Long-Term, Super-Resolution Imaging Of Amyloid Structures Using Transient Amyloid Binding Microscopy, Tianben Ding, Kevin Spehar, Jan Bieschke, Matthew D. Lew

Amyloid fibrils and tangles are signatures of Alzheimer disease, but nanometer-sized aggregation intermediates are hypothesized to be the structures most toxic to neurons. The structures of these oligomers are too small to be resolved by conventional light microscopy. We have developed a simple and versatile method, called transient amyloid binding (TAB), to image amyloid structures with nanoscale resolution using amyloidophilic dyes, such as Thioflavin T, without the need for covalent labeling or immunostaining of the amyloid protein. Transient binding of ThT molecules to amyloid structures over time generates photon bursts that are used to localize single fluorophores with nanometer precision. …
Fundamental Limits Of Measuring Single-Molecule Rotational Mobility, Oumeng Zhang, Matthew D. Lew
Fundamental Limits Of Measuring Single-Molecule Rotational Mobility, Oumeng Zhang, Matthew D. Lew
Electrical & Systems Engineering Publications and Presentations
Various methods exist for measuring molecular orientation, thereby providing insight into biochemical activities at nanoscale. Since fluorescence intensity and not electric field is detected, these methods are limited to measuring even-order moments of molecular orientation. However, any measurement noise, for example photon shot noise, will result in nonzero measurements of any of these even-order moments, thereby causing rotationally-free molecules to appear to be partially constrained. Here, we build a model to quantify measurement errors in rotational mobility. Our theoretical framework enables scientists to choose the optimal single-molecule orientation measurement technique for any desired measurement accuracy and photon budget.

Grain yield is a trait of paramount importance in the breeding of all cereals. In wheat (Triticum aestivum L.), yield has steadily increased since the Green Revolution, though the current rate of increase is not forecasted to keep pace with demand due to growing world population and increasing affluence. While several genome-wide association studies (GWAS) on yield and related component traits have been performed in wheat, the previous lack of a reference genome has made comparisons between studies difficult. In this study, a GWAS for yield and yield-related traits was carried out on a population of 322 soft red …

Evaluation Of Soybean [Glycine Max (L.) Merr.] Genotypes For Yield, Water Use Efficiency, And Root Traits, Harrison Gregory Fried, Sruthi Narayanan, Benjamin Fallen
Publications
Drought stress has been identified as the major environmental factor limiting soybean [Glycine max (L.) Merr.] yield worldwide. Current breeding efforts in soybean largely focus on identifying genotypes with high seed yield and drought tolerance. Water use efficiency (WUE) that results in greater yield per unit rainfall is an important parameter in determining crop yields in many production systems, and is often related with crop drought tolerance. Even though roots are major plant organs that perceive and respond to drought stress, their utility in improving soybean yield and WUE under different environmental and management conditions are largely unclear. The …

What Next For Russia’S Front-Line States?, Keir Giles Mr.
Books, Monographs & Collaborative Studies
Russia’s annexation of Crimea and intervention in Eastern Ukraine in 2014, alarmed not only Western-leaning states in Central Europe and the Baltic but also Moscow’s traditional allies. These events signaled that Moscow is now willing and capable of using direct military force against perceived strategic threats in its self-proclaimed region of vested interests. With the exception of Ukraine and the Baltic States, this Letort Paper examines how Russia’s front-line states have adjusted their foreign policy posture since 2014. Belarus, Moldova, the states of Central Asia and the South Caucasus calculate the benefits and risks in their relationship with Moscow and …

The Influence Of Fetch On The Holocene Thermal Structure Of Hidden Lake, Glacier National Park, Jeffery R. Stone, Jasmine E. Saros, Trisha L. Spanbauer
Department of Earth and Atmospheric Sciences: Faculty Publications
We use three-dimensional modeling of the basin of Hidden Lake, Montana, to assess the influence of effective fetch on diatom-inferred changes in mixing depths throughout the Holocene. The basin of Hidden Lake is characterized by a complex morphometry; for example, three-dimensional modeling of the lake basin indicates that a decrease in lake level of 2 m would result in complete isolation of the deepest part of the lake basin from the rest of the lake. Our model suggests that small changes in the lake surface elevation at Hidden Lake would produce threshold-like responses in effective fetch, which in turn would …

The Marquette Palliative Care System, Rechard M. Love, Sheikh Iqbal Ahamed
Computer Science Faculty Research and Publications
Worldwide, major clinical barriers to effective palliative care are the absence of a. current data on symptom type and intensity; b. immediately accessible information on practical, affordable, and effective interventions; and c. self-sustaining systems to facilitate physician engagement in continuing palliative care. Our adaptable system, developed for Bangladesh and Nepal, addresses these barriers. A tele-home palliative care program facilitates home care, minimizes expense, and encourages efficient professional practitioner involvement employing two information technology tools: a patient cell-phone “app” on the Android platform with a 15-item symptom questionnaire and an Internet website with health information sections for patients/families and for clinicians. …

在60、70年代之交香港文化雜誌的政治性視覺生產初覽, Kin Wah Lau
在60、70年代之交香港文化雜誌的政治性視覺生產初覽, Kin Wah Lau
Lingnan Theses (MPhil & PhD)

Ten Questions Concerning Building Performance Analysis, Pjc De Wilde
School of Art, Design and Architecture
Building performance analysis is an important yet surprisingly complex activity. This article explores the current understanding of the concept of building performance, and explains why its analysis is a challenging activity that mostly requires expert intervention. It addresses some of the common questions about building performance, such as: What can be learnt from other disciplines that also deal with performance? What are the benefits of applying building performance analysis in the building design process? How can building performance analysis support building operation and facility management? What is the relation between building performance analysis and the class of high performance buildings? …

Drought is a major challenge in rice production system worldwide. We conducted a phenotypic screening of the USA rice genotypes for drought tolerance and assessed genetic diversity using SSR markers. Identification of QTLs for drought tolerance during both vegetative and reproductive stage was done using genotyping by sequencing (GBS) based saturated SNP linkage map. The rice genotypes Jes, Leah, Roy J, Jazzman, and Madison were found to be drought tolerant. Population structure analysis grouped the USA rice genotypes into California, Louisiana, and Arkansas types. Marker-trait association showed that markers RM570 and RM351 were significantly associated with grain yield, spikelet fertility, …
