The serine palmitoyltransferase (SPT) complex catalyzes the first and rate-limiting step in sphingolipid biosynthesis in all eukaryotes. ORM/ORMDL proteins are negative regulators of SPT that respond to cellular sphingolipid levels. However, the molecular basis underlying ORM/ORMDL-dependent homeostatic regulation of SPT is not well understood.We determined the cryo–electron microscopy structure of Arabidopsis SPT-ORM1 complex, composed of LCB1, LCB2a, SPTssa, and ORM1, in an inhibited state. A ceramide molecule is sandwiched between ORM1 and LCB2a in the cytosolic membrane leaflet. Ceramide binding is critical for the ORM1-dependent SPT repression, and dihydroceramides and phytoceramides differentially affect this repression. A hybrid β sheet, formed …

A Reparatory Model Of Ethical Silence, Yuanmei Qu, Mayowa Babalola, Mozzam Ali, Jennifer Harrison, Muhammad Usman

Drawing from the appraisal theory of emotion and self-conscious emotions literature, this study proposes a reparatory model of ethical silence at work. We posit that when employees maintain silence on ethical issues, they experience feelings of guilt. This guilt, in turn, propels them to engage in ethical performance as reparatory behaviors. Results from a multisource, three-wave field study supported these hypotheses. Overall, this research contributes to silence literature and provides insights into how and when ethical silence may, paradoxically, facilitate more ethical performance later on.

Sound decision-making relies on accurate prediction for tangible outcomes ranging from military conflict to disease outbreaks. To improve crowdsourced forecasting accuracy, we developed SAGE, a hybrid forecasting system that combines human and machine generated forecasts. The system provides a platform where users can interact with machine models and thus anchor their judgments on an objective benchmark. The system also aggregates human and machine forecasts weighting both for propinquity and based on assessed skill while adjusting for overconfidence. We present results from the Hybrid Forecasting Competition (HFC)—larger than comparable forecasting tournaments—including 1085 users forecasting 398 real-world forecasting problems over 8 months. …

Stabilizing G-Quadruplex Dna And Rna Structures With Ionic Liquids, Timothy Vaden

Stabilizing DNA and RNA for long-term room-temperature storage is important for many biotechnological applications including oncology pharmaceuticals and mRNA-based vaccines (e.g. Covid vaccines). This poster shows that ionic liquids can improve the thermal stability of G-quadruplex DNA and RNA structures, and motivates further studies of ionic liquid-based materials for DNA / RNA stabilization.

The Impact Of Family Support And Rejection On Suicide Ideation And Attempt Among Transgender Adults In The U.S., Guadalupe Marquez-Velarde, Gabe H. Miller, Jesse Ezra Shircliff, Mario Itzel Suárez

We evaluate the association of familial factors and suicidality among transgender adults in the U.S. by estimating the odds of lifetime suicide ideation and attempt using the 2015 U.S. Transgender Survey. Predictors include family support, family rejection, and specific experiences related to both. About 79% of sample respondents have experienced suicidal ideation and nearly 43% have made a suicide attempt. The predicted probability of suicide attempt is 0.35 for those with no family rejection experiences, 0.75 for those who have had all five experiences in our models. Rejection predicts both outcomes and experiences of rejection have a cumulative impact.

Gender-affirming surgery (GAS) has been proven to be successful in the treatment of gender dysphoria. The benefits of providing insurance coverage for transition-related surgeries far surpass the costs of suffering from persistent gender dysphoria, including many positive health outcomes such as decreased rates of substance use, psychiatric illness, and suicide. Despite being deemed a medical necessity, discrepancies in access to treatment and insurance coverage for GAS persist. The purpose of this review is to understand the impact of limited insurance coverage on the well-being of transgender patients. The primary goal of this scoping review was to identify studies where an intervention of education and awareness of surgical supply costs with surgeons was employed as a possible stimulus for healthcare organization cost reduction in the surgical/procedural service space.
Surgical procedures are performed on approximately 25 percent of all inpatient hospital admissions. In 2018 that equated to 14.4 million surgical procedures and $210.3 billion dollars in health system costs. Surgical Procedures are a substantial portion of healthcare system expenditure and are one of the highest revenue producing services provided to patients. Though black boards and white boards have been a fundamental tool in the classroom for over a century, little research has been done on how to best design and present information using these boards. My study takes visual design principles and applies them to boardwork in a mathematics classroom to better organize and clarify the content. This research shows that students notice boardwork, have strong opinions on what makes boardwork good, and that the application of design principles on boards has a significant impact on students and the teacher. Bright Star is a musical written and composed by Steve Martin and Edie Brickell. It is set in the Blue Ridge Mountains of North Carolina in 1945–46 with flashbacks to 1923. The musical is inspired by their Grammy-winning collaboration on the 2013 bluegrass album Love Has Come for You and, in turn, the folk story of the Iron Mountain Baby.
