Study On Centrifugal And Freeze Drying Of Lactobacillus Plantarum Lp-S2 Dvs, Sui Chunguang, Liang Jinzhong, Wang Fengqing

Lactobacillus plantarum Lp-S2 which has the ability to produce gamma-aminobutyric acid, was used as starting strain to develop Direct vat set(DVS). Then the centrifugation process and freeze-drying process were optimized. By orthogonal experiment the optimal conditions of centrifugation were identified as: centrifugal temperature 4 ℃, Time-consuming 30 minutes, centrifugal force 6 000 r/min. In this condition, the centrifugal recovery rate of Lactobacillus Lp-S2 is 83.8%. The cryoprotectant was optimized by single factor and orthogonal test. Slices of tiegun yam were dried at different temperature and thickness without any chemical preservatives added. To dry the slices with constant temperature, optical fiber was used to detect the center temperature of the slices. As the material, fresh tiegun yam was dried by single factor experiment. Litsea cubeba oil microcapsule were prepared by spray drying method. The effect of several shell materials on emulsion stability and surface morphology of microcapsule was investigated, and process conditions of Litsea cubeba oil microcapsule were optimized. The results of scanning electronic microscope (SEM) showed that Litsea cubeba oil microcapsule prepared with hydroxypropyl-β-cyclodextrin/ arabic gum(HP-β-CD/AG) as wall material is similar to smooth spheres with uniform size and good fluidity, and dents, wrinkles and aggregation for microcapsules prepared with other wall materials were observed. In order to improve the color quality of dried banana slices and obtain the best color protecting method, the effects of five different colors protecting agents, i.e. phytic acid, ascorbic acid, sodium chloride, EDTA and citric acid, on the color quality of dried banana slices evaluating were studied, by detecting the L* value and browning degree. The results showed that the order of the effects was phytic acid > citric acid > EDTA > sodium chloride > ascorbic acid. The total content of polyphenols in polygonum captitatum was determined by Folin-Cioculteu colorimetry. 10 types of macroporous resins were tested by static adsorption and desorption experiments for purifying polyphenols in polygogum captitatum and purifying conditions by the best resin were investigated. The results showed that HPD-722 was the best resin. Purifying conditions were as follows: concentration of adcorption sample 2.0~4.5 mg/mL, 2 BV/h adsoption velocity, 7.5 BV 50% ethanol solution as eluent, the content of the total polyphenol sample was up to 59.02%.

A vinegar beverage combined with purple sweet potato and bitter buckwheat was developed, and the flavor compounds in it were analyzed. Purple sweet potato and buckwheat were processed through smashing, size mixing, saccharification, alcoholic fermentation and acetic fermentation to produce brew purple sweet potato and buckwheat vinegar, respectively.. Corn steep liquor was used as the sole carbon and nitrogen source for recombinant E. coli culture medium. Based on the single factor optimization, three factors of corn steep liquor, MgSO4 and trace elements were chosen to design the response surface of corn steep liquor medium components. With Box-Behnhen experimental design and response surface analysis, the optimum components of corn steep liquor medium were determined. Conventional coffee brewing techniques generate vast quantities of spent espresso grounds (SEGs) rich in lignocellulose and valuable bioactives. These bioactive compounds can be exploited as a nutraceutical or used in a range of food products, while breakdown of lignocellulose generates metabolizable sugars that can be used for the production of various high-value products such as biofuels, amino acids and enzymes. Response surface methodology (RSM) was used to optimize the enzymatic saccharification of lignocellulose in SEGs following a hydrothermal pretreatment. The inhibition of pumpkin lao-zao and pure rice lao-zao was compared, and the inhibition effect of fermented supernatant of the different concentrations and different fermented time and different adding amount of pumpkin were studied. Using electronic nose measure and evaluation model as a guidance method for inhibiting halitosis, the strongest bioactive fractions(EⅡM20) were separated from the 95% ethanol emblica extract by organic solvent extraction, repeated normal and reverse phase column chromatography. Consequently, three compounds were isolated from the EⅡM20 by semi-preparative HPLC. Their quantity-quality were 6.8 mg, 4.5 mg, 4.9 mg, repectively. The structures of the three compounds were elucidated by comprehensive spectroscopic methods such as 1H NMR, 13C NMR, HR-ESI-MS. The spectroscopy analysis indicated that the compounds were maltose, Bis(2-methylheptyl)phthalate and 1-(4’-amino-phenyl)-ethane-1,2-diol.

In this study,the detection approach was established for spectrophotometric determination of iron (Ⅲ), existing in the form of isomaltooligosaccharide-iron (Ⅲ) complex. The method was optimized by operating experiments considering the factors of the process. Several factors effected on isomaltooligosaccharide-iron (Ⅲ) complex digestion, including the volume fraction of acid, the heating temperature and time, and this approach yielded to accurate results except the kinds of acid. Titration acidity of different buffalo raw milk samples were investigated in both in the same and different season according to GB 4789.18—2010 of “milk and dairy products inspection”. Among the samples, 228 of them were from major buffalo farming community in Yunnan Province, and 655 were from 26 farmers. The alcohol and alkali-doped were test, and the composition of the diet, feeding patterns and parity and lactation days of buffalo were also surveyed. Studied the effect of the speed, the material of guide, feeding methods and the properties of cashew nut on the state of cutting during the process of shell. The results showed that within the scope of test level, the rate of correct position was 65.1%, when the cashew was boiled in the 115 ℃, for 20 min, with the pushing speed 20 r/min, wiht the rigid plastic guide , and article vertical feeding.
