Targeted protein degradation (TPD) has rapidly emerged as a therapeutic modality to eliminate previously undruggable proteins by repurposing the cell’s endogenous protein degradation machinery. However, the susceptibility of proteins for targeting by TPD approaches, termed “degradability”, is largely unknown. Here, we developed a machine learning model, model-free analysis of protein degradability (MAPD), to predict degradability from features intrinsic to protein targets. MAPD shows accurate performance in predicting kinases that are degradable by TPD compounds [with an area under the precision–recall curve (AUPRC) of 0.759 and an area under the receiver operating characteristic curve (AUROC) of 0.775] and is …

PURPOSE: We investigated the feasibility of thoracic spine stereotactic body radiotherapy (SBRT) using the Elekta Unity magnetic resonance-guided linear accelerator (MRL) in patients who received prior radiotherapy. We hypothesized that Monaco treatment plans can improve the gross tumor volume minimum dose (GTVmin) with spinal cord preservation and maintain consistent plan quality during daily adaptation.
METHODS: Pinnacle clinical plans for 10 patients who underwent thoracic spine SBRT (after prior radiotherapy) were regenerated in the Monaco treatment planning system for the Elekta Unity MRL using 9 and 13 intensity-modulated radiotherapy (IMRT) beams. BACKGROUND: There is an unmet need for COVID-19 prevention in patient populations who have not mounted or are not expected to mount an adequate immune response to complete COVID-19 vaccination. We previously reported that a single subcutaneous 1200 mg dose of the monoclonal antibody combination casirivimab and imdevimab (CAS + IMD) prevented symptomatic SARS-CoV-2 infections by 81·4% in generally healthy household contacts of SARS-CoV-2-infected individuals over a 1-month efficacy assessment period. Here we present additional results, including the 7-month follow-up period (months 2-8), providing additional insights about the potential for efficacy in pre-exposure prophylaxis settings.

Study 309/KEYNOTE-775 is a phase 3 open-label, randomized trial of lenvatinib plus pembrolizumab versus treatment of physician's choice (TPC) in patients with advanced endometrial cancer with progression after platinum-based therapy. Primary endpoints of superiority for lenvatinib plus pembrolizumab were met for progression-free survival (PFS) and overall survival (OS) in all-comers (ie, regardless of mismatch repair [MMR] status) and patients with MMR proficiency (pMMR). OBJECTIVE: Social networking has been shown to improve health outcomes in certain patient populations. While patients with rheumatoid arthritis (RA) increasingly use social networking to communicate with peers, the effects of these interactions are largely unknown.
METHODS: In a randomized controlled trial, we compared RA patients who participated in a social networking group moderated by peer leaders and who had access to a static website offering RA materials with a control group, who only had access to the website. BACKGROUND: Use of surveillance mammography and magnetic resonance imaging (MRI) has been understudied among women with variant of uncertain significance (VUS) compared to pathogenic and likely pathogenic variants (P/LP).
METHODS: Using data from two cancer settings, we calculated use of risk-reducing mastectomy (RRM) and surveillance during each 13-month span after genetic testing up to 6 years afterwards for a cohort of genetically elevated risk women.

OBJECTIVES: Pathologists have routinely observed distinct histologic patterns of growth in early-stage lung adenocarcinoma (LUAD), which have been suggested to be associated with prognosis. Herein, we investigated the relationship between LUAD patterns of growth, as defined by the updated international association for the study of lung cancer (IASLC) grading criteria, and differences in the tumor immune microenvironment to identify predictors of response to immunotherapy.

The backscattering cross section of cylindrical and elliptical disk-shaped scatterers was investigated in this study, utilising a new numerical solution method called the relaxed hierarchical equivalent source algorithm (RHESA). The results were compared with the backscattering cross section of similar cases, using analytical method validation from literature. Variation in the local thermal history during the Laser Powder Bed Fusion (LPBF) process in Additive Manufacturing (AM) can cause micropore defects, which add to the uncertainty of the mechanical properties (e.g., fatigue life, tensile strength) of the built materials. In-situ sensing has been proposed for monitoring the AM process to minimize defects, but successful minimization requires establishing a quantitative relationship between the sensing data and the porosity, which is particularly challenging with a large number of variables (e.g., laser speed, power, scan path, powder property). Physics-based modeling can simulate such an in-situ sensing-porosity relationship, but it is computationally costly. …

Trials were conducted in two experimental runs at the Purdue University Horticulture Greenhouses, West Lafayette, IN, to determine ‘Redefined Murray Mitcham’ peppermint tolerance to tiafenacil. Established peppermint in 20-cm-diameter polyethylene pots was subjected to a simulated harvest by removing aboveground biomass at the substrate surface; then, tiafenacil was applied at 0, 25, 50, 100, and 200 g ai ha−1. Visible crop injury, height, and aboveground dry biomass data were subjected to regression analysis to generate predictive models. Liquid Composite Molding (LCM) techniques including the Resin Transfer Molding (RTM) and Vacuum Assisted Resin Transfer Molding (VARTM) are gaining significant importance for fabricating aerospace and automotive composite parts, owing to the low investment costs. During the curing process, the resin undergoes a property change due to cross-linking of polymers, where it transitions from the liquid state to the solid state. Further, during the cooling process, there is a change in the glass transition temperature, resulting in residual stress and strains. We present a study of the dynamic interactions between actors located on complex networks with scale-free and hierarchical scale-free topologies with assortative mixing, that is, correlations between the degree distributions of the actors. The actor’s state evolves according to a model that considers its previous state, the inertia to change, and the influence of its neighborhood. We show that the time evolution of the system depends on the percentage of cooperative or competitive

Some individuals can flexibly adapt to life’s changing demands while others, in particular those with Autism Spectrum Disorder (ASD), find it challenging. The successful dissemination of the Peer-Led Team Learning (PLTL) model at multiple institutions of higher education, in the United States and other countries, is reflected in the number of publications (see www.pltlis.org). However, many PLTL campus programs are no longer active or exist. This may be due, more recently, to the COVID pandemic and its disruptions. Context: This project investigates the role of state-level institutions in explaining variation in population health in the American states. Although cross-national research has established the positive effects of democracy on population health, little attention has been given to subnational units. The authors leverage a new data set to understand how political accountability and a system of checks and balances are associated with state population health. Methods: The authors estimate error correction models and two-way fixed effects models to estimate how the strength of state-level democratic institutions is associated with infant mortality rates, life expectancy, and midlife mortality. This mixed-methods study investigated the perceptions educators have during the initial stages of implementing the Multi-Tiered System of Support (MTSS) model. A survey and semi-structured interviews were collected to identify if the perceived barriers found in a rural context were consistent with prior research. Quantitative survey data indicated that participants felt confident about their practicing foundational skills of MTSS, and felt MTSS will benefit both staff and students. However, quantitative survey results also indicated a discrete difference in the data between the answers regarding participant confidence, knowledge, and the answers regarding attitude regarding time, resources, and leadership.
