Inhibiting Glutamine Utilization Creates A Synthetic Lethality For Suppression Of Atp Citrate Lyase In Kras-Driven Cancer Cells, Ahmet Hatipoglu, Deepak Menon, Talia Levy, Maria A. Frias, David A. Foster

Publications and Research

Metabolic reprogramming is now considered a hallmark of cancer cells. KRas-driven cancer cells use glutaminolysis to generate the tricarboxylic acid cycle intermediate α -ketoglutarate via a transamination reaction between glutamate and oxaloacetate. We reported previously that exogenously supplied unsaturated fatty acids could be used to synthesize phosphatidic acid–a lipid second messenger that activates both mammalian target of rapamycin (mTOR) complex 1 (mTORC1) and mTOR complex 2 (mTORC2). A key target of mTORC2 is Akt–a kinase that promotes survival and regulates cell metabolism. We report here that mono- unsaturated oleic acid stimulates the phosphoryla tion of ATP citrate lyase (ACLY) at …

BACKGROUND: The current COVID-19 pandemic and the previous SARS/MERS outbreaks of 2003 and 2012 have resulted in a series of major global public health crises. We argue that in the interest of developing effective and safe vaccines and drugs and to better understand coronaviruses and associated disease mechenisms it is necessary to integrate the large and exponentially growing body of heterogeneous coronavirus data. Ontologies play an important role in standard-based knowledge and data representation, integration, sharing, and analysis. Accordingly, we initiated the development of the community-based Coronavirus Infectious Disease Ontology (CIDO) in early 2020.

RESULTS: As an Open Biomedical Ontology …

Diversity In Children's Books, Hala Qarini

Dissertations, Theses, and Projects

Abstract

This project aimed to determine whether Leveled Literacy Intervention (LLI) curriculum represents diversity in the books in a way that reflects the diversity of students in Fargo Public Schools (FPS). I used the green system in LLI curriculum level B-level I, which represents first grade. For this study, I analyzed eighty books total (lesson 21-lesson 100) based on the Fargo Public Schools definition of diversity. This method was a good match for my research question because it included gender/age, race/ethnicity, socioeconomic, religion, disability, and culture which Fargo Public School includes in its description.

After reviewing and completing a content …

Not Just A Trick: A Survey Study Exploring How ‘Exposing’ Exhibition Visitors To Science Of Magic Concepts Impacts Their Appreciation Of Magic, Gabriel Medeiros, Mathew Tompkins, Steve Bagienski, Gustav Kuhn

School of Psychology

The recent rise in scientific research on magic raises important issues about the impact that the dissemination of magic knowledge has on people’s appreciation of magic.Deception, secrecy, and mystery are inexorably intertwined with the idea of performance magic. Magicians traditionally do not reveal their secret methods to non-magicians. This study used a survey to assess how people’s appreciation of magic was impacted by a magic exhibition designed to highlight and reveal the psychological mechanisms that underpin magic. Visitors to the exhibition were asked to rate the impact of the exhibition on a range of measures assessing people’s interest and appreciation …

Biological Networks And Complexity In Early-Onset Motor Neuron Diseases, Matthew E. R. Butchbach, Rod C. Scott Oct 2022

Biological Networks And Complexity In Early-Onset Motor Neuron Diseases, Matthew E. R. Butchbach, Rod C. Scott

Department of Pediatrics Faculty Papers

Motor neuron diseases (MNDs) are neuromuscular disorders where the spinal motor neurons–either the cell bodies themselves or their axons–are the primary cells affected. To date, there are 120 different genes that are lost or mutated in pediatric-onset MNDs. Most of these childhood-onset disorders, aside from spinal muscular atrophy (SMA), lack viable therapeutic options. Previous research on MNDs has focused on understanding the pathobiology of a single, specific gene mutation and targeting therapies to that pathobiology. This reductionist approach has yielded therapeutic options for a specific disorder, in this case SMA. Unfortunately, therapies specific for SMA have not been effective against …

Genetic Conditions Of Short Stature: A Review Of Three Classic Examples, Merlin G Butler, Bradley S Miller, Alicia Romano, Judith L. Ross, M Jennifer Abuzzahab, Philippe Backeljauw, Vaneeta Bamba, Amrit Bhangoo, Nelly Mauras, Mitchell Geffner

Department of Pediatrics Faculty Papers

Noonan, Turner, and Prader-Willi syndromes are classical genetic disorders that are marked by short stature. Each disorder has been recognized for several decades and is backed by extensive published literature describing its features, genetic origins, and optimal treatment strategies. These disorders are accompanied by a multitude of comorbidities, including cardiovascular issues, endocrinopathies, and infertility. Diagnostic delays, syndrome-associated comorbidities, and inefficient communication among the members of a patient's health care team can affect a patient's well-being from birth through adulthood. Insufficient information is available to help patients and their multidisciplinary team of providers transition from pediatric to adult health care systems. …

Staying Out Of Trouble? Effect Of High School Career Counseling On Crime, Stephen Barnes, Louis-Philippe Beland, Swarup Joshi, Barton Willage Oct 2022

Staying Out Of Trouble? Effect Of High School Career Counseling On Crime, Stephen Barnes, Louis-Philippe Beland, Swarup Joshi, Barton Willage

Economics Faculty Works

Human capital-based models of crime imply that increasing educational attainment will decrease crime. A growing body of empirical work has supported this hypothesis, however the evidence is still limited. We investigate if a high school career coaching program, Career Compass of Louisiana, decreases youth crime during and after high school. Career Compass provides students counseling services regarding college admissions, financial aid, and career exploration. We use student-level administrative education and crime convictions data and a difference-in-differences empirical strategy to test for an education-crime relationship among high school seniors. While existing research shows the Career Compass program increases post-secondary enrollment, we …

School of Veterinary and Biomedical Sciences: Faculty Publications

Background: Moraxella bovis and Moraxella bovoculi both associate with infectious bovine keratoconjunctivitis (IBK), an economically significant and painful ocular disease that affects cattle worldwide. There are two genotypes of M. bovoculi (genotypes 1 and 2) that differ in their gene content and potential virulence factors, although neither have been experimentally shown to cause IBK. M. bovis is a causative IBK agent, however, not all strains carry a complete assortment of known virulence factors. The goals of this study were to determine the population structure and depth of M. bovis genomic diversity, and to compare core and accessory genes and …

Insulin resistance (IR) and chronic low-grade inflammation are risk factors for chronic diseases including type 2 diabetes (T2D) and cardiovascular disease. This study aimed to investigate two dietary indices: Mediterranean Diet Score (MDS) and Dietary Inflammatory Index (DII®), and their associations with direct measures of glucose metabolism and adiposity, and biochemical measures including lipids, cytokines and adipokines in overweight/obese adults. This cross-sectional study included 65 participants (males = 63%; age 31.3 ± 8.5 years). Dietary intake via 3-day food diaries was used to measure adherence to MDS (0–45 points); higher scores indicating adherence. Energy-adjusted DII (E-DII) scores were calculated with …

Perturbative Theoretical Model Of Electronic Transient Circular Dichroism Spectroscopy Of Molecular Aggregates, Paul C. Arpin, Daniel B. Turner Oct 2022

Perturbative Theoretical Model Of Electronic Transient Circular Dichroism Spectroscopy Of Molecular Aggregates, Paul C. Arpin, Daniel B. Turner

Materials Science and Engineering Faculty Publications and Presentations

A chiral analog of transient absorption spectroscopy, transient circular dichroism (TCD) spectroscopy is an emerging time-resolved method. Both spectroscopic methods can probe the electronic transitions of a sample, and TCD is additionally sensitive to the dynamic aspects of chirality, such as those induced by molecular excitons. Here, we develop a theoretical description of TCD for electronic multi-level models in which the pump pulse is linearly polarized and probe pulse is alternately left- and right-circularly polarized. We derive effective response functions analogous to those often used to describe other four-wave mixing methods and then simulate and analyze TCD spectra for three …

Maternal Prenatal Cortisol Trajectories Predict Accelerated Growth In Infancy, Jennifer Hahn-Holbrook, Elysia Poggi Davis, Curt A. Sandman, Laura M. Glynn

Psychology Faculty Articles and Research

Higher maternal cortisol in pregnancy has been linked to childhood obesity. Much of the previous research has been limited in that cortisol in pregnancy is only measured at one time-point, precluding the ability to examine critical timing effects of prenatal maternal cortisol. To fill this gap, this longitudinal study measured maternal plasma cortisol at 15, 19, 25, and 31 weeks of pregnancy, and assessed infant body mass index percentile (BMIP)1 at birth, 3, 6, 12, and 24 months in 189 mother-infant pairs. Three distinct patterns of maternal cortisol in pregnancy (typical, steep, and flat trajectories) were identified using general growth …

Visualization Of Cathode Spot Control Using Laser Irradiation And Oxide Addition In Wire Arc Additive Manufacturing Of Titanium Alloys, Tae Hyun Lee, Cheolhee Kim, Je Hoon Oh, Dong Hyuck Kam

Mechanical and Materials Engineering Faculty Publications and Presentations

Arc instability is one of the most critical problems in gas-metal-arc (GMA) based wire arc additive manufacturing of titanium (Ti) alloys. It can result in a poor bead surface, surface oxidation, and spattering. In particular, the relocation of the cathode spot area is the main cause of big spatters because of the high thermal energy of the molten droplet at the molten pool surface. In this study, two cathode spot control techniques were applied using auxiliary laser heating and prelaid oxides, and the behaviors of the cathode spots and arc were visualized using high-speed photography. When the laser beam was …

Married Working Women And Work-Family Interface In Urban Ghana : A Holistic Approach, Kwaku Abrefa Busia Oct 2022

Married Working Women And Work-Family Interface In Urban Ghana : A Holistic Approach, Kwaku Abrefa Busia

Lingnan Theses (MPhil & PhD)

The intersection of work and family has gained traction across various disciplines in an attempt to study how work and family life affect individuals, families, organisations and societies. Nonetheless, most studies on work-family interface (WFI) remain dominated by theories, concepts and experiences rooted in white, middle class and homogenous individuals in Western societies. Owing to these concerns, there have been widespread calls for more cross-cultural studies that capture the plurality of work-family realities and conceptualisation, especially from the Global South. Emphasis has also been made on the need to expand workfamily research beyond the often-studied professional workers and their families …

Department of Medical Oncology Faculty Papers

Hypomethylating agents (HMA) prolong survival and improve cytopenias in individuals with higher-risk myelodysplastic syndrome (MDS). Only 30-40% of patients, however, respond to HMAs, and responses may not occur for more than 6 months after HMA initiation. We developed a model to more rapidly assess HMA response by analyzing early changes in patients’ blood counts. Three institutions’ data were used to develop a model that assessed patients’ response to therapy 90 days after the initiation using serial blood counts. The model was developed with a training cohort of 424 patients from2 institutions and validated on an independent cohort of 90 patients. …
