A Phase I Dose-Escalation Study Of The Hif-2 Alpha Inhibitor Dff332 In Patients With Advanced Clear-Cell Renal Cell Carcinoma, Sumanta K Pal, Joel Picus, Et Al.

PURPOSE: Mutations or silencing of the von Hippel-Lindau tumor suppressor gene accumulate hypoxia-inducible factors (HIF). HIF-2α is implicated in the oncogenesis of ∼50% of patients with clear-cell renal cell carcinoma (ccRCC) but has been considered "undruggable." DFF332, an orally administered novel allosteric inhibitor of HIF-2α, showed dose-dependent antitumor efficacy in preclinical models of ccRCC.

PATIENTS AND METHODS: This first-in-human study evaluated the safety, tolerability, antitumor activity, pharmacokinetics, and pharmacodynamics of DFF332 in patients with heavily pretreated advanced ccRCC. Sterile acute kidney injury (AKI) is common in the clinic and frequently associated with unexplained hypoxemia that does not improve with dialysis. AKI induces remote lung inflammation with neutrophil recruitment in mice and humans, but which cellular cues establish neutrophilic inflammation and how it contributes to hypoxemia is not known. Here we report that AKI induced rapid intravascular neutrophil retention in lung alveolar capillaries without extravasation into tissue or alveoli, causing hypoxemia by reducing lung capillary blood flow in the absence of substantial lung interstitial or alveolar edema. Mechanisms that mediate allograft tolerance differ between organs. We have previously shown that Foxp3+ T cell-enriched bronchus-associated lymphoid tissue (BALT) is induced in tolerant murine lung allografts and that these Foxp3+ cells suppress alloimmune responses locally and systemically. Here, we demonstrated that Foxp3+ cells that reside in tolerant lung allografts differed phenotypically and transcriptionally from those in the periphery and were clonally expanded. Using a mouse lung retransplant model, we showed that recipient Foxp3+ cells were continuously recruited to the BALT within tolerant allografts. Over the past few decades, numerous efforts have been made to increase the proportion of women in the construction industry, coupled with various calls for legislation and rules to prohibit gender discrimination. Despite these efforts, minimal progress has been noticed in the construction industry. While recruitment remains crucial, the current culture in construction reveals a knowledge gap in recruitment and retention in employment—a concept known as a ‘leaky pipeline’. Living with dementia is a challenging journey for both those with dementia and their families. Besides memory loss and cognitive decline, persons with dementia often face challenges of independent living and loss of quality of life. To enhance and extend the period of independent living, an open-source software and commercial sensor-based home monitoring system is being developed. These sensors are relatively compatible with home environments, can be personalized for different users, and provide constant support including reminders, also allowing for early identification of physical or behavioural changes. There is much information available on dementia, but information may not be written at an appropriate level for a lay reader. One aim of the CoDESIGN project is to simplify information available for those who have lower levels of literacy. We investigate the potential to simplify documents to reflect the literacy abilities of the reader. We report on the ability of ChatGPT to modify documents to such target reading levels while retaining the accuracy and completeness of the documents, while avoiding the introduction of errors. Results suggest that human supervision is required before documents can be made available to readers.

This study explores the factors influencing the availability of assistive technology for people with dementia through the application of machine learning. The analysis identified key factors, including carer support, protective legislation, and access to reimbursable products from the WHO Global Health Observatory. A random forest model with high accuracy emphasises the importance of targeted resources and policies in improving dementia care infrastructure.

Based on seminal work in placental species (eutherians)1,2,3,4,5,6,7,8,9,10, a paradigm of mammalian development has emerged wherein the genome-wide erasure of parental DNA methylation is required for embryogenesis. Whether such DNA methylation reprogramming is, in fact, conserved in other mammals is unknown. Here, to resolve this point, we generated base-resolution DNA methylation maps in gametes, embryos and adult tissues of a marsupial, the opossum Monodelphis domestica, revealing variations from the eutherian-derived model. Facial dermatoses in e-cigarette users arise from nicotine-induced oxidative stress, inflammatory signaling, and barrier dysfunction, contributing to an increased prevalence of acneiform eruptions, perioral dermatitis, and rosacea exacerbations. E-cigarette aerosols contain nicotine, volatile organic compounds, and reactive oxygen species (ROS) that disrupt cutaneous homeostasis by impairing antioxidant defense mechanisms and promoting lipid peroxidation in the epidermis. Nicotine exposure induces mitochondrial dysfunction in keratinocytes and sebocytes, leading to dysregulated sebum production, enhanced Cutibacterium acnes colonization, and increased susceptibility to inflammatory acne. Additionally, nicotine-mediated vasoconstriction and endothelial dysfunction reduce dermal microcirculation, compromising oxygen delivery and delaying wound healing, while heightened expression of …

Confronting The Duty To Capitalize In Veil-Piercing, Douglas C. Michael

In corporate law, the doctrine of “piercing the corporate veil” allows courts to hold shareholders personally liable for corporate obligations. The doctrine—which now also applies to entities other than corporations—has been widely criticized as imprecise and unworkable. I respond to that criticism in the area of veil-piercing law which remains most intractable—the case of the tort victim who remains unsatisfied after exhausting corporate assets. Although most commentators have eschewed talk of any duty owed by the business owner to vest the business with sufficient assets, I argue that courts and legislatures should recognize—confront—such a duty. Ncaa Women Athletes And Nil Pay Disparities: Are They Students Under Title Ix, Employees Under Title Vii, Or Both?, Michael H. Leroy

Title IX requires schools to eliminate gender disparities in financial benefits and publicity for athletes. This Article shows that men’s basketball players in major National Collegiate Athletic Association (NCAA) conferences were paid an average of $171,272 in 2024, compared to $16,222 for women. Analysis of a prominent basketball program shows that it coordinates with its Name, Image, and Likeness (NIL) collective to monetize NIL donor access in favor of men.

How does visual cortex form increasingly complex and invariant representations? We address this question starting from our understanding of natural images and V1. Although it is well-known that the relative phases of frequency components in images contain important structure, the standard model of V1 complex cells discards phase information within the receptive field. We propose a model for disentangling invariances and their accompanying transformations based on a complex-valued sparse coding model adapted from Cadieu & Olshausen 2012. Perceptual spaces are representations of a sensory or cognitive domain in which the domain’s elements correspond to points, and distances between these points are perceptual differences. Proximity relationships within a perceptual space can support a variety of functions, including discrimination, grouping, learning, and generalization. These diverse functions may use the features of the domain in different ways, resulting in task-dependent influences on the geometry of the space.
