highly heterogenic histological PD-L1 has shown low accuracy. Liquid biopsy is the analysis of biomarkers in body fluids and represents a minimally invasive tool that can be used to monitor tumor evolution and treatment effects, potentially reducing biases associated with tumor heterogeneity associated with tissue biopsies. Seasonal commodity price fluctuations can potentially offer farmers arbitrage opportunities to increase their income. However, smallholder farmers in most of sub-Saharan Africa often do not exploit these opportunities to the fullest extent possible. To inform this issue, we conducted a randomized controlled trial among 1739 smallholder farmers in Malawi to estimate the impact of two key post-harvest constraints, lack of appropriate storage technology and commitment issues, on farmers' legume storage and sales decisions. Dimensional reduction is one of the methods to ensure the quality of a questionnaire. This study examined two methods to reduce the dimension of the questionnaire: multidimensional scaling (MDS) and exploratory factor analysis (EFA). The questionnaire, Awareness of Academic Dishonesty consists of 30 questions. Participants included 110 college students. Multidimensional scaling analysis reduced the multidimensions to essentially two dimensions. The exploratory factor analysis reduced the multidimensions to three dimensions. MDS allowed the researchers to evaluate the questionnaire items by looking at the similarities of these data points. EFA provided an alternative thought about the construct of the questionnaire.

Tulisan ini mengkaji fenomena rendahnya elektabilitas perempuan di Daerah Pemilihan Banten III pada Pemilihan Umum Legislatif tahun 2019. Daerah ini memiliki karakter pemilih rasional dan ramah gender, namun karakter tersebut ternyata tidak memberi pengaruh terhadap elektabilitas perempuan untuk menjadi anggota parlemen. Many recent studies show the development of an increasingly diverse elite characterized by the emergence of a new elite and, on the other hand, the collapse of the old elite. However, these studies mainly infer democratic political change as an explanatory factor. This article takes a different standpoint to explain why some local elites collapse, survive, and emerge as new elites. This study aims to quantify the techno-economic potential of using solar photovoltaics (PV) to support heat pumps (HP) towards the replacement of natural gas heating in a representative North American residence from a house owner's point of view. For this purpose, simulations are performed on: (1) a residential natural gas-based heating system and grid electricity, (2) a residential natural gas-based heating system with PV to serve the electric load, (3) a residential HP system with grid electricity, and (4) a residential HP+PV system. Extended very-high-energy (VHE; 0.1-100 TeV) γ-ray emission has been observed around several middle-aged pulsars and referred to as “TeV halos.” Their formation mechanism remains under debate. It is also unknown whether they are ubiquitous or related to a certain subgroup of pulsars. With 2321 days of observation, the High Altitude Water Cherenkov (HAWC) Gamma-Ray Observatory detected VHE γ-ray emission at the location of the radio-quiet pulsar PSR J0359+5414 with >6σ significance. By performing likelihood tests with different spectral and spatial models and comparing the TeV spectrum with multiwavelength observations of nearby sources, we show that this excess is consistent with …

Multicollinearity Applied Stepwise Stochastic Imputation: A Large Dataset Imputation Through Correlation‑Based Regression, Benjamin D. Leiby, Darryl K. Ahner

This paper presents a stochastic imputation approach for large datasets using a correlation selection methodology when preferred commercial packages struggle to iterate due to numerical problems. A variable range-based guard rail modification is proposed that benefits the convergence rate of data elements while simultaneously providing increased confidence in the plausibility of the imputations. A large country conflict dataset motivates the search to impute missing values well over a common threshold of 20% missingness. The Multicollinearity Applied Stepwise Stochastic imputation methodology (MASS-impute) capitalizes on correlation between variables within the dataset and uses model residuals to estimate unknown values. Machine learning has been increasingly applied to neuroimaging data to predict age, deriving a personalized biomarker with potential clinical applications. The scientific and clinical value of these models depends on their applicability to independently acquired scans from diverse sources. Accordingly, we evaluated the generalizability of two brain age models that were trained across the lifespan by applying them to three distinct early-life samples with participants aged 8-22 years. These models were chosen based on the size and diversity of their training data, but they also differed greatly in their processing methods and predictive algorithms. In this chapter, the use of memory by Indigenous activists and movements in North America to unsettle the status quo by bringing attention to a contested past is reviewed to demonstrate how memory activism has become an integral component of the Indigenous rights movement of the twenty-first century. Centered on awaking the memory of settler-colonial society to unsettle the dominant narrative that pervades the understanding of Indigenous peoples as something of the past serves to spark the settler-colonial state's remembrance that Indigenous people are of the present. This paper reviews the architecture of collaboration that exists within inter-organizational natural resource management (NRM) networks. It presents an integrative conceptual framework designed to help operationalize the multi-level interactions that occur between different dimensions of trust, risk perception, and control as key concepts in inter-organizational collaboration. The objective is to identify and justify a series of propositions considered suitable for assessing inter-organizational NRM network collaboration through empirical work. Marine water temperatures are increasing globally, with eastern Australian estuaries warming faster than predicted. There is growing evidence that this rapid warming of coastal waters is increasing the abundance and virulence of pathogenic members of the Vibrionaceae, posing a significant health risk to both humans and aquatic organisms. Fish disease, notably outbreaks of emerging pathogens in response to environmental perturbations such as heatwaves, have been recognised in aquaculture settings. Considerably less is known about how rising sea surface temperatures will impact the microbiology of wild fish populations, particularly those within estuarine systems that are more vulnerable to warming. Spinocerebellar ataxia type 1 (SCA1) is a paradigmatic neurodegenerative disease in that it is caused by a mutation in a broadly expressed protein, ATXN1; however, only select populations of cells degenerate. The interaction of polyglutamine-expanded ATXN1 with the transcriptional repressor CIC drives cerebellar Purkinje cell pathogenesis; however, the importance of this interaction in other vulnerable cells remains unknown. Here, we mutated the 154Q knockin allele of Atxn1154Q/2Q mice to prevent the ATXN1-CIC interaction globally. This normalized genome-wide CIC binding; however, it only partially corrected transcriptional and behavioral phenotypes, suggesting the involvement of additional factors in disease pathogenesis. Glioblastoma (GBM) is the most common primary intracranial malignant tumor and consists of three molecular subtypes: proneural (PN), mesenchymal (MES) and classical (CL). Transition between PN to MES subtypes (PMT) is the glioma analog of the epithelial-mesenchymal transition (EMT) in carcinomas and is associated with resistance to therapy. CXCR4 signaling increases the expression of MES genes in glioma cell lines and promotes EMT in other cancers. Sushi, von Willebrand factor type A, EGF and pentraxin domain containing 1 (SVEP1) is an extracellular matrix protein that causally promotes vascular disease and associates with platelet reactivity in humans. Here, using a human genomic and proteomic approach, we identify a high affinity, disease-relevant, and potentially targetable interaction between SVEP1 and the orphan receptor Platelet and Endothelial Aggregation Receptor 1 (PEAR1). This interaction promotes PEAR1 phosphorylation and disease associated AKT/mTOR signaling in vascular cells and platelets. Mice lacking SVEP1 have reduced platelet activation, and exogenous SVEP1 induces PEAR1-dependent activation of platelets. SVEP1 and PEAR1 causally and concordantly relate to platelet …

Emerging evidence suggests that Gram-negative bacteria release bacterial outer membrane vesicles (OMVs) and that these play an important role in the pathogenesis of bacterial infection-mediated inflammatory responses and organ damage. Despite the fact that scattered reports have shown that OMVs released from Gram-negative bacteria may function via the TLR2/4-signaling pathway or induce pyroptosis in macrophages, our study reveals a more complex role of OMVs in the development of inflammatory lung responses and macrophage pro-inflammatory activation. We first confirmed that various types of Gram-negative bacteria release similar OMVs which prompt pro-inflammatory activation in both bone marrow-derived macrophages and lung alveolar macrophages. …

This research focuses on the investigation of two machine learning methodologies, Reconstructability Analysis (RA) and Bayesian Networks (BN). Both methods are probabilistic graphical modeling (PGM) methodologies. RA was developed in the systems community and has applications including time-series analysis, classification, decomposition, compression, pattern recognition, prediction, control, and decision analysis. BNs have origins in path models and have applications similar to those of RA. A 38-year-old with Turner syndrome presented with acute myocardial infarction due to multivessel spontaneous coronary artery dissection (SCAD) complicated by left ventricular free wall rupture. Conservative management for SCAD was pursued. She underwent sutureless repair for an oozing-type left ventricular free wall rupture. SCAD has not been previously reported in Turner syndrome. (

Adverse in-utero insults during fetal life alters offspring's developmental trajectory, including that of the cardiovascular system. Gestational hyperandrogenism is once such adverse in-utero insult. Gestational testosterone (T)-treatment, an environment of gestational hyperandrogenism, manifests as hypertension and pathological left ventricular (LV) remodeling in adult ovine offspring. Furthermore, sexual dimorphism is noted in cardiomyocyte number and morphology in fetal life and at birth. This study investigated transcriptional changes and potential biomarkers of prenatal T excess-induced adverse cardiac programming. Background Maximal inspiratory pressure (MIP) is currently the most commonly used measure for respiratory muscle strength (RMS) estimation, however, requires significant effort. Falsely low values are therefore common, especially in fatigue-prone subjects, such as neuromuscular disorder patients. In contrast, sniff nasal inspiratory pressure (SNIP) requires a short, sharp sniff; this is a natural manoeuvre, decreasing required effort.
