This thesis looks at Buffalo’s legacy breweries from 1870 to 1972.  Buffalo had a dynamic brewing past with over 30 breweries in existence on the eve of Prohibition. Buffalo’s breweries always displayed remarkable entrepreneurial ingenuity, which allowed them to adapt, change, and shift their businesses to survive. The brewers faced setbacks well before Prohibition ranging from fires to overcompetition to licensing challenges to outside brewers poaching their local markets. In covering Prohibition challenges, this thesis will only look at local brewers’ attempts to survive inside the law.  Plenty of work has already been done on illegal forms of coping.  In …

Water containing high levels of dissolved minerals, mainly calcium and magnesium, is known as hard water, which is commonly found across the world due to calcium’s abundance in the earth’s crust. Hard water can lead to calcium carbonate buildup (also referred to as limescale) that reduces flow, clogs pipes, and lowers heat exchanger efficiency, all of which can drive up costs. This work evaluated two commercially available chemical-free treatment devices from HydroFLOW USA LLC, called the Pearl Plus and s38 devices. Bacteriophages (phages) are viruses that infect bacteria and play a role in bacterial evolution. Through horizontal gene transfer (HGT), phages integrate parts of their genomes into their bacterial hosts. While the molecular mechanisms of this process are well understood, the evolutionary impacts remain less certain. One evolutionary explanation is that auxiliary metabolic genes (AMGs) modulate host gene expression to favor phage reproduction or improve the host’s survival. While libraries provide valuable academic support and resources, many students do not consistently use these services. Drawing on existing literature, this research explores how library design, service awareness, and emotional factors shape help-seeking behavior. Prior studies suggest that unclear layouts, low visibility of help desks, and limited understanding of librarian roles can discourage engagement. According to the 2024 National College Health Assessment, roughly one in five college students experience serious psychological distress (ACHA, 2024). High levels of psychological distress among college students are well-documented (Emmerton et al., 2024; Eisenberg et al., 2013; Bewick et al., 2010), yet less is known about the specific experiences that contribute to this distress. Grounded in multiple theoretical frameworks, this study examines students’ negative academic experiences, their perceived causes, and associated outcomes. Lipid nanoparticles (LNPs) have become effective delivery vehicles in nanomedicine, particularly for nucleic acid-based treatments. However, a major limitation remains in the field regarding the simultaneous monitoring of the nanoparticle carrier, its encapsulated payload, and the resulting functional protein production in both in vitro and in vivo systems. Furthermore, traditional lipophilic dyes employed for labeling LNPs frequently exhibit incompatibility with tissue-clearing methods, such as CLARITY, due to signal loss following lipid removal. We Are Still Lipan: Identity Erasure, Settler Colonialism, Historical Memory And The Persistence Of The Non-Reservation Lipan Apache, Freddy Cabral

This dissertation challenges the long-standing historical narrative that the Lipan Apache disappeared in the late nineteenth century. Instead, it argues that the non-reservation Lipan Apache survived through strategic adaptation, mobility, kinship networks, and cultural continuity across the United States-Mexico borderlands. By examining the Lipan experience under Spanish colonial rule, Mexican governance, the Republic of Texas, and the State of Texas, this study demonstrates that shifting political regimes consistently pursued Indigenous land, labor, and subordination through structures of settler colonialism and racial hierarchy. Using the frameworks of settler colonialism, white supremacy, genocide studies, oral history, and autoethnography, this dissertation shows how …

This thesis, Blood, Stone, and Steel, explores the intergenerational impact of trauma, memory, and history through a unique blend of poetry, photography, and personal family narratives. Drawing on archival materials, family photographs, and the tradition of documentary poetry, the work investigates how the legacy of war and immigration shapes identity across generations. Inspired by Marianne Hirsch's concept of postmemory, the author examines inherited memories and their affective transmission, focusing on the experiences of her great-grandfather - a German immigrant and WWI veteran - and the subsequent effects on her family. Modern extracellular neural probes like Neuropixels, allow for high-density extracellular neural probes to record terabytes of electrophysiological brain data. Spike sorting is the process of isolating individual neurons on this data through various clustering and filtering techniques. Modern spike sorting algorithms are held back by their need for human intervention to separate high-quality from low quality clusters, due to the volume of data that can be collected a completely automated approach is required. This phenomenological qualitative study explored the use of the Dungeons and Dragons (D&D) Role-Playing Game as a Game-Based Learning (GBL) model, measuring middle school student perceptions of engagement, motivation, and skill development through interactive storytelling. The study used a Practitioner Research theoretical framework and Thematic Analysis to identify emergent themes from the collected data. The study included gameplay of D&D after school with a small group of participants grades 6-8. Results indicated that participants reported consistently high levels of motivation and engagement. This scoping review examines post-retraction citations as a proxy for epistemic risk (i.e., the likelihood that one holds a false or inaccurate belief). Cranes (Gruidae) are a globally distributed clade of large, wetland-associated birds. Prior phylogenetic analyses have shown there to be two major subfamilies, Balearicinae and Gruinae, and five consistently recovered clades within Gruinae. However, the relationships among those five clades has remained contentious. In addition, little has been done to reconstruct the biogeographic history of cranes. To that end, I first inferred phylogeny using a large, nuclear dataset sampled from all 15 extant crane species and two outgroup species. The phylogeny mostly agreed with previous analysis using whole mitochondrial genomes but differed in that the Anthropoides species group formed a sister …

The presence of delays has a significant influence on system performance both positively and negatively. This project approaches a long-term experience of living in a cult through a performance studies lens. Utilizing autoethnographic methods, it explores personal insights and observations from years within the inner-circle of a coercive religious environment under tyrannical leadership. A variety of systems and relationships are narrated and analyzed according to three categories: ritualization, deployability, and occupation. Ritualization explains how a body becomes imbued with the rhythms and mantras of a cultic milieu. Deployability describes how this body is put into action at the service of the leadership. Rooted in the understanding that effective leadership significantly influences organizational outcomes, this study examines process-oriented relationships between leader initiating structure and subordinate outcomes. Specifically, it extends the leadership literature by focusing on the cognitive resources underlying initiating structure behaviors, such as role-related clarity, that may enhance positive subordinate outcomes and mitigate negative ones. Proactive public-safety surveillance requires deep learning systems that can detect and segment weapons reliably under real-world conditions. This research develops and evaluates a progressive, multi-phase pipeline that moves (i) from RGB object detection to pixel-level semantic segmentation and (ii) from visible-spectrum cameras to thermal imagery to improve robustness under low-light, clutter, and occlusion. Early phases establish strong detection baselines using one-stage and two-stage detectors (YOLOv5 and Faster R-CNN) for multi-class weapon localization, followed by practical optimization through pruning and ensembling to reduce false positives and computational overhead while maintaining real-time feasibility. The principled combination of symbolic execution and random testing lacks a formal foundation, especially in deciding which inputs to symbolize. We propose selective concolic testing, a cost-aware framework that formulates this choice as an optimized policy problem of a MDP (Markov Decision Process). We model program exploration over a finite control-flow graph, where MDP states represent covered statements, actions partition path constraints into symbolic and random fragments, rewards reflect coverage gain, and costs account for SMT solving effort and sampling inefficiency. NRAS mutations occur in 10%-30% of cutaneous melanomas and are associated with high tumor mutational burden. Mutant NRAS signaling drives aberrant cell growth and proliferation, in part, through activation of the RAF-MEK-ERK1/2 kinase pathway; however, targeted therapies to this pathway have limited effectiveness in patients with NRAS mutant melanoma. The role of other targetable signaling pathways in NRAS mutant melanoma is poorly characterized. Here, we demonstrated that one isoform of diacylglycerol kinase, diacylglycerol kinase eta (DGKη), a lipid signaling regulator, was highly expressed in NRAS mutant melanoma patient samples.
