8-oxoguanine (8-oxoGua) is one of the most frequent forms of oxidative DNA base lesions, repaired by 8-oxoguanine DNA glycosylase 1 (OGG1) via base excision repair (BER) pathway to maintain genome fidelity. The human allelic variant hOGG1S326C, prevalent in Caucasians and Asians, has been regarded as a susceptibility factor for various diseases, yet its pathogenic mechanism remains elusive. In this study, we demonstrate that Ogg1S326C/S326C mice exhibit increased and sustained airway inflammation compared with wild-type (WT) Ogg1S326/S326 mice. Mechanistically, in response to inflammatory stimulation, OGG1S326C undergoes reactive oxygen species-induced dimerization, which impairs its base excision function, but …

Sjögren's disease (SjD) is a systemic autoimmune disorder primarily causing dry eyes and mouth. It frequently overlaps with other autoimmune diseases (AIDs), including rheumatoid arthritis (RA) and systemic lupus erythematosus (SLE). However, the genetic basis of SjD remains underexplored, limiting our understanding of its connections to other immune-mediated conditions. In this study, we aimed to identify gene networks associated with SjD through the integration of genetic, transcriptomic, and epigenomic data. We further compared the genetic factors of SjD with other immune-mediated diseases. We analyzed genome-wide association studies (GWAS) summary statistics, DNA methylation, and transcriptomic data using our in-house network-based methods, …

ABSTRACT

Bullying is perceived in different ways according to the experience or training of individuals. In my practice, adults often misidentify disputes or incidents as bullying due to a lack of evidence or varying perceptions. This problem must be a theme of reflection for all stakeholders in traditional or charter schools. My study aimed to understand how teachers and parents perceive bullying in terms of social implications, cultural factors, and professional training. In my research, I provided insight into how the stakeholders in a public charter school perceive this phenomenon. The participants highlighted their perceptions and topics of communication, exclusion, …

Assessment Of The Risk Factors Affecting Diabetes Outcome In Kumasi Metropolis Of Ghana, Audrey Jacobs May 2025

Assessment Of The Risk Factors Affecting Diabetes Outcome In Kumasi Metropolis Of Ghana, Audrey Jacobs

Honors College Theses

This paper examines Kumasi Metropolis, Ghana risk factors for diabetes outcomes, considering the prominent determinants such as geographic location, income, and educational level. Such determinants play a role when it comes to shaping health outcomes, with research indicating positive correlation between per capita expenditure on healthcare and life expectancy. In spite of that, inequalities do occur, even in developing nations such as Ghana, where minimal health care investment impacts poor health outcomes. Socioeconomic factors, including low living conditions, poor access to healthcare, and illiteracy, reinforce health disparities, especially in rural areas. Informed by a review of past research, we hypothesized …

Invasion As A Structure: Carceral Capitalism, Necropolitics, And The Persistence Of Settler-Colonialism, Ethan Peacock May 2025

Invasion As A Structure: Carceral Capitalism, Necropolitics, And The Persistence Of Settler-Colonialism, Ethan Peacock

Honors College Theses

In the United States, settler-colonialism is often dismissed as a "bad chapter" of the past, an embarrassing episode of the nation's history, considered irrelevant to contemporary socio-political discussions. In contrast, while ongoing, Canada's reconciliation efforts remain performative, failing to address underlying colonial power structures. This project argues that settler-colonialism is not merely a historical phenomenon but a pervasive socio-political and cultural structure shaping the contemporary world. Capitalism depends on racialized and gendered hierarchies, continually produced through dispossession, marginalization, and exploitation. The prison-industrial complex (PIC) exemplifies this dynamic by industrializing dehumanization and weaponizing state violence against marginalized communities through mass incarceration, …

Identity And The Russian-Ukrainian War: Evidence From The American Community Survey, Matthew Holian, Oleh Wolowyna

Faculty Research, Scholarly, and Creative Activity

The Russian invasion of Ukraine in 2022 led to significant changes in ethnic identity and language preferences among U.S. residents. We observe an 18.9% increase in U.S.-born individuals identifying as ethnic Ukrainian from 2021 to 2022 and a 6.8% decrease for U.S.-born ethnic Russians, which cannot be attributed to prior trends or sampling variation. The changes among Ukrainians were more pronounced among college graduates and those living in Democratic-leaning counties. We document an increase in the use of the Ukrainian language among Ukrainian immigrants, which cannot be explained by prior trends or immigration patterns. We test for labor market discrimination, …

Can Flywheel Training Serve As A Viable Intervention For Dynamic Strength Index Based Programs?, Jeffrey Ruiz May 2025

Can Flywheel Training Serve As A Viable Intervention For Dynamic Strength Index Based Programs?, Jeffrey Ruiz

Doctorate in Kinesiology

The purpose of this case series was to examine the effects that flywheel training had on a person’s dynamic strength index in comparison to concurrent training (DSI). It has been shown that with strength training alone there can be an overall decrease in the DSI, due to an increase the IMTP peak force and no significant change in the CMJ performance (Comfort et al., 2018). Flywheel training’s effect on the DSI specifically have not been previously examined. It was hypothesized that a regimented flywheel training intervention will demonstrate an

improvement in the peak force for the countermovement jumps as well …

Avoiding Recidivism Of Recently Released Offenders In North Carolina, Josue Genaro Galvez

Walden Dissertations and Doctoral Studies

There is a problem with recidivism in the North Carolina prison system and the United States. This study fills the gap in the literature by focusing on what life experiences contribute to behaviors in recently released offenders not to reoffend in North Carolina. Edmund Husserl's (1913) Transcendental Phenomenology used several angles of perception and a thematic analysis of the participant’s lived experiences. This study employed a purposive sampling method by targeting 17 participants who may have represented characteristics of the intended population. Researchers do not know what kept these released offenders from avoiding prison. This phenomenological design focused on the …

Lived Experiences Of Hispanic Parents Considering Early Intervention Services For Developmentally Delayed Children, Karina Proffer May 2025

Lived Experiences Of Hispanic Parents Considering Early Intervention Services For Developmentally Delayed Children, Karina Proffer

Walden Dissertations and Doctoral Studies

Studies have demonstrated that children benefit significantly from Early Intervention (EI) services due to brain plasticity and rapid early development. EI supports growth and establishes foundational skills in cognitive, social, emotional, motor, language, and daily living domains. Research indicates that children who participate in EI programs experience positive outcomes in their academic trajectories. However, limited studies examine the lived experiences of Hispanic parents with children diagnosed with autism or intellectual disabilities who did not utilize EI services. This study explores the factors influencing these parents' decisions and experiences that hinder their children’s participation in the program. The Theory of Planned …

How School Professionals Perceive They Can Engage Parents In Helping Children In Fifth Grade Cope Adaptively With Stress, Hector Alejandro Soto

Walden Dissertations and Doctoral Studies

National education statistics indicate issues of aggressive behaviors in middle schools, such as bullying and disrespecting teachers. This suggests that fifth-grade students could benefit from parental intervention to manage stress and reduce such behaviors. However, there is limited information on effectively engaging parents in this process. The study involved a purposive sample of 20 school professionals from four schools in a large southwestern U.S. school district who were invited to address this gap. Nine professionals completed the online survey of six qualitative, open-ended questions regarding engaging parents in helping fifth graders cope with stress. The questions focused on establishing meaningful …

Center on Aging Staff Publications

Hematopoietic stem cells (HSCs) are defined by their capacity to regenerate all main components of peripheral blood, but individual HSCs exhibit a range of preferences for generating downstream cell types. Their propensities are thought to be epigenetically encoded, but few differential regulatory mechanisms have been identified. In this work, we explored the role of DNA methyltransferase 3A (DNMT3A) in the megakaryocyte-biased HSC population, which is thought to reside at the top of the hematopoietic hierarchy. We demonstrate that heterozygous loss of DNMT3A (Dnmt3a+/-) in these megakaryocyte-biased HSCs has distinct consequences compared with the rest of the HSC pool. These megakaryocyte-biased …

White Matter Connections Within The Central Sulcus Subserving The Somato-Cognitive Action Network, Georgios P Skandalakis, Evan M Gordon, Nico U F Dosenbach, Et Al.

2020-Current year OA Pubs

The somato-cognitive action network (SCAN) consists of three nodes interspersed within Penfield's motor effector regions. The configuration of the somato-cognitive action network nodes resembles the one of the 'plis de passage' of the central sulcus: small gyri bridging the precentral and postcentral gyri. Thus, we hypothesize that these may provide a structural substrate of the somato-cognitive action network. Using microdissections of 16 human hemispheres, we consistently identified a chain of three distinct plis de passage with increased underlying white matter in locations analogous to the somato-cognitive action network nodes. We mapped localizations of plis de passage into standard stereotactic space …

Novel Treatment Approaches Utilizing Antibody-Drug Conjugates In Breast Cancer, Andrew A Davis, Jennifer Hesse, Patrícia M R Pereira, Cynthia X Ma

2020-Current year OA Pubs

Antibody-drug conjugates (ADCs) are rapidly changing the way we treat patients with breast cancer. Despite this progress, many unanswered questions remain regarding the sequencing of different ADCs with similar payloads, optimal combinations, drug design strategies to limit off-target toxicities, biomarkers to define antigen positivity, and the use of ADCs in the neoadjuvant and adjuvant settings. In this review, we summarize novel ADC approaches in breast cancer treatment, including potential improvements in ADC payloads, linkers, targets, and drug delivery. We also evaluate novel strategies to combine ADCs with other agents, such as targeted drugs and immune checkpoint inhibitors. To improve patient …

The conjugation of small ubiquitin-like modifiers (SUMO) to target proteins, known as SUMOylation, plays a crucial role in regulating protein homeostasis, activity, interaction with other proteins, and subcellular localization. Loss of SUMOylation in nongrowing oocytes by conditional deletion of the E2 SUMO conjugating enzyme, Ube2i, at the primordial follicle stage leads to female sterility due to complex changes in oocyte development, including altered folliculogenesis, defective meiotic progression, and premature loss of the ovarian reserve. In this study, proteomics was used to compare control and Ube2i conditional knockout ovaries during the first wave of folliculogenesis to identify key differences that may …
