Pharmacy and Wellness Review
Alcohol use disorder (AUD) is a medical condition that arises from the inability to control alcohol consumption regardless of negative consequences. Over 10% of the US population met the diagnostic criteria for having AUD in 2022.1 Each individual presents with symptoms of AUD differently and may therefore need more personalized treatment. Research has shown that using a combination of currently available pharmacological and non-pharmacological treatment options provides the best results for patients with AUD. However, there still remains a pressing need for the development of newer treatment options. Psilocybin, a naturally occurring psychedelic compound, is currently under investigation due to …

Lived Experience Of Burnout And Self-Efficacy In Intensive Care Unit Registered Nurses Caring For Covid-19 Patients In U.S. Hospitals, Trixie Alohilani Harris
Walden Dissertations and Doctoral Studies
The COVID-19 pandemic has impacted the physical and mental health of intensive care unit registered nurses (ICU RNs). Burnout in ICU RNs existed long before the pandemic; however, the massive increase in critically ill patients and the scarcity of supplies and qualified staff to care for these patients put RNs in a dangerous situation. Research addressed this topic early in the pandemic but were primarily quantitative, with minimal qualitative studies appearing later. There remained a gap in examining ICU RNs in rural hospitals in the United States. The research question for this hermeneutic phenomenological study explored the interpretation of the …

PURPOSE: Develop and deploy a robust discovery platform that encompasses heterogeneity, clinical annotation, and molecular characterization and overcomes the limited availability of prostate cancer models. This initiative builds on the rich MD Anderson (MDA) prostate cancer (PCa) patient-derived xenograft (PDX) resource to complement existing publicly available databases by addressing gaps in clinically annotated models reflecting the heterogeneity of potentially lethal and lethal prostate cancer.
EXPERIMENTAL DESIGN: We performed whole-genome, targeted, and RNA sequencing in representative samples of the same tumor from 44 PDXs derived from 38 patients linked to donor tumor metadata and corresponding organoids. The cohort includes models derived …

A challenge with studying cancer transcriptomes is in distilling the wealth of information down into manageable portions of information. In this resource, we develop an approach that creates and assembles cancer type-specific gene expression modules into flexible barcodes, allowing for adaptation to a wide variety of uses. Specifically, we propose that modules derived organically from high-quality gold standards such as The Cancer Genome Atlas (TCGA) can accurately capture and describe functionally related genes that are relevant to specific cancer types. We show that such modules can: (1) uncover novel gene relationships and nominate new functional memberships, (2) improve and speed …

Longitudinal monitoring of patients with advanced cancers is crucial to evaluate both disease burden and treatment response. Current liquid biopsy approaches mostly rely on the detection of DNA-based biomarkers. However, plasma RNA analysis can unleash tremendous opportunities for tumor state interrogation and molecular subtyping. Through the application of deep learning algorithms to the deconvolved transcriptomes of RNA within plasma extracellular vesicles (evRNA), we successfully predicted consensus molecular subtypes in patients with metastatic colorectal cancer. Analysis of plasma evRNA also enabled monitoring of changes in transcriptomic subtype under treatment selection pressure and identification of molecular pathways associated with recurrence. This approach …

Extracellular vesicles mediate intercellular communication by transporting biologically active macromolecules. Our prior studies have demonstrated that the nuclear factor of activated T cell cytoplasmic member 3 (NFATc3) is activated in mouse pulmonary macrophages in response to lipopolysaccharide (LPS). Inhibition of NFATc3 activation by a novel cell-permeable calcineurin peptide inhibitor CNI103 mitigated the development of acute lung injury (ALI) in LPS-treated mice. Although pro-inflammatory lipid mediators are known contributors to lung inflammation and injury, it remains unclear whether the calcineurin-NFATc pathway regulates extracellular vesicle (EV) lipid content and if this content contributes to ALI pathogenesis. In this study, EVs from mouse …

Cellular sensitivity to ferroptosis is primarily regulated by mechanisms mediating lipid hydroperoxide detoxification. We show that inositol-requiring enzyme 1 (IRE1α), an endoplasmic reticulum (ER) resident protein critical for the unfolded protein response (UPR), also determines cellular sensitivity to ferroptosis. Cancer and normal cells depleted of IRE1α gain resistance to ferroptosis, while enhanced IRE1α expression promotes sensitivity to ferroptosis. Mechanistically, IRE1α's endoribonuclease activity cleaves and down-regulates the mRNA of key glutathione biosynthesis regulators glutamate-cysteine ligase catalytic subunit (GCLC) and solute carrier family 7 member 11 (SLC7A11). This activity of IRE1α is independent of its role in regulating the UPR and is …

BACKGROUND: Respiratory syncytial virus (RSV) is a leading cause of acute respiratory illnesses in children. RSV can be broadly categorized into 2 major subtypes: A and B. RSV subtypes have been known to cocirculate with variability in different regions of the world. Clinical associations with viral subtype have been studied among children with conflicting findings such that no conclusive relationships between RSV subtype and severity have been established.
METHODS: During 2016-2020, children aged/guardian interviews, chart reviews, and collection of midturbinate nasal plus/minus throat swabs for RSV (RSV-A, RSV-B, and untyped) using reverse transcription polymerase chain reaction.
RESULTS: Among 6398 RSV-positive …

The Wnt receptor ROR1 has generated increased interest as a cancer therapeutic target. Research on several therapeutic approaches involving this receptor is ongoing; however, ROR1 tissue expression remains understudied. We performed an immunohistochemistry analysis of ROR1 protein expression in a large cohort of multiple tumor and histologic types. We analyzed 12 anonymized multi-tumor tissue microarrays (TMAs), including mesothelioma, esophageal and upper gastrointestinal carcinomas, and uterine endometrioid carcinoma, among other tumor types. Additionally, we studied 5 different sarcoma types of TMAs and 6 patient-derived xenografts (PDX) TMAs developed from 19 different anatomic sites and tumor histologic types. A total of 1142 …

We test the predictions of hadronic interaction models regarding the depth of maximum of air-shower profiles, Xmax, and ground-particle signals in water-Cherenkov detectors at 1000 m from the shower core, S(1000), using the data from the fluorescence and surface detectors of the Pierre Auger Observatory. The test consists of fitting the measured two-dimensional (S(1000), Xmax) distributions using templates for simulated air showers produced with hadronic interaction models epos-lhc, qgsjet-ii-04, sibyll 2.3d and leaving the scales of predicted Xmax and the signals from hadronic component at ground as free-fit parameters. The method relies on the assumption that the mass composition remains …

“How does one discover solace and belonging within these layered narratives?”
In mythology, narratives were once created to answer the incomprehensible questions of an era. These narratives unveil half-truths, customs, and convictions. K(now) W(here) is based on the experience of the artist who is Chinese and immigrated to South Africa at a young age; she elaborates the story about assimilation, authenticity, tales of her ancestral roots, and, most often, myths of identity.
The artist used narratives from Chinese mythology, collaged physically and metaphorically using tangible objects from other’s homes in combination with photography, digital media, and domestic items and assemblage …

Abstract: Aiming at large fluctuations of intermission and low prediction accuracy in iron and steel industry, based on the classification of intermission characteristics, a converter gas generation predicting model(CPSO-Elman) based on Elman neural network(ENN) optimized by chaotic PSO(CPSO) algorithm is proposed. The intermittent characteristics of converter gas generation time series are extracted and raw data is classified according to intermittent duration. The PSO algorithm improved by chaotic disturbance is introduced to optimize the initial weight and threshold of ENN and inertia weight of nonlinear updating is designed to balance global search ability and local search ability. Construct the combined prediction …

Dose-Dependent Effects Of Gamma Radiation Sterilization On The Collagen Matrix Of Human Cortical Bone Allograft And Its Influence On Fatigue Crack Propagation Resistance, Dylan B. Crocker, Thomas M. Hering, Ozan Akkus, Clare M. Rimnac
Student Scholarship
Fatigue crack propagation resistance and high-cycle S–N fatigue life of cortical bone allograft tissue are both negatively impacted in a radiation dose-dependent manner from 0 to 25 kGy. The standard radiation sterilization dose of 25–35 kGy has been shown to induce cleavage of collagen molecules into smaller peptides and accumulation of stable crosslinks within the collagen matrix, suggesting that these mechanisms may influence radiation-induced losses in cyclic fracture resistance. The objective of this study was to determine the radiation dose-dependency of collagen chain fragmentation and crosslink accumulation within the dose range of 0–25 kGy. Previously, cortical bone compact tension specimens …

H2 is increasingly regarded as a decarbonized solution for the increasing energy demand. However, due to its low density, H2 cannot be easily compressed, and there is a need for materials that can store large amounts of H2. Here we tested H2 adsorption capacity of micro-polyethylene (MicroPE) – a significant environmental pollutant - at 1–70 bar and 30 °C. H2 adsorption capacity increased with increasing the adsorption pressure (from 0.06 mol/kg at 17.21 bar to 0.20 mol/kg at 68.11 bar for raw MicroPE). MicroPE activated with aqueous NaOH significantly changed H2 adsorption under the same operating conditions. H2 adsorption increased …

Research outputs 2022 to 2026
Quantifying the interlinked behaviour of the soil microbiome, fluid flow, multi-component transport and partitioning, and biodegradation is key to characterising vapour risks and natural source zone depletion (NSZD) of light non-aqueous phase liquid (LNAPL) petroleum hydrocarbons. Critical to vapour transport and NSZD is transport of gases through the vadose zone (oxygen from the atmosphere, volatile organic compounds (VOCs), methane and carbon dioxide from the zone of LNAPL biodegradation). Volatilisation of VOCs from LNAPL, aerobic biodegradation, methanogenesis and heat production all generate gas pressure changes that may lead to enhanced gas fluxes apart from diffusion. Despite the importance of the gaseous …

It is imperative for individuals to exhibit flexible behaviour according to ecological context, such as available resources or predation threat. Manipulative studies on responses to threat often focus on behaviour in the presence of a single indicator for the potential of predation, whereas in the wild perception of threat will probably be more nuanced. Here, we examine the collective behaviour of eastern mosquitofish (Gambusia holbrooki) subject to five differing threat scenarios relating to the presence and hunger state of a jade perch (Scortum barcoo). Across threat scenarios, groups exhibit unique behavioural profiles that differ in the durations that particular collective …

Trace metals (TMs) manganese (Mn), cobalt (Co), and aluminium (Al) have important geochemical and biological roles in the ocean. Here, we present full depth profiles of dissolved (d) and particulate Al, Mn, and Co along the latitude of 40 °S in the South Atlantic Ocean from the GEOTRACES GA10 cruises that operated in austral spring 2010 and summer 2011. The region is characterized by enhanced primary productivity and forms a key transition zone between the Southern Ocean and South Atlantic Subtropical Gyre. The mean concentrations of dAl, dCo, and dMn (±standard deviation) were 3.36 ± 2.65 nmol kg−1, …

Quantum computing has the potential to solve problems that are currently intractable to classical computers with algorithms like Quantum Phase Estimation (QPE); however, noise significantly hinders the performance of today’s quantum computers. Machine learning has the potential to improve the performance of QPE algorithms, especially in the presence of noise. In this work, QPE circuits were simulated with varying levels of depolarizing noise to generate datasets of QPE output. In each case, the phase being estimated was generated with a phase gate, and each circuit modeled was defined by a randomly selected phase. The model accuracy, prediction speed, overfitting level …

Prostatic artery embolization (PAE) has proven to be an efficacious treatment for urinary symptoms of benign prostatic hyperplasia. PAE is performed in a complex and challenging anatomical field which may pose difficulties from procedural standpoint. Cone beam computed tomography (CBCT) has been proposed as an invaluable tool during the PAE procedure. A review of different techniques and advancements, as well as demonstration of CBCT benefits via a pictorial overview of the salient examples is lacking. The techniques of CBCT are discussed herein and the virtual injection technology as an advancement in CBCT is discussed. To show the merits of CBCT …
