Structural And Functional Characterization Of Sulfonium Carbon-Oxygen Hydrogen Bonding In The Deoxyamino Sugar Methyltransferase Tyim1, Robert J. Fick, Scott Horowitz, Brandon G. Mcdole, Mary C. Clay, Ryan A. Mehl, Hashim M. Al-Hashimi, Steve Scheiner, Raymond C. Trievel

The N-methyltransferase TylM1 from Streptomyces fradiae catalyzes the final step in the biosynthesis of the deoxyamino sugar mycaminose, a substituent of the antibiotic tylosin. The high-resolution crystal structure of TylM1 bound to the methyl donor S-adenosylmethionine (AdoMet) illustrates a network of carbon-oxygen (CH•••O) hydrogen bonds between the substrate’s sulfonium cation and residues within the active site. These interactions include hydrogen bonds between the methyl and methylene groups of the AdoMet sulfonium cation and the hydroxyl groups of Tyr14 and Ser120 in the enzyme. To examine the functions of these interactions, we generated Tyr14 to phenylalanine (Y14F) and Ser120 to alanine …

Six students from Gardner-Webb University competed recently in the annual Big South Conference Quiz Bowl hosted by High Point University. The team finished second in preliminary competition, and Cameron Walters, a junior music composition major from Bessemer City, N.C., had the third highest individual score. The annual Quiz Bowl is organized by the Big South Academic Consortium.

Osteoblasts Are "Educated" By Crosstalk With Metastatic Breast Cancer Cells In The Bone Tumor Microenvironment., Alexus D. Kolb, Alison B. Shupp, Dimpi Mukhopadhyay, Frank C. Marini, Karen M. Bussard

INTRODUCTION: In a cancer-free environment in the adult, the skeleton continuously undergoes remodeling. Bone-resorbing osteoclasts excavate erosion cavities, and bone-depositing osteoblasts synthesize osteoid matrix that forms new bone, with no net bone gain or loss. When metastatic breast cancer cells invade the bone, this balance is disrupted. Patients with bone metastatic breast cancer frequently suffer from osteolytic bone lesions that elicit severe bone pain and fractures. Bisphosphonate treatments are not curative. Under ideal circumstances, osteoblasts would synthesize new matrix to fill in erosion cavities caused by osteoclasts, but this is not what occurs. The Anthropocene has emerged as the dominant conception of the contemporary moment, centering the human individual as both responsible for and bearing the responsibility to counteract its numerous interrelated socioeconomic, political, and environmental issues including the staggering loss of biodiversity across the globe and the reality of anthropogenic climate change. This constitutes a significant psychological impasse that disempowers and disenfranchises humans living in this epoch, discouraging any substantive individual effort. The Viceroyalty of Brazil (c. 1720–1815) refers to a polity that, at its greatest extent, roughly corresponded in geographic area to the modern nation-state of Brazil. Lying on the upper Atlantic coast of South America, it is bounded on the northeast by the Guyanas, to the northwest by the Viceroyalty of New Granada, to the west by the Viceroyalty of Peru, and to the southwest and south by the Viceroyalty of the Río de la Plata. In six well-regarded hydrology and water resources journals published in 2017, the estimated percentage of studies whose results could be fully reproduced was only between 0.06 and 6.8 percent. This low level of reproducibility is not uncommon in hydrology studies — a fact many scientists readily acknowledge. However, a team of researchers at Utah State University may have found a solution to make these studies more reproducible.

BACKGROUND

Transjugular intrahepatic portosystemic shunts (TIPS) can alleviate complications of portal hypertension such as ascites and variceal bleeding by decreasing the portosystemic gradient. In limited clinical situations, parallel TIPS may be only solution to alleviate either variceal bleeding or ascites secondary to portal hypertension when the primary TIPS fails to do so. Data specifically addressing the use of this partially polytetrafluoroethylene covered nitinol stent (Viatorr®) is largely lacking despite Viatorr® being the current gold standard for modern TIPS placement.

CASE SUMMARY

All three patients had portal hypertension and already had a primary Viatorr® TIPS placed previously. …

Exploring Local Environmental Factors Influencing Geographic Distribution Of Black-Legged Tick Questing Activity, Chong Di

The deer tick (Ixodes scapularis or I. scapularis), also known as the black-legged tick, is the primary vector that transmits Lyme Disease (LD) in Northeastern United States. To contain the geographic expansion of Lyme disease ticks across the US in recent decades, ecological studies have been conducted to understand the biotic and abiotic environmental factors affecting tick activity. We observed in preliminary surveys that the tick host-seeking activity varies across small local areas.
