Technical measurement is foundational in large-scale complex engineered systems (LSCES) design and contract awards due to the state of practice of systems engineering. Although technical measurement is ubiquitous in systems engineering practice, the selection and assessment of the impact of a set of technical measures is often heuristics-based, rather than based on empirical evidence or theory. This dissertation studies the existence and stability of solutions for a class of non-autonomous systems of differential equations in multi-dimensional time, within the framework of Banach space. Focusing on both hyperbolic and parabolic cases, it presents a comprehensive range of results on existence and stability over finite and infinite time intervals. By leveraging the theory of evolution families, this work uncovers the conditions under which these systems have solutions and provides an in-depth analysis of how these solutions evolve, offering fresh perspectives on their dynamic behavior over time.

Cloud base height (CBH) and cloud base vertical velocity (CBVV) are essential parameters for understanding cloud formation, boundary layer evolution, and air exchange between the atmosphere and the boundary layer. CBH is linked to aviation safety and impacting the amount of solar radiation reaching the surface, while CBVV influences cloud development, condensation (droplet formation) and precipitation potential. High CBVV can erode capping inversions and trigger convection. These two parameters are the focus of this dissertation. The origin of solar energetic electrons has been a long-standing problem, namely the production of a huge number of energetic electrons with power-law energy spectra within a sufficiently short time. Using particle-in-cell (PIC) magnetic reconnection (MR) simulations based on a new vortical electron acceleration model proposed by Che and Zank (CZ) [21], I first investigated the scaling of the PIC MR simulations to the observational scales of solar flares. The CZ model reproduced the sharp two-stage transition observed in soft-hard-harder electron energy spectra, implying that the electron acceleration timescale must be a small fraction of the solar flare duration. High-latitude neutral winds have a number of drivers, both from solar and magnetospheric origins as well as from the lower atmosphere. The efficiency of the individual thermospheric forces, in addition to the relative strength of ion-neutral coupling, are not well characterized. Because of this, the neutral wind response to changes in ionospheric convection is not well understood, with response times ranging on the order of tens of minutes to hours. Using both data driven and modeling simulation efforts, we aim to develop a better understanding of high-latitude ionosphere-thermosphere coupling. With the advent of videogames as the world's first mass-market interactive media, gamification has emerged as the premier way to harness powerful behavior-affecting gamified elements and place them in other contexts, such as education, fitness, and rehabilitation. This transposition, however, ignores a key tenant of user engagement: reward ought to be aligned with effort. Most, if not all, current gamification systems dispatch static reward allocations for their spread of gamified behaviors. In this paper, an alternative is presented: dynamic gamification. The District of Columbia ranks first nationally for health insurance coverage; however, African American/Black children have disproportionately experienced barriers to care compared to the District of Columbia’s pediatric population overall. The development of polymeric micelles for biomedical applications has been of significant interest due to their potential for targeted delivery for therapeutic and diagnostic agents. This dissertation focusses on the synthesis, characterization, and application of amphiphilic PEGylated poly(amino acid) block copolymers as nanocarriers for superparamagnetic iron oxide nanoparticles (SPIONs) in magnetic particle imaging (MPI). Series of novel diblock and triblock copolymers were synthesized through ring-opening polymerization of amino acid N-Carboxyanhydrides (NCAs) to afford HO-PEG77-b-p(L-Leu)m and HO-PEGx-b-p(Allyl-L-Gly)10-b-p(L-Leu)n. This research is focused on the study of the trajectory tracking of a Hypersonic Missile (HM) in its terminal phase of flight in light of the cross coupling, high nonlinearity, and uncertainty in the missile dynamics. Stream Interaction Regions (SIRs) play a critical role in energetic particle acceleration and transport in the heliosphere. While shocks and compression regions associated with SIRs can accelerate particles to several MeV energies, most of our current understanding of these events is based on observations near 1 AU and beyond. Observations inside 1 AU, particularly by PSP and SolO, provide a unique opportunity to disentangle the roles of acceleration and transport. When the elements of an antenna array are closely spaced, the electromagnetic coupling between them can distort the radiation patterns of the individual elements and change their input impedances, causing significant degradation in the array’s performance. A challenge in neuroscience is understanding how neural circuits generate and sustain behavior over time. Neural circuits consist of interconnected groups of neurons that process and integrate sensory information to perform specific tasks and behaviors. These circuits organize behaviors by maintaining internal states—stable neural activity patterns influenced by sensory and physiological contexts, as discussed in Chapter One.

To understand how neural circuits generate internal states, we examined how the well-characterized C. elegans egg-laying circuit transitions between inactive and active states to produce egg-laying behavior. Pseudomonas aeruginosa is an opportunistic pathogen known to cause acute infections in individuals with epithelial damage or medically placed devices, leading to sepsis or death in immunocompetent and immunocompromised individuals. Like many other Gram-negative bacteria, P. aeruginosa uses a Type III Secretion System (T3SS) to intoxicate eukaryotic cells. This T3SS is essential for P. aeruginosa to establish an infection and avoid clearance by immune cells and is also linked to increased morbidity and mortality in infections. Despite this necessity, activation of the T3SS slows P. aeruginosa’s cell growth and permits innate immune system detection and resulting inflammation. To balance …

Nerve injury causes neuropathic pain and multilevel nerve barrier disruption. Nerve barriers consist of perineurial, endothe- lial and myelin barriers. So far, it is unclear whether resealing nerve barriers fosters pain resolution and recovery. To this end, we analysed the nerve barrier property portfolio, pain behaviour battery and lipidomics for precursors of specialized pro-resolving meditators (SPMs) and their receptors in chronic constriction injury of the rat sciatic nerve to identify tar- gets for pain resolution by resealing the selected nerve barriers. Off-target neuronal injury is a serious side-effect observed in cancer survivors. It has previously been shown that pediatric acute lymphoblastic leukemia (ALL) survivors have a decline in neurocognition compared to healthy age-matched counterparts. Elevated oxidative stress has been documented to be a mediator in off- target tissue damage in cancer survivors. Early detection of oxidative stress markers may provide an opportunity to prevent off-target tissue damage. Few evidence-based trainings exist on how to equip healthcare providers, particularly nurses, with the skills to engage in cost of care conversations with patients/caregivers to mitigate the impact of cancer-related financial toxicity. This study evaluated a pilot training developed in collaboration with Triage Cancer® to prepare oncology nurses to identify and assist patients/caregivers facing financial and/or legal barriers to care. Ten pediatric oncology nurses completed the training and pre/post-surveys on behaviors related to financial and legal need screening, frequency and comfort level of answering questions, knowledge, and behavior changes, along with training evaluation questions. Cardiomyocytes (CMs) lost during ischemic cardiac injury cannot be replaced due to their limited proliferative capacity. Calcium is an important signal transducer that regulates key cellular processes, but its role in regulating CM proliferation is incompletely understood. Here we show a robust pathway for new calcium signaling-based cardiac regenerative strategies. A drug screen targeting proteins involved in CM calcium cycling in human embryonic stem cell-derived cardiac organoids (hCOs) revealed that only the inhibition of L-Type Calcium Channel (LTCC) induced the CM cell cycle.
