BACKGROUND: Metabolic Syndrome (MetS) is a clinical diagnosis where patients exhibit three out of the five risk factors: hypertriglyceridemia, low high-density lipoprotein (HDL) cholesterol, hyperglycemia, elevated blood pressure, or increased abdominal obesity. MetS arises due to dysregulated metabolic pathways that culminate with insulin resistance and put individuals at risk to develop various comorbidities with far-reaching medical consequences such as non-alcoholic fatty liver disease (NAFLD) and cardiovascular disease. As it stands, the exact pathogenesis of MetS as well as the involvement of the gastrointestinal tract in MetS is not fully understood. Coinage metal nanoparticles remain an intriguing subject for research due to their industrial versatility. Primary applications of coinage metal nanoparticles include printed electronics, solar panels, and sensors. Inks formulated with the nanoparticles are conductive and thus useful for fabricating sensors. Silver-copper nanoalloy inks are viable for the fabrication of flexible sensing devices for the detection of volatile organic compounds. One of the challenges is the ability to synthesize composition-controllable alloy nanoparticles at room temperature through wet chemical methods and achieve controllable sintering at room temperature. Protein carbonylation is one manifestation of oxidative stress, which is characteristic of many diseases. Our lab has developed several coumarin-hydrazine based fluorescent probes capable of detecting carbonylation in live cells via the formation of hydrazone conjugates. As an effort to expand and improve on these achievements, this work presents new data regarding the synthesis and characterization of three such probes. Two of the probes—julolidine coumarin hydrazide (JCH) and trifluoromethyl coumarin hydrazine (TFCH)—were previously synthesized by our lab. Background: Lung cancer is the leading cause of cancer death, partially owing to its extensive heterogeneity. The analysis of intertumor heterogeneity has been limited by an inability to concurrently obtain tissue from synchronous metastases unaltered by multiple prior lines of therapy.

Methods: In order to study the relationship between genomic, epigenomic and T cell repertoire heterogeneity in a rare autopsy case from a 32-year-old female never-smoker with left lung primary late-stage lung adenocarcinoma (LUAD), we did whole-exome sequencing (WES), DNA methylation and T cell receptor (TCR) sequencing to characterize the immunogenomic landscape of one primary and 19 synchronous metastatic tumors. …

DKK1 inhibits the canonical Wnt signaling pathway that is known to be involved in various cancers. However, whether DKK1 acts as an oncogene or tumor suppressor gene remains controversial. Furthermore, the DKK1-regulating mechanism in gastric cancer has not yet been defined. The aim of this study was to explore whether the ultraconserved region UC.145 regulates epigenetic changes in DKK1 expression in gastric cancer. Microarray analysis revealed that UC.145 exhibited the highest binding affinity to EZH2, a histone methyltransferase. The effects of UC.145 inactivation were assessed in gastric cancer cell lines using siRNA. The results indicated that UC.145 triggers DKK1 methylation …

Amidst global shifts in the distribution and abundance of wildlife and livestock, we have only a rudimentary understanding of ungulate parasite communities and parasite-sharing patterns. We used qPCR and DNA metabarcoding of fecal samples to characterize gastrointestinal nematode (Strongylida) community composition and sharing among 17 sympatric species of wild and domestic large mammalian herbivore in central Kenya. We tested a suite of hypothesis-driven predictions about the role of host traits and phylogenetic relatedness in describing parasite infections. Host species identity explained 27 – 53% of individual variation in parasite prevalence, richness, community composition and phylogenetic diversity. Host and parasite phylogenies …

Background/Purpose: In acute ischemic stroke patients meeting established criteria, thrombolytics may aid in the reperfusion of ischemic brain regions leading to improvement or resolution of neurological deficits. Alteplase is the only FDA approved thrombolytic for acute ischemic stroke. Recent evidence suggests tenecteplase is as safe and efficacious as alteplase, while providing a more favorable pharmacokinetic profile. The purpose of this project is to conduct a review of alteplase compared to tenecteplase for treatment of acute ischemic stroke, following a formulary conversion in the thrombolytic of choice across a large health system.

Providing care to child with a chronic, rare diseases has been identified as a major life stressor with a myriad of negative physical and mental health consequences. These stresses have previously been found to create burden that may impair caregiving efforts and increase the risk of mortality for both the caregiver and affected child. Many rare diseases affect children. It is not known how caregiver burden and the source of stress varies by disease. Qualitative methods have been used to understand caregiver experience with the rare disease, epidermolysis bullosa (EB), but no such studies have occurred in the United States. …

OBJECTIVE: We investigated patient experiences with medication- and test-related cost conversations with healthcare providers to identify their preferences for future informatics tools to facilitate cost-sensitive care decisions.

MATERIALS AND METHODS: We conducted 18 semistructured interviews with diverse patients (ages 24-81) in a Midwestern health system in the United States. We identified themes through 2 rounds of qualitative coding.

RESULTS: Patients believed their providers could help reduce medication-related costs but did not see how providers could influence test-related costs. Patients viewed cost conversations about medications as beneficial when providers could adjust medical recommendations or provide resources. However, cost conversations did not …

Workplace safety is under serious threat due to the increasing trend of active shooter violence in recent years. Therefore, it becomes essential that the safety of a workplace is rigorously and, most importantly, methodologically assessed against active shooter violence. To serve this purpose, this study proposes a machine learning-optimization framework to assess the safety of a building against possible active shooter violence. While much scholarship has considered Deborah Brandt’s concept of sponsors of literacy, there remains a need to consider relationships between literacy sponsors and larger implications of literacy sponsorship at national and institutional levels. Utilizing academic theories, U.S. federal government budgets and financed reports, and discoursal analysis, this dissertation investigates literacy sponsorship at the federal, postsecondary institutional, postsecondary institutional writing programmatic, and individual levels to tease out how, and in what ways, through “enabling” and “supporting” literacy these sponsors also” regulate, suppress, and withhold literacy” (Brandt 166). Precision agriculture continues to be prevalent within row-crop production. The purpose of this study was to investigate the adoption status of precision agricultural practices among selected row-crop (soybean, wheat, corn, cotton, peanuts, and rice) producers in Alabama, Arkansas, Georgia, Louisiana, Mississippi, and Tennessee. Seventy-four percent of row-crop producers surveyed in this study had adopted precision agricultural practices in their farming operations. Eighty-three percent of respondents indicated they were using automated GPS technology such as autosteer, 66% were using manual guidance such as lightbar, 63% variable-rate prescription map, and 34% auto-sprayer boom section or nozzle control. This work is made up of three parts. Part one looks to establish design values for two types of three ply access mats from the U.S. South and Midwest. The mats were subject to 3 point bending tests to determine strength and stiffness values. Values for MOE (Modulus of Elasticity) and MOR (Modulus of Rupture) are reported by region and mat design. Part two tested five species groups of hardwoods for wear resistance and hardness. These species groups include white oak, red oak, ash, sweetgum and hickory.
