During infection, Listeria adhesion protein (LAP), a housekeeping enzyme, acts as a tight junction modulator (TJM) through interaction with Hsp60 to facilitate Listeria monocytogenes translocation across the intestinal epithelial barrier. Here, we used purified LAP as a potential TJM to overcome the limiting and variable effects observed by other agents in the class. We structurally determined the LAP interaction alone and in complex with Hsp60 utilizing cryo-EM and computational analysis. LAP structure resolved at 2.83 Å, forms multimeric interlocking dimers and tetramers, and the N-domain interacts with Hsp60, while the C-domain bridges the bacterial surface receptor InlB. Climate change is expected to intensify salinity and temperature fluctuations in estuarine and coastal habitats. However, the impact of salinity variation on ectotherms’ thermal tolerance remains underexplored, limiting our understanding of the interplay between these stressors. This study investigated salinity-dependent changes in life history traits (survival, growth rate, fecundity, and egg size) over a 33-day exposure to a salinity gradient (12, 17, 22, 27, 32, 35, 40, 45, 50) in the marine annelid Ophryotrocha labronica. Climate change has had devastating effects on tropical coral reefs; however, much less is known regarding how heat stress affects temperate coral. This research focuses on Astrangia poculata (Ellis and Solander, 1786) collected from Narragansett Bay, RI, during the summer and winter seasons and understanding the effect of experimental thermal extremes (i.e., 26 °C) on seasonally different populations. Photosynthetic efficiency (Fv/Fm), symbiont density (via an inverse relationship with pixel intensity), and oxidative stress via reactive oxygen species (ROS) concentrations were measured on symbiotic and aposymbiotic A. poculata. Climate change has caused drastic declines in corals. As sessile organisms, corals acclimate to environmental shifts through genome-wide changes in gene expression, epigenetic modifications, and alterations in microbiome composition. However, alternative splicing (AS), a conserved mechanism of stress response in many organisms, has been under-explored in corals. Using short-term acute thermal stress assays, we investigated patterns of AS in the scleractinian coral Acropora cervicornis during response to low (33°C), medium (35°C), and high (37°C) heat stress and subsequent overnight recovery. Our findings demonstrate reproducible dynamic shifts in AS of at least 40 percent of all genes during response to heat …

Coral reefs are vital marine ecosystems that support many different species and ecosystem services through protecting coastlines from storms and erosion, providing raw materials for medication, and facilitating diversity amongst marine species. Anthropogenic climate change is causing ocean temperatures to exceed the range of coral thermal tolerance resulting in more coral bleaching events globally. However, some corals have the capacity to withstand high temperatures through different thermal tolerance strategies. Amblyomma maculatum Koch (Acari: Ixodidae), Ixodes scapularis Say, and Dermacentor variabilis Say are three hard-bodied ticks responsible for vectoring pathogens that cause most human tick-borne diseases in the United States of America (USA). Tick surveillance is critical to elucidate high-risk areas for targeted vector control and public health interventions. Despite tick-borne diseases having a higher annual incidence compared to mosquito-borne diseases in the USA, tick traps used for surveillance are grossly under-developed compared to the suite of sophisticated mosquito traps available on the commercial market. Knowledge gaps on the dynamics of cattle fever tick-cattle-habitat-climate interactions in South Texas and their influence on the efficacy of treatments to eliminate infestations with the Rhipicephalus (Boophilus) microplus prevent optimal interventions by the Cattle Fever Tick Eradication Program (CFTEP). The CFTEP has been operating in the USA since 1907. The future of coral reefs remains uncertain as intensifying ocean warming continues to drive coral decline and demise worldwide. Corals occurring in extreme environments such as the Persian/Arabian Gulf (PAG), the world’s warmest sea, represent a promising resource to study the genetic architecture of thermal tolerance. However, their thermal capacities remain unresolved. Mixotrophic reef-building corals acquire nutrition via photosynthate translocation from endosymbiotic microalgae and by heterotrophic prey/particle capture. Heterotrophy promotes resistance to, and recovery from, environmental stress, but quantifying coral heterotrophy remains difficult due to complex resource exchanges within the coral holobiont. We interrogated the response of multiple biomarkers to coral heterotrophy using fatty acid profiling and stable isotope analysis of Stylophora pistillata grown along a controlled feeding gradient from pure autotrophy to pure heterotrophy. Introduction: Heart failure with preserved ejection fraction (HFpEF) is a multifactorial cardiovascular disorder characterized by diastolic dysfunction and often associated with hypertension and metabolic disturbances. We aimed to determine the inter-relationship between C/EBP homologous protein (CHOP) in b-cells and HFpEF development.
Methods: Eight-week-old male mice b-cellflox/flox and b-cellCHOP-/- were randomly divided into four groups: control b-cellflox/flox and b-cellCHOP-/- mice subjected to standard diet and water. b -cellflox/flox and b-cellCHOP-/- mice fed a high-fat diet (HFD) and L-NAME (0.5 g/L) for five weeks. Motivation: Here, we make available a second version of the BioTIME database, which compiles records of abundance estimates for species in sample events of ecological assemblages through time. The updated version expands version 1.0 of the database by doubling the number of studies and includes substantial additional curation to the taxonomic accuracy of the records, as well as the metadata. Moreover, we now provide an R package (BioTIMEr) to facilitate use of the database.

Urine is an attractive biomarker analyte for non-invasive longitudinal monitoring of health and disease, particularly for diseases of the genitourinary tract, like prostate and bladder cancer. The composition of an individual's urine reflects both genetic and lifestyle characteristics that differ across geographies and populations, like diet, hydration and other socio-economic factors. While men of African ancestry have elevated prostate cancer risk, it is unclear to what extent this influences urinary biomarkers. We therefore quantified the urinary proteomes of 329 localized prostate cancer patients: 135 self-identifying as White and 194 self-identifying as Black. Introduction
Identifying which patients are at risk for respiratory decompensation, whether from airway compromise or inhalation injury, is an important component of initial burn resuscitation. Findings that traditionally suggest the need for intubation include cutaneous burns to the face and neck, soot seen in the upper airway, and carbonaceous sputum. However, evidence suggests that a significant number of pre-burn center intubations are ultimately unnecessary. The purpose of this investigation was to determine if head and neck burns or sputum seen in the upper airways were correlated with the need for intubation.

We investigate the tidal Love numbers for a non-rotating dark matter admixed neutron star (NS). Tidal Love numbers, which quantify a star's response to the tidal field of a companion object, are an important feature in the gravitational wave (GW) signal and vary depending on the star’s equation of state and dark matter profiles. By determining how the internal properties of the NS affect the GW signal, we hope that improvements in gravitational wave detection will help constrain these parameters by providing additional data from neutron star mergers with other compact objects. Lower extremity injuries are common among basketball athletes. Majority of these injuries are non-contact and therefore preventable. While previous research has explored high-risk movements associated with lower extremity injuries, limited research has explored the influence of settings for these movement patterns, particularly on a basketball court. Betrayal trauma (Freyd, 1996) is a specific form of trauma that occurs when a child is significantly harmed by someone they depend on or trust. Prior research has demonstrated that betrayal trauma is more strongly associated with adverse mental health outcomes than traumas lower in interpersonal betrayal, yet few studies have examined its relationship with suicidal ideation or identified mechanisms that explain this association. Accurate parameter estimation is fundamental to statistical modeling, shaping the validity of inference, prediction, and decision-making. Although Maximum Likelihood Estimation (MLE) and Bayesian methods are well established theoretically, their performance relative to alternative estimators in small-sample, skewed-data contexts remains less clear. This thesis addresses this gap through a systematic evaluation of estimation techniques applied to human response time data from a controlled seek-and-place jigsaw puzzle task, where distributions are characteristically right-skewed.

This dissertation introduces a novel Two-Stage Clustering Framework for the automated identification of human skeletal muscle fiber types using fluorescence microscopy image intensity data. By integrating two complementary clustering methods, this framework overcomes the subjectivity and labor-intensive nature of traditional manual methods, offering an objective and efficient solution for large-scale muscle physiology research. This thesis presents a unified framework for modeling, optimizing, and analyzing collective behavior in flocking systems through a thermodynamic and statistical lens. In the first study, we employ entropy-regularized Gaussian Mixture Models (GMM) to identify distinct thermodynamic regimes—ordered, transitional, and disordered—based on the energy and entropy profiles of simulated agent-based flocks. The second study introduces the Organic Adaptive Flocking Algorithm (OAFA), a biologically inspired, entropy-aware extension of the Boids model. Finite mixture models have been widely used to cluster data consisting of homogeneous subpopulations. In forensic palynology, pollen is used as a proxy to link individuals or items to a crime scene. Mixtures of pollen data-including willow and mustard and blank samples-were analyzed using flow cytometry. Willow and mustard clusters tend to have multivariate normal distributions, while a background cluster has multivariate non-normal distribution. We propose a finite mixture model capable of handling the mixtures of pollen in terms of univariate and multivariate distribution. The growing prevalence of high-frequency, high-dimensional data streams in domains such as finance, cybersecurity, and industrial monitoring has intensified the demand for real-time multiple change point detection methods. These methods are expected to identify distributional shifts as they occur while also determining the number and locations of change points—even in the presence of noise, outliers, and limited labeled data. Claudia Emerson’s Infusion Suite section within her poetry collection Impossible Bottle is a significant contemporary literary representation of cancer. In this twelve-poem sequence, Emerson dispenses with the typical martial metaphors so often used to represent this lived experience. Instead, she focuses on more mundane everyday moments in a way that acknowledges and respects the humanity of all those at the treatment center—especially that of the patients. Emerson refuses to airbrush out the daunting, debilitating aspects of cancer, but she also preserves space for light and life.

This mixed-methods study explores how university presidents leverage adaptive leadership principles and learning organization frameworks to generate institutional growth and sustainability in higher education. Through analysis of presidential narratives, the research examines the intersection of Heifetz et al.’s (2009) adaptive leadership theory and Senge’s (2006) five disciplines of learning organizations, revealing patterns in leadership communication that drive change. The study analyzed narratives from eight university presidents who demonstrated success through their institutions’ minimum 30% endowment growth over five years.
