People with HIV (PWH) have often experienced chronic stressors across their lifespan, including adverse childhood experiences (ACES), lifetime economic hardship (LEH), and concurrent stressors associated with living in urban areas (urban stress). Prolonged exposure to stressors might result in differential coping patterns among PWH that can impact care trajectories. We utilized a life course-informed approach to examine chronic stressors as antecedents of latent coping strategies among PWH in care. High-risk alcohol use and non-adherence to anti-retroviral therapy (ART) were examined as consequences of latent coping strategies. Although the basolateral amygdala (BLA) and corticotropin releasing hormone receptor type I (CRHR1) signaling are both central to the stress response, the spatial and circuit-specific distribution of CRHR1 have not been identified in the BLA at a high resolution. We used transgenic male and female CRHR1-Cre-tdTomato rats to topographically map the distribution of BLA neurons and identify whether they are activated by acute stress. Additionally, we used the BLA circuits projecting to the central amygdala (CeA) and nucleus accumbens (NAc) as a model to test circuit-specific expression of CRHR1 in the BLA. Phosphorus (P) deficiency is one of the major constraints for sustainable crop production in calcareous soils. This study aimed to elucidate the key soil characteristics modulating the variability of soil Olsen P in these typical soils. A comprehensive soil sampling initiative (1.5 samples per hectare) was conducted on a 100-ha farm, considering 31 attributes that included soil physical and chemical properties, and geographic attributes. The physical mechanisms usually applied to explain the relativistic electron enhancement have been delved into to elucidate non-adiabatic electron acceleration resulting in the ultra-relativistic electron population observed in the outer radiation belt. We considered multisatellite observations of the solar wind parameters, magnetospheric waves, and particle flux to report an unusual local acceleration of ultra-relativistic electrons under a prolonged high-speed solar wind stream (HSS). A corotating interaction region reaches the Earth’s bowshock on August 3, 2016, causing a minor geomagnetic storm. This dissertation is focused on the development of novel class of ionic materials (IMs), and ionic nanomedicines (INMs) for cancer treatment. These IMs are composed of two therapeutics combined via electrostatic attraction. The impact of various counterions on the drug’s cytotoxic and photophysical properties are unveiled. Moreover, the mechanism of internalization, enhanced toxicity of these materials is investigated by determining their cellular uptake, subcellular localization, and cell death mechanism. Maternal mortality in the United States is significantly higher than in most developed nations and has been increasing since 2000. Persistent between-group disparities among White, Black, and Hispanic women occur in terms of maternal mortality; however, the reason for this is largely unknown. Using the intersectionality theory, this quantitative study examined if racial groups predicted maternal mortality and to what extent relationships were moderated by geographic area and health risk factors. The Department of Veterans Affairs (VA) is responsible for providing care to millions of veterans and has encountered significant obstacles in its EHR modernization efforts, leading to budget overruns, extended timelines, and operational disruptions. The purpose of this qualitative study was to explore law enforcement officers’ perceptions in combating carjacking and auto theft during the COVID-19 pandemic in the southeast region of Louisiana. The framework selected to guide the study was the crime pattern theory by Brangtingham and Brangtinham. Using the purposive sampling method and snowballing sampling method, 10 active law enforcement officers were recruited from agencies across the Southeast region of Louisiana until data saturation occurred. Purpose: Nonsurgical treatment options are increasingly needed for endometrial atypical hyperplasia (AH) and endometrioid endometrial cancer (EEC). Despite promising initial response rates, prospective long-term data and determinants for relapse are limited.

Materials and methods: Follow-up data from patients in our prospective phase II trial of levonorgestrel intrauterine device (LIUD) for AH/G1EEC were collected from medical records. Spatial transcriptomics (Nanostring GeoMX digital spatial profiling) with in silico cell type deconvolution and pathway analyses were employed on longitudinal biopsy samples from five patients across pre-treatment, on-treatment, and relapse.

Results: Of 43 participants exhibiting initial response to LIUD, 41 had follow-up data. Sixteen …

Homeostasis is necessary for epithelia to maintain barrier function and prevent the accumulation of defective cells. Unfit, excess, and dying cells in the larval zebrafish tail fin epidermis are removed via controlled cell death and extrusion. Extrusion coincides with oscillations of cell area, both in the extruding cell and its neighbors. Here, we develop a biophysical model of this process to explore the role of autonomous and non-autonomous mechanics. We vary biophysical properties and oscillatory behaviors of extruding cells and their neighbors along with tissue-wide cell density and viscosity. Purpose: BRAFV600E-mutated colorectal cancer exhibits a strong correlation with DNA hypermethylation, suggesting that this subgroup of tumors presents unique epigenomic phenotypes. Nonetheless, 5-azacitidine, which inhibits DNA methyltransferase activity, is not efficacious in BRAFV600E colorectal cancer in vivo.

Experimental design: We randomized and treated mice implanted with patient-derived tumor xenografts harboring BRAFV600E mutation with control, 5-azacitidine, vemurafenib (BRAF inhibitor), or the combination. Comprehensive epigenomic profiling was conducted on control and 5-azacitidine-treated tumor samples, including DNA methylation, histone modifications, chromatin accessibility, and gene expression. Combinations of epigenetic agents were explored in preclinical BRAFV600E colorectal cancer models.

Geopolitical, health and economic events have led to an accumulated surge in energy prices in the UK at the beginning of the 2020s. In response, studies and media reports show that households have had to reduce the running of their heating systems to cut their energy bills. However, there is limited research on the impact of this context on indoor thermal conditions and occupants’ ability to meet their thermal comfort needs.
