In this brief, authors Jess Carson and Sarah Boege describe child care use and gaps among respondents to the 2022 New Hampshire Preschool Development Grant Family Needs Assessment Survey. The authors report that three-quarters of responding parents with children under age five had searched for child care in the past 12 months; half reported that their search was difficult, with a lack of openings as the main challenge. Despite these difficulties, three-quarters of respondents with young children reported using some form of regular child care. Supportive Program Strengths And Gaps For New Hampshire Families: "Just Enough Money To Barely Pay For Most Things", Sarah Boege, Jess Carson

In this brief, authors Sarah Boege and Jess Carson describe child and family program use and gaps among respondents to the 2022 New Hampshire Preschool Development Grant Family Needs Assessment Survey. Parents with children under age nine were asked about programs to help meet family food needs and to support child development and education. The authors find that nearly half of parent respondents used at least one of the programs in question, with the National School Lunch Program having the widest reach. Responding parents were generally familiar with food assistance programs, but less familiar with child development and education programs. …

Mutations in Nucleophosmin 1 (NPM1) are associated with a favorable prognosis in newly diagnosed acute myeloid leukemia (AML), however, their prognostic impact in relapsed/refractory (R/R) settings are unknown. In a retrospective analysis, we identified 206 patients (12%) with mutated NPM1 (NPM1c) and compared their outcomes to 1516 patients (88%) with NPM1 wild-type (NPM1wt). NPM1c was associated with higher rates of complete remission or complete remission with incomplete count recovery compared with NPM1wt following each line of salvage therapy (first salvage, 56% vs 37%; P < .0001; second salvage, 33% vs 22%; P = .02; third salvage, 24% vs 14%; P = .02). However, NPM1 mutations had no impact on relapse-free survival (RFS) and overall survival (OS) with each salvage therapy with a median OS following salvage 1, 2 or 3 therapies in NPM1c vs NPM1wt of 7.8 vs 6.0; 5.3 vs 4.1; and 3.5 vs 3.6 months, respectively. Notably, the addition of venetoclax to salvage regimens in patients with NPM1c improved RFS and OS (median RFS, 15.8 vs 4.6 months; P = .05; median OS, 14.7 vs 5.9 months; P = .02). In conclusion, NPM1 mutational status has a minimal impact on prognosis in relapsed or refractory AML; therefore, novel treatment strategies are required to improve outcomes in this entity.

Purpose Despite the number of quality management procedures being currently applied, construction defects in the domestic sector are acknowledged to contribute to the energy performance gap of buildings. This paper investigates the limitations and challenges to the implementation of project quality plans (PQPs) and their impact on the achievement of expected thermal performance in the UK social housing projects. Design/methodology/approach A qualitative approach, guided by grounded theory, was used in this research. Aim: Despite well-established protocols for cardiopulmonary resuscitation training, performance during real-life cardiac arrests can be suboptimal. Understanding personal characteristics which could influence performance of high-quality chest compressions could provide insight into the practice-performance gap. This study examined chest compression performance, while employing feedback and introducing code team sounds as an anxiety-inducing factor in registered nurses using a cardiopulmonary resuscitation training manikin.

Methods: Participants included 120 registered nurses with basic life support certification randomized to one of the following groups: no feedback and no code team sounds, feedback without code team sounds, or feedback with code team sounds. Chest compression sessions …

The United States of America, as the richest nation in the world, must determine how to allocate more resources toward health promotion, prevention, and the public health of its citizens. This exploratory study investigates the phenomenon of the advanced practice nurse as healthcare clinician for improved implementation of clinical and community preventive services. The following dissertation explains the psychologism debate as it played out in the 19thand early 20th Centuries and then shows how Martin Heidegger radicalized the debate by undermining its key themes and assumptions. First, I explain each side of the psychologism debate, starting with the psychologicists. I explore the philosophies of Jakob Friedrich Fries and John Stuart Mill in order to encapsulate the full spectrum of psychologism in the 19th Century, from Neo-Kantian to British Empiricist. How do communities living with chronic environmental contamination cope with the social, political, and economic impacts of the contamination? This research employs a community-engaged oral history approach with participant observation and archival research to address this question. We focus on the case of Tallevast, Florida, where the local groundwater has been contaminated with chlorinated solvents for over 60 years and where cleanup is estimated to take another 100 years. DNA replication needs to be strictly monitored to ensure proper duplication of the genome. During DNA synthesis, the DNA replication machinery encounters multiple obstacles such as incorrect dNTP incorporation, RNA-DNA hybrids, modification of DNA nucleobases, collision with the transcription-replication machinery. These endogenous and exogenous sources of DNA damage may result in single-strand breaks (SSBs) and double-strand DNA breaks (DSBs), thereby impeding fork progression. To prevent stalling of DNA synthesis a multitude of DNA repair pathways are specifically designed to deal with these kinds of blockages. In this paper, we present a unifying analytical framework for identifying conditions for transport effects such as reflectionless and transparent transport, lasing, and coherent perfect absorption in non-Hermitian nonreciprocal systems using a generalized transfer matrix method. This provides a universal approach to studying the transport of tight-binding platforms, including higher-dimensional models and those with an internal degree of freedom going beyond the previously studied case of one-dimensional chains with nearest-neighbor couplings. For a specific class of tight-binding models, the relevant transport conditions and their signatures of non-Hermitian, nonreciprocal, and topological behavior are analytically tractable from a general perspective. 'I Can’T Vote If I Don’T Leave My Apartment’: The Problem Of Residential Violence And Its Impact On The Politics Of Black American Women Living Below The Poverty Line, Alex J. Moffett-Bateau

Prior research examining political behavior outside of the United States, has shown that violence can have a mixed impact on political engagement. Building on that work, this research examines whether violence shapes the political lives of poor Black women within the United States. I argue, neighborhood violence in the United States can and often does, shape the political behavior of Black women living below the poverty line in public housing. I use ethnographic data to parse out a conceptual framework which articulates connections between residential violence experienced by Black women living in poverty and their politics. Enterocytes modulate the extent of postprandial lipemia by storing dietary fats in cytoplasmic lipid droplets (cLDs). We have previously shown that the integrin ligand MFGE8 links absorption of dietary fats with activation of triglyceride (TG) hydrolases that catabolize cLDs for chylomicron production. Here, we identify CES1D as the key hydrolase downstream of the MFGE8-αvβ5 integrin pathway that regulates catabolism of diet-derived cLDs. Mfge8 knockout (KO) enterocytes have reduced CES1D transcript and protein levels and reduced protein levels of the transcription factor HNF4γ. Both Ces1d and Hnf4γ KO mice have decreased enterocyte TG hydrolase activity coupled with retention of TG in …

BACKGROUND: Both Alzheimer's disease (AD) genetic risk factors and indices of cognitive reserve (CR) influence risk of cognitive decline, but it remains unclear whether they interact. This study examined whether a CR index score modifies the relationship between AD genetic risk factors and long-term cognitive trajectories in a large sample of individuals with normal cognition.

METHODS: Analyses used data from the Preclinical AD Consortium, including harmonized data from 5 longitudinal cohort studies. Participants were cognitively normal at baseline (M baseline age = 64 years, 59% female) and underwent 10 years of follow-up, on average. AD genetic risk was measured by …

Elastic laminae, an elastin-based, layered extracellular matrix structure in the media of arteries, can inhibit leukocyte adhesion and vascular smooth muscle cell proliferation and migration, exhibiting anti-inflammatory and anti-thrombogenic properties. These properties prevent inflammatory and thrombogenic activities in the arterial media, constituting a mechanism for the maintenance of the structural integrity of the arterial wall in vascular disorders. The biological basis for these properties is the elastin-induced activation of inhibitory signaling pathways, involving the inhibitory cell receptor signal regulatory protein α (SIRPα) and Src homology 2 domain-containing protein tyrosine phosphatase 1 (SHP1). BACKGROUND: Understanding how heterogeneous β-cell function impacts diabetes is imperative for therapy development. Standard single-cell RNA sequencing analysis illuminates some factors driving heterogeneity, but new strategies are required to enhance information capture.

RESULTS: We integrate pancreatic islet single-cell and bulk RNA sequencing data to identify β-cell subpopulations based on gene expression and characterize genetic networks associated with β-cell function in obese SM/J mice. We identify β-cell subpopulations associated with basal insulin secretion, hypoxia response, cell polarity, and stress response. Network analysis associates fatty acid metabolism and basal insulin secretion with hyperglycemic-obesity, while expression of Pdyn and hypoxia response is associated …

Eye tracking has proven to be a valuable tool for a number of disciplines such as imaging science, vision science, psychology, and neurology. They have used eye-tracking to study the human visual system and to understand how humans attend to their environment for different tasks. Analyzing the gaze data obtained from eye-trackers begins with visualizing the gaze point. By monitoring gaze we can answer questions including, ‘where was the person looking?’ and ‘what image region or object was the person looking at?’ Traditional methods aimed at answering such questions from gaze data: required frame-by-frame analysis of videos, manually annotating gaze. …
