Vision impairments have a high prevalence in adults with a stroke or traumatic brain injury (TBI), potentially affecting up to 90% of this population. Biochemical Characterization Of Fsa1572 From Fervidibacter Sacchari, The First Hyperthermophilic Gh50 With Β-1, 4-Glucanase Activity, Jonathan Covington

The bacterium Fervidibacter sacchari is an aerobic hyperthermophile that catabolizes various polysaccharides and is the only cultivated member of the class Fervidibacteria within the phylum Armatimonadota. Among its glycoside hydrolase (GH) cache is an enzyme from GH family 50 (GH50), an understudied family with only 25 characterized representatives and two known activities from 1,518 predicted members in the Carbohydrate-Active EnZyme (CAZy) Database. Here, we expressed, purified, and functionally characterized an extracellular GH50 from F. sacchari called Fsa1572. Using colorimetric assays, we show it has novel β-1,4-glucanase activity and only weak agarose activity that is typical for GH50 enzymes. The purified …

Long-Range Regulation By The Shigella Virulence Gene Regulator, Virb, Cody Cris

Nucleoid-structuring proteins (NSPs) in bacterial cells can exert gene regulatory effects over great distances on DNA. This contrasts with the traditional view of bacterial transcriptional regulation, where transcription factors bind cis-acting regulatory sites proximal (<250 bp) to promoters to influence gene expression. In Shigella species, the histone-like nucleoid-structuring protein (H-NS) transcriptionally silences many genes on the large (~220 kb) virulence plasmid. During host colonization, however, silencing by H-NS is alleviated by the anti-silencer protein, VirB, which remodels the H-NS:DNA complex. This transcriptional anti-silencing is mediated by VirB:DNA interactions and the modulation of DNA supercoiling, which lead to the up-regulation of virulence genes. VirB is a central regulator of Shigella virulence, but the mechanistic basis for transcriptional regulation is incompletely characterized. Here, I focus on the potential for VirB to regulate transcription from remotely located DNA binding sites. Employing complementary in vivo approaches, I assessed the spatial stringency of the VirB binding site and elucidated how varying cellular VirB levels impacts regulation over different distances. I also evaluated how a molecular roadblock on DNA influences remote regulation by VirB. Additionally, I developed control tools to validate the ability of VirB to spread along DNA, an activity of VirB that putatively drives transcriptional regulation. These data are consistent with transcriptional anti-silencing being driven by VirB spreading along DNA. Overall, these findings highlight how VirB is not a conventional transcriptional regulator and broaden our perspective on transcriptional control in bacteria. Furthermore, this work contributes to our understanding of long-range gene regulatory effects, which are frequently overlooked and poorly characterized in bacteria.

This manuscript describes a quality improvement project completed by a student in the University of Nevada Las Vegas Occupational Therapy Doctorate Program. The project tests the efficacy of an educational toolkit for occupational therapists staffed at an acute care hospital in Las Vegas, Nevada, to increase their perceived knowledge of and ability to educate families on conservative management techniques for pediatric reflux management. Results indicate that the toolkit demonstrated efficacy in achieving this objective. Fatigue is an exercise-induced reduction in muscle force generating capacity. Several studies have shown that some forms of non-invasive brain stimulation can increase the resistance to fatigue during sustained isometric contractions. The purpose was to determine the influence of transcranial alternating current stimulation (tACS) on the time to task failure (TTF) of a precision grip task in young adults. Twenty-two healthy young men and women participated in the study. The study utilized a randomized, double-blind, SHAM-controlled, within-subjects, crossover design. Terrorist groups from the Middle East and North Africa (MENA) region remain one of the most formidable threats to international security. While their organization and motives have been significantly different over time, a commonality is that they tend to disproportionately target Western democracies. This dissertation argues that this trend is mainly a consequence of the foreign policies that these states pursue in the region. Purpose: The aim of this project is to determine if the development and implementation of an independent living skills workshop would improve the perceived competence of unhoused youth in performing independent living skills.Methods: The Casey Life Skills Short Assessment was used as a pre-test and post-test. It was found that unhoused young adults generally scored low on finding affordable housing, budgeting, banking skills, education training, and feeling a sense of pride in their lives. Teachers with low levels of self-efficacy are correlated with lower job satisfaction, higher levels of stress and anxiety, and lower student achievement gains in the classroom. Such levels of volatility have highlighted a need to understand the effects of the COVID-19 pandemic on flight crew communication. This study explores the impact of the COVID-19 pandemic on intra-flight crew communication and the emotional labor flight crew members experienced. In this dissertation, silicon-based materials for photovoltaics and chalcopyrite-based materials for photoelectrochemical water splitting are investigated using various spectroscopic and microscopic techniques. Although silicon dominates the photovoltaic market, further improvement can be made by using an alternative low temperature passivation approach. Currently, thermally grown SiO2 passivation is commonly used for silicon solar cells. However, this technique requires high processing temperatures (>800 °C), which increases the thermal budget, potentially decreases the bulk quality of Si, and can lead to difficulties in implementing in production lines. Here, a S-based passivation approach is studied that require lower processing temperatures of ~550 °C. …

Static compression experiments on water ice are needed for precise characterization and discovery of new ice phases near room temperature. Here I present my efforts and the developments that have been made to reduce detrimental effects found in high-pressure experiments in order to acquire precise measurements on ice approaching 3 Mbar. The objective of this study was to develop a predictive model that utilizes a Light Gradient Boosting Machine (Light-GBM) to accurately assess the risk of severe radiation-induced toxicities in cancer patients undergoing chemoradiotherapy. The Savanah River Basin is an area of 9,850 square miles that stretches from the border between South Carolina and Georgia, which empties into the Atlantic Ocean near Savannah, Ga. This area is home to surrounding cities, industrial sites, and the Savanah River Site. In this dissertation, the methods of fundamental solutions (MFS) and the methods of particular solutions (MPS) are used to solve the boundary value problems of the Poisson and Helmholtz equations, where the particular solutions of the Poisson and Helmholtz equations in [13, 14] are used. Then the initial boundary value problems of the diffusion and wave equations are discretized into a sequence of boundary value problems of the Helmholtz equation by using either the Laplace transform or time difference methods along the lines of [8]. I analyze new HIRES Radial Velocity (RV) data in conjunction with transit mid-times and uncertainties from the full 17 quarters of Kepler data to reassess the orbital parameters of the Kepler-36 system. Six additional RV measurements were taken by the Keck-HIRES spectrograph from September 21, 2021 to October 11, 2021. I carry out a differential evolution Markov Chain Monte Carlo-based (DEMCMC) analysis to infer improved orbital elements for the two known planets in the system. Leveraging additional information provided by the new RV measurements, I extend this DEMCMC analysis to a possible three-planet configuration. I explore the likelihood of a …
