Reniform nematode (Rotylenchulus reniformis, RN) is a major plant-pest affecting key crops in Arkansas such as cotton and soybean, causing economic losses exceeding $757 million in the U.S. over the past decade. While some genetic resistance exists in cotton, soybean remains highly susceptible, exacerbating RN populations in crop rotations. To address this gap, this research investigates the mechanisms of nematode infection focusing on effectors, the specialized proteins secreted by the parasite to manipulate host cell biology and suppress plant immunity. Diphenylalanine (FF) has demonstrated the tendency to form several nanostructures, a subset of which are electrically conductive and have exciting potential applications in biotechnology and nanoelectronics. However, the exact mechanisms underlying this self-assembly process remain largely ambiguous. Atomistic-scale molecular dynamics (MD) simulations allow for the evaluation of atomic-level interactions crucial in understanding the driving thermodynamic forces behind self-assembly, but are too computationally expensive to span the necessary timescale to observe the self-assembly process in action. This reaction occurs between minerals present in aggregates and alkalis in the cement, and produces a gel-like substance called alkali silicate gel. Duchenne muscular dystrophy, or DMD, is a genetic neuromuscular disorder caused by the mutation of the DMD gene, resulting in alterations in the functionality or production of dystrophin, a protein essential to maintain muscle fiber integrity and strength. This mutation leads to progressive muscle degeneration, causing motor loss, cardiopulmonary failure, and ultimately, premature death. Gene therapies using CRISPR aim to replace the mutated gene to provide long-term correction. Changes in land use, such as agricultural expansion, have impacted stream systems by modifying discharge, substrate diversity, channel morphology, and nutrient inputs. The Spokane River Watershed is heavily impacted by human activity, including urban and agricultural development. Rehabilitation efforts are being organized to reintroduce Salmonidae species in the Spokane River Watershed. The first step of this reintroduction is to perform habitat studies to inform allocation of restoration efforts. As part of this effort, I performed benthic macroinvertebrate surveys throughout the watershed.

The Quantum-Well User Entered Simulation Tool (QUEST), originally developed at the University of Texas at Arlington in 2005, is a simulation program built in MATLAB for computing energy eigenvalues and wavefunctions in user-defined semiconductor quantum well structures. This thesis presents a new revision and extension of QUEST with three primary contributions: compatibility updates to the existing MATLAB codebase, intersubband absorption modelling, and a redesigned graphical user interface for ease-of-use in testing.

A Bayesian Late-Fusion Supportability Framework For Rare-Disease Severity Prediction In Glut1 Deficiency Syndrome, Jordan M. Rodriguez

Glucose transporter type 1 deficiency syndrome (GLUT1-DS) is a rare neurometabolic disorder with heterogeneous neurological and developmental severity. Because patient-level severity is not observed as a single validated outcome, this dissertation develops a Bayesian late-fusion supportability framework for constructing and predicting an ordered latent severity phenotype from clinical, genetic, and EEG-derived evidence. The primary target was constructed in a larger clinical cohort using age-5 symptom burden and learning cognition, then assigned to an aligned multimodal prediction cohort. Target-defining variables were excluded from supervised predictors, and models were evaluated using patient-exclusive cross-validation with training-fold preprocessing and fold-wise EEG PCA.

Background: Mesenchymal stromal cell (MSC)-derived extracellular vesicles (EVs) offer a promising acellular therapy for immune modulation, but clinical translation is hindered by variability and storage constraints. Lyophilization (freeze-drying) could enable shelf-stable EV therapeutics, although its effects on biological activity remain incompletely defined.

Study design: We compared the immunomodulatory effects of frozen and lyophilized EVs from bone marrow (BM), adipose tissue (AD), and umbilical cord (UC) MSCs using lipopolysaccharide-stimulated rodent splenocytes and microglial models and human peripheral blood mononuclear cells (PBMCs). Background and aims: Deviations from the population mean in sleep duration have been associated with increased risk for developing dyslipidemia and atherosclerotic cardiovascular disease, but the mechanism of effect is poorly characterized. We performed large-scale genome-wide gene-sleep interaction analyses of lipid levels to identify genetic variants underpinning the biomolecular pathways of sleep-associated lipid disturbances and to suggest possible druggable targets.

Methods: We collected data from 55 cohorts with a combined sample size of 732,564 participants (87 % European ancestry) with data on lipid traits (high-density lipoprotein [HDL-c] and low-density lipoprotein [LDL-c] cholesterol and triglycerides [TG]). Short (STST) and long (LTST) …

Blood flow modifies oxygen availability and biomechanical forces within the vasculature of the embryo as the hematopoietic system develops. The aorta-gonad-mesonephros (AGM) envelops the largest artery in the body and is a critical site for the emergence of hematopoietic stem cells (HSCs). Herein, I discuss the role of hypoxia-inducible factors (HIFs) and force as determinants of metabolism and fate determination. To address the effects of blood flow on hematopoietic development, I employ mouse embryo models and biomimetic culture.
