DYT-PRKRA (dystonia 16 or DYT-PRKRA) is caused by mutations in the PRKRA gene that encodes PACT, the protein activator of interferon (IFN)-induced double-stranded (ds) RNA-activated protein kinase (PKR). PACT participates in several cellular pathways, of which its role as a PKR activator protein during integrated stress response (ISR) is the best characterized. Previously, we have established that the DYT-PRKRA mutations cause enhanced activation of PKR during ISR to sensitize DYT-PRKRA cells to apoptosis. In this study, we evaluate if the most prevalent substitution mutation reported in DYT-PRKRA patients alters PACT’s functional role in induction of type …

Skeletal stem cells (SSCs, or mesenchymal stromal cells typically referred to as mesenchymal stem cells from the bone marrow) are a dynamic progenitor population that can enter quiescence, self-renew or differentiate depending on regenerative demand and cues from their niche environment. However, ex vivo, in culture, they are grown typically on hard polystyrene surfaces, and this leads to rapid loss of the SSC phenotype. While materials are being developed that can control SSC growth and differentiation, very few examples of dynamic interfaces that reflect the plastic nature of the stem cells have, to date, been developed. Achieving such interfaces is …

BACKGROUND: The role of neuromuscular blocking agents (NMBAs) in coronavirus disease 2019 (COVID-19) acute respiratory distress syndrome (ARDS) is not fully elucidated. Therefore, we aimed to investigate in COVID-19 patients with moderate-to-severe ARDS the impact of early use of NMBAs on 90-day mortality, through propensity score (PS) matching analysis.
METHODS: We analyzed a convenience sample of patients with COVID-19 and moderate-to-severe ARDS, admitted to 244 intensive care units within the COVID-19 Critical Care Consortium, from February 1, 2020, through October 31, 2021. Patients undergoing at least 2 days and up to 3 consecutive days of NMBAs (NMBA treatment), within 48 …

Problematic smartphone use is rising across the world. We tested an intervention with ten strategies that nudge users to reduce their smartphone use, for example by disabling non-essential notifications and changing their display to greyscale. Participants first completed baseline measures of smartphone use, well-being, and cognition before choosing which intervention strategies to follow for 2 to 6 weeks. Study 1 (N = 51) used a pre–post design while study 2 (N = 70) compared the intervention to a control group who monitored their screen time. Study 1 found reductions in problematic smartphone use, screen time, and depressive symptoms …

High-resolution computed tomography (CT) is widely used to assess bone structure under physiological and pathological conditions. Although the analytic protocols and parameters for micro-CT (μCT) analyses in mice are standardized for long bones, vertebrae, and the palms in aging mice, they have not yet been established for craniofacial bones. In this study, we conducted a morphometric assessment of craniofacial bones, in comparison with long bones, in aging mice. Although age-related changes were observed in the microarchitecture of the femur, tibia, vertebra, and basisphenoid bone, and were more pronounced in females than in males, the microarchitecture of both the interparietal bone …

Biochar has proven its potential in removing heavy metal ions from water. The objective of this study was to evaluate locally obtained biomass feedstocks for biochar production and their efficiency as a sorbent for aqueous lead (Pb2+) removal. The biomass feedstocks consisted of avocado seed, avocado peel, grapefruit peel, and brown seaweed, which represent agricultural and marine biomasses. The biochar materials were produced in two different methods: (1) a laboratory tube furnace at 300 °C and (2) a Do-It-Yourself (DIY) biochar maker, “BioCharlie Log”. The biochars were characterized for selected physicochemical properties, and batch adsorption tests with 10 …
Technical And Economic Evaluation Of The First Ever Polymer Flood Field Pilot To Enhance The Recovery Of Heavy Oils On Alaska's North Slope Via Machine Assisted Reservoir Simulation, Cody D. Keith
Engineering
Polymer flooding has become globally established as a potential enhanced oil recovery method for heavy oils. To determine whether this technology may be useful in developing the substantial heavy oil resources on the Alaska North Slope, a polymer flood field pilot commenced at the Milne Point Unit in August 2018. This study seeks to evaluate the results of the field pilot on a technical and economic basis. A reservoir simulation model is constructed and calibrated to predict the oil recovery performance of the pilot through machine-assisted reservoir simulation techniques. To replicate the early water breakthrough observed during waterflooding, transmissibility contrasts …

Design Of A Solar Assisted Flying Wing Small Unmanned Aerial System, Zion A. Alioto
Engineering
The purpose of this project is to further advance Unmanned Aerial System flight capabilities with solar assisted flight. To reduce drag and increase flight efficiency, a flying wing configuration was selected and developed modeling the tradeoff between additional drag and power received from photovoltaic cells. To initiate the design process, strategies based on the needs were implemented to determine initial constraints. As the design methodology progressed, the selected parameters were finetuned and optimized to best meet the performance requirements and simplify the fabrication process. The chosen design was modeled for computational fluid dynamics simulations, stability and control analysis, and finite …
The Multidimensional Test Anxiety Scale: A Latent Profile Analysis And An Examination Of Measurement Invariance, Gabrielle Francis
The Multidimensional Test Anxiety Scale: A Latent Profile Analysis And An Examination Of Measurement Invariance, Gabrielle Francis
USF Tampa Graduate Theses and Dissertations
Standardized testing is an integral part of the English and American education systems. The objectives of these tests are to evaluate students, teachers, and schools. However, this evaluation has unintended consequences, one of which is test anxiety. Over the last 50 years, there has been an increase in studies on test anxiety because of the widespread use of standardized tests (Hembree, 1988; von der Embse et al., 2019). However, two areas that have seen little attention are the measurement invariance of test anxiety scales across demographic groups, and the creation of classification standards for these test anxiety scales to increase …

Feedback To Feed Forward: Creating Formative Feedback To Improve Student Writing, Adam R. Huttenga
Feedback To Feed Forward: Creating Formative Feedback To Improve Student Writing, Adam R. Huttenga
Masters Projects
Feedback to students has been highlighted in the literature as an area where improvements are needed. Students need high quality, specific, usable, and timely feedback. They also need guidance and tools to help them engage with, reflect on, and learn from that feedback in order to close the gap between their writing and the expected standards. In order to help students improve their writing, teachers must develop written feedback that is formative and feeds forward into future writing assignments. This project explores how effective formative feedback can be achieved within a social constructivist theoretical approach to learning, in which an …
How Federal Pandemic Relief Helped Replenish State Unemployment Reserves, Christopher J. O'Leary, Kenneth J. Kline
How Federal Pandemic Relief Helped Replenish State Unemployment Reserves, Christopher J. O'Leary, Kenneth J. Kline
Upjohn Institute Policy Papers
Unemployment insurance (UI) is a joint federal-state program that pays temporary partial earnings replacement to involuntarily unemployed workers while they seek reemployment. During the COVID-19 pandemic, UI claims surged and became a primary source for income replacement for workers who lost their jobs. However, despite previous federal incentives for states to shore up their UI funding reserves, the scale of claims during the pandemic was unprecedented, and the federal government needed to step in to help pay not only for direct, expanded benefits but for additional assistance to states themselves. Although this effort helped backstop the successful operation of states’ …

Extended interval dosing of tobramycin is recommended for treatment of pulmonary exacerbations in adults and older children with cystic fibrosis (CF), but data are limited in patients less than 5 years of age. We performed a retrospective population pharmacokinetic (PK) analysis of hospitalized children with CF <5 years of age prescribed intravenous tobramycin for a pulmonary exacerbation from March 2011 to September 2018 at our hospital. Children with normal renal function who had ≥1 tobramycin concentration available were included. Nonlinear mixed effects population PK modeling was performed using NONMEM using data from the first 48 h of tobramycin treatment. Monte Carlo simulations were implemented to determine the fraction of simulated patients that met published therapeutic targets with regimens of 10-15 mg/kg/day once-daily dosing. Fifty-eight patients received 111 tobramycin courses (range 1-9/patient). A two-compartment model best described the data. Age, glomerular filtration rate, and vancomycin coadministration were significant covariates on tobramycin clearance. The typical values of clearance and central volume of distribution were 0.252 L/hr/kg^0.75 and 0.308 L/kg, respectively. No once-daily regimens achieved all pre-specified targets simultaneously in >75% of simulated subjects. A dosage of 13 mg/kg/dose best met the predefined targets of C

Increases in snack consumption associated with Westernized lifestyles provide an opportunity to introduce nutritious foods into poor diets. We describe two 10-wk-long open label, single group assignment human studies that measured the effects of two snack prototypes containing fiber preparations from two sustainable and scalable sources; the byproducts remaining after isolation of protein from the endosperm of peas and the vesicular pulp remaining after processing oranges for the manufacture of juices. The normal diets of study participants were supplemented with either a pea- or orange fiber-containing snack. We focused our analysis on quantifying the abundances of genes encoding carbohydrate-active enzymes …
Necessary Myths, Jessica Ramsey
Necessary Myths, Jessica Ramsey
MFA in Illustration & Visual Culture
My thesis essay was inspired by my search for a belief system that could transform despair over what will be lost through climate change into valuing what we still have. In researching the earliest iterations of belief structures, I came across the Maros-Pangkep cave paintings. These paintings are the oldest known works of art, and by my interpretation the first evidence of religious life. They are a series of representational paintings which tell a story, and I was inspired to emulate this methodology in my own exploration of belief.
My essay investigates the relationship between images and religion. Through W.J.T …
Interpretations Of Climate Change On Grazing Systems: The Comparison Of Arctic And Subarctic Carex, Iris Cato Harritt
Interpretations Of Climate Change On Grazing Systems: The Comparison Of Arctic And Subarctic Carex, Iris Cato Harritt
Biological Sciences
Stresses imposed by climate change are altering arctic and subarctic ecosystem structure and function. On the Yukon-Kuskokwim River Delta (YKD) in subarctic western Alaska, Pacific Black Brant geese (Branta bernicla nigricans) are losing their available grazing lawns of shortstatured Carex subspathacea due to its conversion into a taller, less nutritious growth form. However, C. subspathacea on Alaska's Arctic Coastal Plain (ACP) grows in extensive lawns that remain short even when ungrazed. Our goal was to compare the growth responses between arctic C. subspathacea and subarctic C. subspathacea when grown in arctic and subarctic conditions. We used reciprocal common gardens to …
Drivers And Mechanisms Of Migration In An Arctic Caribou Herd, Matthew D. Cameron
Drivers And Mechanisms Of Migration In An Arctic Caribou Herd, Matthew D. Cameron
Biological Sciences
Migration is one of the world's great natural wonders and the scale of some migratory journeys is astounding. Yet migration is globally imperiled and effective conservation of the remaining migrations will require a thorough understanding of the drivers and mechanisms underlying how migrants complete such journeys. In this dissertation, I present three chapters that sought to better understand spring and autumn migration for the Western Arctic Herd, a population of barren-ground caribou (Rangifer tarandus) that complete some of the longest terrestrial migrations on the planet. In the first chapter, I applied and validated an analytical method to infer parturition events …
Snow As Structural Habitat For Wolverines In A Changing Arctic, Thomas Rutherford Glass
Snow As Structural Habitat For Wolverines In A Changing Arctic, Thomas Rutherford Glass
Biological Sciences
Arctic snowpack provides critical wintertime habitat for animals to facilitate thermoregulation and avoid predators. Wolverines (Gulo gulo) are iconic among such animals, relying on snow burrows for resting sites and reproductive dens. Most of the knowledge regarding this mesocarnivore's association with snow, however, has so far originated in more southerly latitudes. In this dissertation, I investigated Arctic wolverines' behaviors associated with snow, focusing on how specific snow properties influence resting, habitat selection, and avoiding predators. Motivated by the paucity of published descriptions of wolverine resting burrows and reproductive dens on tundra, I first described terrain features and architecture of such …
Seeing The Forest Through The Trees: How Site Conditions Mediate White And Black Spruce Responses To Climate In Interior Alaska, E. Fleur Nicklen
Seeing The Forest Through The Trees: How Site Conditions Mediate White And Black Spruce Responses To Climate In Interior Alaska, E. Fleur Nicklen
Biological Sciences
The boreal forest provides essential ecosystem services and helps regulate global climate. With climate change occurring at a faster rate at high latitudes, including in the boreal forest biome, it is critical to understand how boreal forests are responding to these unprecedented changes. Despite much effort, uncertainty remains as to how boreal forest productivity has and will change with ongoing climate changes. Some of the uncertainty reflects the complex mosaic of regional climatic patterns, direct and indirect species-specific responses to regional climate, and heterogenous local site conditions that affect boreal forest productivity. I focused on the latter uncertainty: the potential …

Analyzing The News Media Trust At Different Levels And Its Relationship With Civic Engagement, Dina Ahmed Ali
Analyzing The News Media Trust At Different Levels And Its Relationship With Civic Engagement, Dina Ahmed Ali
Theses and Dissertations
"People want trusted news," Faulconbirdge said. Trust has long been deemed a critical element that influences audiences' relationship with the news media. Particularly after the percentage of online users is rapidly expanding around the world during the COVID-19 outbreak and its continuous existence. The new technologies and social media have the power to polarize, manipulate, and entrench public opinion and response. Where trust has eroded, conspiracy theories flourish, the media's authority has waned, and emotions have taken precedence over facts. With high-choice media ecosystems in today's world, news media trust and its relationship with civic engagement have taken on new …

The Prime Aesthetic: Exploratory Research On The Relationship Between The Imprinting Of Aesthetic Dress Preferences And The Sensitive Period Of Emerging Adulthood, Jeremy Bernardoni
LSU Doctoral Dissertations
The purpose of this research was to explore the theoretical concept of Prime Aesthetic across generational cohorts through the investigation of individual aesthetic dress preferences (ADP), which are formed during the Prime of Life, and their potential influence on present-day ADP. The Prime Aesthetic concept is defined as a theoretical concept representative of the cumulative ADP imprinted from all influences during an individual’s Prime of Life. The Prime Aesthetic was derived from Holbrook and Schindler’s (1989) study on the development of musical tastes during late adolescence and early adulthood and linked to theories on imprinting during sensitive periods (Bornstein, 1989) …
