Mitophagy, the process by which cells eliminate damaged mitochondria, is mediated by PINK1 (PTEN induced kinase 1). Our recent research indicates that PINK1 functions as a tumor suppressor in colorectal cancer by regulating cellular metabolism. Interestingly, PINK1 ablation activated the NLRP3 (NLR family pyrin domain containing 3) inflammasome, releasing IL1B (interleukin 1 beta). However, inhibiting the NLRP3-IL1B signaling pathway with an IL1R (interleukin 1 receptor) antagonist or NLRP3 inhibitor did not hinder colon tumor growth after PINK1 loss. To identify druggable targets in PINK1-deficient tumors, ribonucleic acid sequencing analysis was performed on colon tumors from pink1 knockout and wild-type mice. …

Strategies For Lgbtq+ Inclusivity Among Elementary School Aged Youths In Extension, Maru Gonzalez, Michael Kokozos, Sudha Sankar

While there is a growing body of scholarship related to LGBTQ+ inclusive practices among adolescents in 4-H, there is currently no research focused specifically on elementary school aged youths. This paper identifies research-based strategies to foster LGBTQ+ inclusivity among elementary school aged youths in 4-H and provides recommendations for implementation at the individual and institutional level. These strategies include ongoing professional development, supportive adult relationships, inclusive curriculum and programming, and LGBTQ+ inclusive guidance and protocols. The strategies and recommendations for implementation underscore the need for a multi-faceted approach to create more welcoming spaces that benefit all youths.

Automation is becoming increasingly common in manufacturing and assembly plants. The future lies in hybrid workspaces where the strengths of humans and robots complement each other, with robots excelling in precision, speed, and strength, and humans excelling in creativity, emotional intelligence, and complex decision-making. Collaborative robots can foster a more efficient and productive work environment by bridging the gap between human and machine capabilities. Recreational fisheries policy decisions have social and ecological outcomes that depend on whether regulations are understood and meet expectations, which in turn enhances compliance. This study investigates perceptions of management and compliance among boat-based recreational fishers in Western Australia with consideration of demography (age, residence) and fishing behaviour (avidity, bioregion fished). Most respondents considered themselves to be informed about rules governing their fishing and there was strong support for current input and output controls, including area and seasonal closures and possession and size limits. Extension has an active and dynamic health insurance literacy program with certified educators nationwide, but dental insurance literacy is an unmet need. There is an increasing trend of dental practitioners willing to perform basic preventative health measures, particularly hypertension screening. Lack of insurance literacy is a barrier to screening request and follow-up. Participants in our longitudinal study visited their dentists as regularly as they visited their primary health practitioners. This study shows the need for Extension to add dental insurance literacy to the menu of health insurance literacy programs to impact the prevalence of hypertension.

Thorough investigations into air temperature variation across urban environments are essential to address concerns about city livability. With limited research on smaller cities, especially in the American Midwest, the goal of this research was to examine the spatial patterns of air temperature across multiple small to medium-sized cities in Iowa, a relatively rural US state. Extensive fieldwork was conducted utilizing manually built mobile temperature sensors to collect air temperature data at a high temporal and spatial resolution in ten Iowa urban areas during the afternoon, evening, and night on days exceeding 32 °C from June to September 2022. Cooperative Extension can make a significant impact on health in the United States through effective community engagement that focuses on the complex socio-ecological factors that impact health. The Stakeholder Engagement in Question Development Method (SEED Method) employs community based participatory research principles to engage communities in research, problem-solving, and action planning. Projects employing the SEED Method engage community members from diverse backgrounds, create community capacity, result in tangible outcomes, and foment inter-organizational trust and networking. A SEED Toolkit is available on the Extension Foundation Campus site and provides implementation instructions, tools and timelines, and potential applications within Extension.

With climate-smart agriculture as the context, we explore one aspect of transdisciplinarity, the co-development of knowledge (CDK). Challenges were identified via matrix coding and word frequency queries of field day, meeting, and interview transcripts. Focusing on the Research-Extension team, we found they highly value collaboration and participatory-based projects, but developing trust-based relationships, enhancing long-term stakeholder investment, and understanding both team and stakeholder capacity was difficult. This paper informs transdisciplinary projects aimed at climate adaptation by illustrating the need for Research-Extension teams to take incremental steps towards CDK, to engage stakeholders before program planning, and to increase both interaction and reflexivity.

Faithful chromosome segregation in eukaryotes requires the assembly of a bipolar spindle and the faithful attachment of kinetochores to spindle microtubules, which are regulated by various spindle-associated proteins (SAPs) that play distinct functions in regulating spindle dynamics and microtubule-kinetochore attachment. The protozoan parasite Trypanosoma brucei employs evolutionarily conserved and kinetoplastid-specific proteins, including some kinetoplastid-specific nucleus- and spindle-associated proteins (NuSAPs), to regulate chromosome segregation. Here, we characterized NuSAP4 and its functional interplay with diverse SAPs in promoting chromosome segregation in T. brucei. NuSAP4 associates with the spindle during mitosis and concentrates at spindle poles where it interacts with SPB1 and MAP103. …

The purpose of this study was to identify the competencies essential for Florida early career 4-H agent success. This four-round Delphi study asked three expert panels to identify the competencies required for Florida early career agent success in years one and three. The Delphi procedure is a multi-round consensus building process, with the preceding round dictating the questions asked in subsequent rounds (Werner, 2017). The panels identified 55 year-one and 67 year-three competencies that fell into 15 competency areas. These competencies can be used to develop professional training programs to meet the needs of early career agents.

Objective: This study delved into the integration of ChatGPT as a medium for English teacher professional development among Vietnamese English as Foreign Language (EFL) teachers.
Methods: Rooted in the Constructivist Learning Theory and the Technological Pedagogical Content Knowledge (TPACK) framework, the qualitative exploration employed semi-structured interviews with nine teachers categorized into novice, mid-career, and near-end career stages. The objective was to understand teacher experiences, perceptions, and insights regarding the role of ChatGPT in their professional journey.

Understanding how farmers prefer to learn is critical for the development and delivery of successful extension programming. Therefore, Michigan State University Extension specialists conducted an interactive hands-on demonstration workshop (Potato University) addressing pressing issues to Michigan potato production at the Michigan Winter Potato Conference. The new session format emphasized alternative, hands-on research presentation formats and promoted small group interactions between researchers and stakeholders. Overall, we developed an alternative framework to deliver information in a novel format by highlighting research activities in an approachable format for stakeholders that better aligned Extension teaching styles with the learning styles of the potato industry.

In this study, the influences of the aging period and temperature on the flexural properties of CFRP laminates were investigated experimentally and numerically. Samples with cross-ply (CP) and angle-ply (AP) layups were prepared and immersed in water tanks with different temperatures (30 ℃ and 50 ℃). Three-point bending tests were performed for unaged, 15, and 35 days-aged composite laminates. SEM, optical, and Fourier transform infrared spectroscopy were employed to investigate the morphology, chemical structures, and failure mechanisms. Leadership development programs are an exploratory journey that develop Extension faculty to better themselves in service to the organization. In an effort to mitigate potential barriers for participants, it is imperative that organizations work to create conditions conducive for success. We posit barriers for extension professionals include, participants having a fixed-mindset (Dweck, 2016), time constraints (Lindle, 2015; McAlearney, 2006), and the employee burnout epidemic (Fisher, 2018; Seppala & Moeller, 2021). Proactively working against common negative influences, sets up leadership program participants to become more fully immersed in learning and subsequently have a better chance for a positive behavior change.

This article describes the use of paper origami as a training tool for Extension evaluation and capacity building workshops. Three paper origami models have been developed and used consistently since 2021 to benefit 131 new Extension professionals. The use of these models added an element of pleasant surprise for workshop participants and fostered collaboration during training. This article describes the construction and usefulness of the origami models to these new employees. Post-event feedback and testimonials are very encouraging to authors who continue to develop additional origami models for training and motivation.

Growers’ Perceptions And Strategies To Mitigate Spring Frost Injury In Grapevines, Vinicius Cervilieri, Imed Dami, Guilherme Signorini

Uncertain weather patterns raise concerns about frequent spring frosts and injury in grapevines. This article quantifies growers’ perceptions of susceptibility to spring frosts and examines their satisfaction with existing mitigation strategies. Departing from a survey with grape growers, results show that over 80% of sampled growers have experienced damage due to frosts. Over-vine sprinklers, proper site selection, and adoption of late budding cultivars are the practices with the highest satisfaction levels. None of the strategies examined reached the highest satisfaction score. In addition, 61% of respondents showed interest in sprayable products to delay budbreak and circumvent grapevine damage.

Extension professionals have used research verification programs to evaluate the effectiveness of research-based management recommendations in several different aquaculture species. In addition to providing a platform on which to examine current best management practices and Extension recommendations from Land Grant Universities, research verification is valuable in identifying gaps for further research. Perhaps the best benefit of research verification programs are the comradery and relationships developed between cooperating commercial producers and Extension personnel that can lay the groundwork to facilitate future collaborative efforts related to technology transfer in the aquaculture industry.
