An accelerated transformation pipeline has been developed for functional genomics and trait gene analysis in Sorghum bicolor based on visual selection using the AmCyan reporter. Here, we combine a GRF4–GIF1 morphogenic regulator with a constitutively expressed AmCyan fluorescent reporter to enable early visual identification of transformed tissues from immature embryo (IE) explants and to shorten time‑to‑regeneration. We demonstrate a practical workflow in which AmCyan‑positive embryogenic clusters are visually recognized and excised 24-48 days after infection (DAI), and then rapidly regenerated directly into plants, in the absence of antibiotic/herbicide selection exposure. Cerebral palsy often results from perinatal hypoxia-ischemia and is characterized by motor and sensory dysfunction associated with altered serotonergic signaling in the spinal cord. The 5-HT₇ receptor, a G-protein-coupled receptor implicated in both motor control and nociceptive modulation, may thus play a critical role in these post-injury changes. However, the regional and developmental patterns of expression of 5-HTR₇ following HI injury are not well understood. Reliable Agrobacterium mediated transformation of Sorghum remains constrained by low transformation frequencies and the requirement for high quality immature embryos (IEs). A useful conceptual framework describes transformation efficiency T=V(Cd×Ci×Cr), as the product of DNA delivery (Cd), stable integration (Ci), regeneration competence (Cr), and experiment specific variables (V). Using this framework, in a multi-year study we systematically evaluated parameters predicted to influence DNA delivery and regeneration. Standard IE workflows established baseline developmental dynamics of somatic embryogenesis and regeneration. This work discusses how to directly extract features from complex-valued spectra using an energy-based probability distribution that represents complex-valued data. As an extension of the well-known restricted Boltzmann machine (RBM), the proposed model, the complex-valued restricted Boltzmann machine (CRBM), is made to deal with complex valued visible units. With the use of contrastive divergence (CD) or Gibbs sampling, the CRBM, like the RBM, develops a grasp of the relationships between visible and hidden units without requiring connections between units in the same layer. Green synthetized nanoparticles have been the object of study for their use as fertilizers in the agricultural field. Among them, manganese oxide nanoparticles (MnONPs) have been used to increase the growth and biomass of rice seedlings, mung bean, moringa plants, among others. In this work, we explore the biosynthesis and purification of MnONPs for these environmental applications, seeing as the biosynthesis of MnONPs consumes less thermal energy compared to chemical synthesis methods. Abstract
The pre-participation school or sports physical, introduced in the mid-20th century, remains a cornerstone of adolescent health screening, primarily aimed at ensuring athletic safety. While most evaluations reveal benign conditions, these encounters occasionally uncover critical yet asymptomatic disorders. This report highlights the incidental discovery of Loeys–Dietz syndrome (LDS) type 4 in a 17-year-old male during a routine sports physical, underscoring the clinical value of thorough cardiovascular assessment and recognition of subtle phenotypic cues in preventive settings.

White matter—the fundamental structure underlying network connectivity—lies at the heart of the brain’s computational power. White matter is organized into fiber tracts, or bundles, in both human and nonhuman primate brains. These bundles consist of long-range axonal projections with a set of shared origin and termination points and exhibit a predictable organization. However, the organization of white matter in the mouse brain remains relatively unknown. Background: Among individuals with type 2 diabetes (T2D), lifestyle improvements can restore glycemic control, yet few studies have examined deprescribing in settings where it was necessitated by improvements in health. This study aimed to (1) identify instances of medication deprescribing among adults with T2D in a primary care setting where patients had access to lifestyle medicine (LM), (2) document lifestyle changes among deprescribed patients, (3) assess changes in body mass index (BMI), glucose, and hemoglobin A1c (HbA1c) following deprescribing, and (4) assess the safety of deprescribing in the context of LM-informed care by identifying adverse events.

Cerebellar ataxias are characterized by impaired motor coordination resulting from neuronal dysfunction within the cerebellum. The mechanisms underlying this pathology and its cerebellar-specific neurodegeneration remain unknown. We uncover how a gain-of-function canonical transient receptor potential member 3 (TRPC3) mutation, coupled with a cerebellum-specific isoform, stabilizes the channel’s open state, resists the leading inhibitor Pyr3, and drives calcium-dependent cell death. Restoring calcium homeostasis by expressing a Purkinje cell calcium pump improves cell viability. Transgenic expression of the TRPC3 hypermorphic variant in Caenorhabditis elegans induces neurodegeneration, confirming its pathogenicity across species. Background: Firearm secure storage (ie, storing firearms unloaded and locked) is recommended to reduce unintentional injuries and suicides. However, the relationship between psychological states, such as social isolation, and firearm secure storage practices is under-researched.
Methods: Data are from 7136 individuals with firearms in their households from the 2022 Behavioral Risk Factor Surveillance System. Multinomial logistic regression was used to explore the relationship between social isolation and firearm storage.
Results: Among respondents, 71.6% reported storing firearms unloaded, 14.2% stored firearms loaded and locked and 14.2% stored firearms loaded and unlocked. Most respondents reported feeling 'never' (40.7%) or 'rarely' (33.3%) socially …

This study investigates the impact of a computer-based divided attention task on spoken language and compares the impact between younger (18–30 years) and older (55–82 years) adults. Sixty adults participated in a speaking task and two computer tasks which varied in difficulty. The Level 1 computer task involved highlighting each instance of the word “the” and the Level 2 computer task involved highlighting each instance of the word “the” while also underlining each instance of the word “a”. The Principle of Sufficient Reason has a long history, but basically put, states that everything that exists has a reason or explanation for its existence. This principle has been a controversial one as it has been used to argue for the existence of God and have been accused of entailing determinism. In this thesis, I will be defending a version of the PSR advocated by Alexander Pruss, that all true contingent propositions need an explanation. I will also argue that the principle of sufficient reason should be restricted to our phenomenal experiences, just like Kant does.

Purpose: X-ray grating interferometry is a form of X-ray phase contrast imaging capable of simultaneously measuring attenuation, differential-phase, and dark-field images arising from X-ray absorption, refraction, and small-angle scattering, respectively. These additional contrast mechanisms have potential clinical and industrial applications, particularly for lung tissue characterization and disease identification. As grating interferometers approach clinical viability, key challenges include reducing X-ray dose, mitigating mechanical instability, and correcting spectral artifacts resulting from beam hardening.

Theta series encode the number of representations of integers by quadratic forms in their Fourier coefficients and connect classical counting problems to the theory of modular forms and elliptic curves. On the other hand, theta series connect with hypergeometric series via Ramanujan’s theory of elliptic functions to alternative bases. This dissertation explores these connections and applies theta series to solve problems involving representations of integers and special values of $L$-functions. We analyze the theta series associated to the quadratic form $Q(\Vec{x})=x_1^2+x_2^2+x_3^2+x_4^2$ with congruence conditions on $x_i$ modulo $2,3,4$ and $6$. Large language models have fundamentally challenged traditional methods of verifying doctoral competency as AI-generated text becomes increasingly difficult to distinguish from human scholarship. This paper argues that thesis committees and doctoral supervisors must reclaim the oral defence as a critical checkpoint for assessing authentic threshold crossing rather than a ceremonial rite of passage. Drawing on historical examples from medieval oral disputations through to the rise of written theses, this paper asserts the necessity of returning to rigorous oral assessment. Grounded in S-D logic and value co-creation theories within the context of consumer decision-making, this study examines how reality technological attributes influence consumer continuous use intention as well as purchase intention. Neuronal function requires high energy expenditure that is likely customized to meet specific signaling demands. However, little is known about diversity of metabolic homeostasis among divergently-functioning types of neurons. To this end, we examined retinal ganglion cells (RGCs), a population of closely related, yet electrophysiologically distinct excitatory projection neurons. Using in vivo 2-photon imaging to measure ATP with single cell resolution, we identified differential homeostatic energy maintenance in the RGC population that correspond to distinct RGC types. Access to justice is “the ability of all people to seek and obtain effective remedies through accessible, affordable, … and culturally competent institutions of justice.”1 Historically, patients and physicians have worked together to protect the integrity of medical care as well as patients’ rights by challenging unlawful government actions in court. Yet the US Supreme Court recently issued decisions that may impede access to justice, potentially empowering federal, state, and local governments to interfere with evidence-based care. INTRODUCTION: Inappropriate antibiotic prescribing is a major concern in low- and middle-income countries (LMICs), particularly at the primary care level. The WHO AWaRe Antibiotic Handbook was introduced to promote rational antibiotic use, yet its real-world feasibility and potential impact remain underexplored. Our study evaluated the impact and usefulness of the WHO AWaRe Handbook training among private primary care providers (PCPs) in Patna, India.
METHODS: We conducted a pre-post pilot study among 145 private PCPs (40 formal PCPs (FPs) and 105 informal PCPs (IPs) in Patna, India. Participants received training from an infectious disease physician on the WHO AWaRe Antibiotic Handbook. …

BACKGROUND: Individuals who have been incarcerated are at high risk of death by homicide after release from carceral settings. Our study aimed to assess the prevalence of criminal legal system involvement (CLSI) among non-fatal violently injured patients enrolled in a hospital-based violence intervention (HVIP) program and describe demographic and clinical characteristics specific to this population.
METHODS: This is an exploratory retrospective cohort study of all patients enrolled in the St. Louis, Missouri based HVIP, Life Outside of Violence (LOV), between 08/15/2018 and 12/31/2024. LOV enrolls patients aged 8-30 who receive violent injury care at a partner hospital. We collected data …

BACKGROUND: Antimicrobial resistance (AMR) is a growing public health threat, and we currently lack accurate measures to track and trend this resistance. We developed the antibiotic resistance index (ARI) that aggregates resistance of Gram-negative bacilli (GNB) into a single metric which can be tracked across healthcare settings and over time.
METHODS: Culture data were collected from adult patients who met the CDC adult sepsis event criteria across 10 Barnes-Jewish HealthCare (BJC) hospitals between January 2018 and December 2023.
