Dynamic Localization Of Endoplasmic Reticulum During Tetrahymena Conjugation, Guerrier Sabrice, Michael Patterson, Kaitlin Crofton, Michael Tucker, Shyhiem Walker

Changes in lipid composition at membrane fusion sites in mating Tetrahymena are thought to involve the endoplasmic reticulum (ER), but its localization to these sites has not been observed. Here we show ER distribution during Tetrahymena mating using TtRET1-GFP and GFP-KDEL. We find that both markers localize to perinuclear membranes and tubular structures that connect perinuclear membrane to plasma membrane at fusion sites. Interestingly, both markers disappear from parental macronuclei after emergence of zygotic macronuclei. These similarities in localization of established ER marker, GFP-KDEL, and TtRET1-GFP reveal TtRET1-GFP as a useful new live cell marker for the ER in Tetrahymena.

Despite a record number of clinical studies investigating various anti-myeloma treatments, the 5-year survival rate for multiple myeloma (MM) patients in the US is only 55%, and almost all patients relapse. Poor patient outcomes demonstrate that myeloma cells are "born to survive" which means they can adapt and evolve following treatment. Thus, new therapeutic approaches to combat survival mechanisms and target treatment resistance are required. Importantly, Mcl-1, anti-apoptotic protein, is required for the development of MM and treatment resistance. To improve data consistency, work efficiency, inspector safety, and cost effectiveness during routine inspections, drones have been increasingly used in recent years to support imaging and scanning over the surface of various elements in a bridge for surface condition assessment. Most drones are operated manually within a visual line of sight and unable to inspect river-crossing bridges completely since not all elements can be viewed by a drone operator using a binocular. This project aimed to develop and evaluate two statistical methods for determining the probability of detection (POD) in corrosion monitoring using Fe–C coated long-period fiber grating (LPFG) sensors. These methods, referred to as the Size-of-Damage-at-Detection (SODAD) method and the Random Parameter Model (RPM), addressed limitations in traditional POD models by considering the time dependency inherent in sensor data. The goal was to assess steel mass loss at 90% detection probability and establish the largest undetectable mass loss threshold at a 95% lower confidence bound. Coatings can prevent their substrate from being contacted by aggressive particles and thus corrosion to preserve structural components. However, the wide selection of coatings for steel girders or truss can greatly impact the serviceability of bridges. This project studied four types of top-coat materials (epoxy, Alkyd, acrylic, and polyurethane) and two types of under-coat materials (epoxy and acyclic). This report details the development and testing of an innovative robotic system for underwater mapping and bridge scour inspection, integrating a 3D sonar with a modified remotely operated vehicle (ROV). The system addresses limitations in traditional scour monitoring methods, such as safety concerns and poor visibility after flooding. The research approach included laboratory experiments, pool testing, and field trials at two bridge sites in Missouri, with procedures developed for both water surface and underwater deployments. This research aimed to delineate the strategies for fostering tolerance among students through multicultural education at Sanggar Anak Alam Nitiprayan in Yogyakarta. Employing a qualitative methodology with a case study approach, the investigation was conducted within the premises of Sanggar Anak Alam Nitiprayan located in Bantul, Yogyakarta. Data gathering methods encompassed interviews, observations, and documentation, with subjects selected purposively. Triangulation was utilised to ensure data validity. Analysis followed the qualitative data analysis technique advocated by Miles & Huberman, involving stages of data collection, reduction, presentation, and conclusion drawing. This research explains how the dimensions of critical thinking develop and are consistent in students by learning directly in a project-based contextual manner through small research that they choose. The research method used is descriptive analysis to analyse the dimensions of students' critical thinking abilities using a quasi-experimental design (a one-group post-test only design). Although certain instances may exhibit indolent characteristics, a significant number of tumors demonstrate rapid progression and a poor prognosis. Mesotheliomas are typically categorized as benign or malignant, with malignant mesothelioma being more frequently linked to asbestos exposure.
