Alzheimer's disease (AD, OMIM: 104300) is an age-related disorder that affects millions of people. One of the underlying causes of AD is generation of hydrophobic amyloid-beta 42 (Aβ42) peptides that accumulate to form amyloid plaques. These plaques induce oxidative stress and aberrant signaling, which result in the death of neurons and other pathologies linked to neurodegeneration. We have developed a Drosophila eye model of AD by targeted misexpression of human Aβ42 in the differentiating retinal neurons, where an accumulation of Aβ42 triggers a characteristic neurodegenerative phenotype. Polyhydroxyalkanoates (PHA) is a biopolymer that has the prospect of becoming a preferred renewable and environmentally friendly resource with various applications including in manufacturing and medicine. Intensive studies have been carried out to find the most practical and economical method for PHA production because current commercial production of PHA is stringent and extremely costly. The potential of microwave and ultrasound was evaluated for the pretreatment of brewer's spent grain (BSG). Under optimal conditions of microwave and ultrasound pretreatments, reducing sugar yields per 1 g of pretreated BSG were 64.4 ± 7 mg and 39.9 ± 6 mg, respectively. Subsequently, the pretreated BSG was evaluated as a substrate for production of Xylanopectinolytic enzymes using fungi isolated from spoiled fruits. Out of twenty-nine (29) isolates recovered, Mucor sp. (AB1) isolated from Bramley apple (Malus domestica) produced xylanopectinolytic enzymes with higher specific activity, and was selected for further studies. The highest enzyme activity (137 U/g, …

Despite our understanding of the biochemistry of the BRCA1/BARD1 E3 ligase, we know very little of the mechanism by which its cellular substrates, how they are chosen, how its E3 Ub ligase activity is regulated, which are the cofactors involved in this regulation, and how the substrates are modified during DDR. In this dissertation, I further evaluate BRCA1/BARD1-mediated E3 ligase activity and how CstF-50 and p97 regulate this reaction working as cofactors. My data show that BRCA1/BARD1 is an E3 ligase that ubiquitinates the RNA binding protein Human antigen R(HuR).

An efficient protocol was developed for rapid propagation and regeneration of the Dioscorea alata L. locally known as ‘ubi badak’ because of their rhinoceros-shaped tubers. In the present study, direct and indirect regeneration were induced to produce plantlets of Dioscorea alata L. For direct regeneration explants (leave, stem and node) were cultured on MS media supplemented with various combinations and concentrations of BAP and NAA to induce microshoots and roots formation. The optimum medium for microshoots formation was MS medium supplemented with 1.0 mg/l BAP and 0.5 mg/l NAA with the number of microshoot is 23.07±0.44 from node explant. While …

Mushroom cultivation and palm oil industry in Malaysia have contributed to the large volumes of lignocellulosic residues. Lignocellulolytic enzymes profiles (lignin peroxidase, laccase, xylanase, endoglucanase, and β-glucosidase) of Pleurotus pulmonarius (parents strains UMP001 and UMP 002, and the hybrid UMH004) during growth in sawdust substrate bags were investigated. The enzymes extracted from the spent mushroom substrate (SMS) of the strain with the highest enzymes productivity was used to hydrolyse palm oil mill effluent (POME). The role of the gut microbiome in human health and disease is the focus of much attention. It has been widely agreed upon that our gut bacteria play a role in host immunity, nutrient absorption, digestion, metabolism, and other key drivers of health. Furthermore, certain microbial signatures and specific taxa have also been associated with the development of diseases, such as obesity; inflammatory bowel disease; and, indeed, colorectal cancer (CRC), which is the focus of this review. The Swiss Constitution was amended by referendum in 1992 to include two unique provisions: Article 119, which imposes strict limits on genetic and reproductive technologies in humans in order to protect ‘human dignity’, and Article 120, which commits the Swiss federal government to limiting genetic technologies in non-human species on the basis of the ‘dignity of the creature’. This article analyzes the role of ‘dignity’ as a limit on biotechnologies in the Swiss constitutional order. Objectives: Iron depletion is common around the world and among certain risk groups in developed countries. The overall purpose was to test the suitability of a novel plasma collection card for minimally invasive iron status assessment.
Methods: Twenty participants (10 f/10 m) participated in this cross-sectional study. Ferritin and hemoglobin were measured from blood collected from a forearm vein, serving as reference method. Blood was also collected from the fingertip using the NoviplexTM Plasma Prep Card as well as capillary collection tubes.

Selective autolysosomal degradation of damaged mitochondria, also called mitophagy, is an indispensable process for maintaining integrity and homeostasis of mitochondria. One well-established mechanism mediating selective removal of mitochondria under relatively mild mitochondria-depolarizing stress is PINK1-Parkin-mediated or ubiquitin-dependent mitophagy. However, additional mechanisms such as LC3-mediated or ubiquitin-independent mitophagy induction by heavy environmental stress exist and remain poorly understood. The present study unravels a novel role of stress-inducible protein Sestrin2 in degradation of mitochondria damaged by transition metal stress. In contrast to efforts focusing on improving inclusion in STEM classrooms from kindergarten through un-dergraduate (K–16), efforts to improve inclusion in scientific meetings and conferences, important hubs of STEM culture, are more recent. Markers of inclusion that are sometimes overlooked at these events can include the composition of panels, how workshops are run, the affordability of conferences, and various other mechanisms that maintain pre-existing hierarchies and norms that limit the participation of early-career researchers and individuals of minoritized cultural, linguistic, and economic backgrounds. This paper intends to explore the rationality and feasibility of modeling dispersed submicron particles in air by a kinetic-based method called the unified gas-kinetic scheme (UGKS) and apply it to the simulation of particle concentration under a transverse standing wave. A gas-particle coupling scheme is proposed where the gas phase is modeled by the two-dimensional linearized Euler equations (LEE) and, through the analogous behavior between the rarefied gas molecules and the air-suspended particles, a modified UGKS is adopted to estimate the particle dynamics. Parkinson’s disease (PD) is the second most common neurodegenerative disorder and the motor symptoms are caused by progressive loss of midbrain dopamine neurons. There is no current treatment that can slow or reverse PD. Our current “DBS-Plus” clinical trial (NCT02369003) features the implantation in vivo of autologous Schwann cells (SCs) derived from a patient’s sural nerve into the substantia nigra pars compacta (SNpc) in combination with Deep Brain Stimulation (DBS) therapy for treating patients with advanced PD.

The risk involved through the transmission of viruses like Dengue, Zika, Yellow-fever and Chikungunya calls for an effective control method for the deadly vector. Objectives: This study aimed to identify carbapenem-resistant Gram-negative bacteria from clinical specimens of patients in Yola, Nigeria.
Methods: Routine clinical specimens were screened for the presence of carbapenem-resistant Gramnegative bacteria using chromogenic agar plates. Susceptibility of all presumptive isolates to carbapenems was tested by MIC and disk diffusion methods. Real-time PCR was used to test for the presence of carbapenemase genes.
Results: Screening of 1741 clinical specimens yielded 119 (6.8%) presumptive carbapenem-resistant Gram-negative bacteria. Antimicrobial susceptibility testing confirmed carbapenem resistance in 105 of these isolates. Distinct populations of steelhead in the wild are in decline. The propagation of steelhead in hatcheries has been used to boost population numbers for recreational fisheries and for use in conservation. However, hatchery breeding practices of steelhead can result in changes in genetic structure. I investigated the genetic structure of winter-run steelhead (Oncorhynchus mykiss) returning to the Mad River, California, where a hatchery has been used enhance production for recreational fisheries since 1971. To identify novel antimalarials from unexplored areas of chemical space, we have utilized two approaches: (a) using Torrey Pines Institute for Molecular Studies (TPIMS) proprietary high-density combinatorial libraries containing a large number of small molecule compounds residing largely in the underexplored areas of chemical space; and, (b) using novel, complexity-to-diversity/ring distortion of available natural products. The first approach involved the screening of more than 30 million compounds derived from 81 small molecule libraries, built on 81 distinct scaffolds. From this, we identified the pyrrolidine bis-cyclic guanidine library (TPI-1955) to be one of the most active and selective for antiplasmodial activity. …

เห็ดเผาะหนัง Astraeus odoratus เป็นราเอ็กโทไมคอร์ไรซาที่มีความสัมพันธ์แบบพึ่งพาอาศัยกันกับไม้วงศ์ไม้ยาง ซึ่งเป็นไม้พื้นถิ่นที่มีความสำคัญทางเศรษฐกิจอย่างมากของประเทศไทย ในปัจจุบันการปลูกไม้วงศ์ไม้ยางได้รับความสนใจเป็นอย่างมาก เนื่องจากเป็นไม้เนื้อแข็งที่มีคุณค่าทางการค้า และเกิดดอกเห็ดของราเอ็กโทไมคอร์ไรซาที่สามารถรับประทานได้อีกด้วย อย่างไรก็ตามความสำเร็จในการปลูกไม้วงศ์ไม้ยางและการปลูกป่าต้องการหัวเชื้อราเอ็กโทไมคอร์ไรซาเพื่อเพิ่มการเจริญเติบโตของพืช อัตราการอยู่รอดหลังการย้ายปลูก และความทนต่อสภาพแวดล้อมที่ไม่เหมาะสม ดังนั้นการพัฒนาหัวเชื้อราเอ็กโทไมคอร์ไรซาเพื่อให้ได้หัวเชื้อที่มีประสิทธิภาพจึงเป็นเรื่องสำคัญอย่างยิ่ง ในการศึกษานี้ เส้นใยของเห็ดเผาะหนังจะถูกดักจับเซลล์ด้วยแคลเซียมอัลจิเนตเพื่อให้ได้เป็นหัวเชื้อที่มีประสิทธิภาพ สายพันธุ์เห็ดเผาะ 9 สายพันธุ์ได้มาจาก 3 แหล่ง ได้แก่ จังหวัดน่าน กาญจนบุรี และสระบุรี โดยเห็ดเผาะสายพันธุ์ K-1 เป็นสายพันธุ์ที่ถูกคัดเลือกมาใช้ในการดักจับเซลล์ด้วยแคลเซียมอัลจิเนต เนื่องจากมีอัตราการเจริญของเส้นใยบนอาหารแข็ง PDA ภายใน 14 วัน มากที่สุด และได้มีการศึกษาผลของอาหารเลี้ยงเชื้อ 6 ชนิด ได้แก่ PDB, PG, MMN, MEB, Ohta และ Czapek ต่อการเจริญของเส้นใยเห็ดเผาะหนังสายพันธุ์ K-1 และ S-24 พบว่า PDB และ PG ส่งผลให้เส้นใยมีน้ำหนักแห้งมากที่สุดหลังการเลี้ยงเชื้อ 30 วัน ผลของสารเติมแต่งต่อการงอกของเส้นใยจากเม็ดแคลเซียมอัลจิเนต พบว่าน้ำตาลกลูโคสความเข้มข้น 0.5 ถึง 2 เปอร์เซ็นต์ และซอร์บิทอลความเข้มข้น 5 เปอร์เซ็นต์ สามารถรักษาอัตราการงอกของเม็ดแคลเซียมอัลจิเนตได้มากที่สุดเท่ากับ 100 และ 96 เปอร์เซ็นต์ ตามลำดับ เม็ดแคลเซียมอัลจิเนตคงเหลืออัตราการงอกของเส้นใย 55 เปอร์เซ็นต์ หลังจากการเก็บรักษา 90 วัน ในน้ำกลั่นที่อุณหภูมิ 25 องศาเซลเซียส โดยอัตราการงอกของเส้นใยจากชิ้นวุ้นที่ถูกเก็บในภาวะเดียวกันมีการลดลงอย่างรวดเร็วหลังการเก็บรักษาเพียง 5 วัน และไม่พบอัตราการงอกของเส้นใยหลังการเก็บรักษา 75 วัน หลังการเก็บรักษาเป็นเวลา 3 เดือน เส้นใยของเห็ดเผาะหนังสายพันธุ์ K-1 ในรูปเม็ดแคลเซียมอัลจิเนตสามารถเข้าติดเชื้อต้นกล้าตะเคียนทอง Hopea odorata ได้ด้วยอัตราการติดเชื้อ 46 เปอร์เซ็นต์ จากการทดลองนี้แสดงให้เห็นว่าเส้นใยของเห็ดเผาะหนังที่ถูกดักจับในเม็ดแคลเซียมอัลจิเนตสามารถนำมาใช้เป็นหัวเชื้อสำหรับการผลิตทางการค้าและใช้ในการอนุบาลกล้าไม้ป่าได้

Innovation in meat production has enabled modern humans to inflict far greater harm on animals, the environment, and public health than was possible just a few decades ago. Wiebers & Feigin aptly express the urgency with which these issues must be addressed. Those advocating for animals on moral grounds face resistance from omnivores citing taste, price and convenience. Further innovation in meat production (plant-based and cultured meat) will enable us to preserve the experience of eating meat whilst phasing out the many problems caused by industrial animal farming.

Utilizing light and genetic engineering, optogenetics permits the manipulation of events within cells via light using the light-sensitive properties of single-component microbial opsins. Microbial opsins are activated by a light source, such as lasers, light-emitting diodes, and incandescent sources that deliver light to the region of interest either directly or indirectly, such as through fiberoptics. In classical in vivo optogenetics, the wiring of optic fibers necessitates tethering of animals by the optic fiber to the light source. Miscanthus is a perennial wild grass that is of global importance for paper production, roofing, horticultural plantings, and an emerging highly productive temperate biomass crop. We report a chromosome-scale assembly of the paleotetraploid M. sinensis genome, providing a resource for Miscanthus that links its chromosomes to the related diploid Sorghum and complex polyploid sugarcanes. The asymmetric distribution of transposons across the two homoeologous subgenomes proves Miscanthus paleo-allotetraploidy and identifies several balanced reciprocal homoeologous exchanges. Analysis of M. sinensis and M. sacchariflorus populations demonstrates extensive interspecific admixture and hybridization, and documents the origin of the highly productive triploid bioenergy crop M. …

Background: Mango is one of the world’s most important tropical fruits. It belongs to the family Anacardiaceae, which includes several other economically important species, notably cashew, sumac and pistachio from other genera. Many species in this family produce family-specific urushiols and related phenols, which can induce contact dermatitis.
Results: We generate a chromosome-scale genome assembly of mango, providing a reference genome for the Anacardiaceae family. Our results indicate the occurrence of a recent whole-genome duplication (WGD) event in mango. Duplicated genes preferentially retained include photosynthetic, photorespiration, and lipid metabolic genes that may have provided adaptive advantages to sharp historical decreases …
