Analyses Of Chromosome Copy Number And Expression Level Of Four Genes In The Ciliate Chilodonella Uncinata Reveal A Complex Pattern That Suggests Epigenetic Regulation, Laure Bellec, Laura A. Katz

Chilodonella uncinata, like all ciliates, contains two distinct nuclei in every cell: a germline micronucleus and a somatic macronucleus. The macronucleus develops from the zygotic nucleus through a series of chromosomal rearrangements. Macronuclear development in C. uncinata yields a nucleus with highly amplified gene-sized chromosomes. The macronucleus is transcriptionally active during vegetative growth while there is no expression in the micronucleus except during a brief period following conjugation. Gene family evolution in ciliates occurs through complex processes including gene duplication and an alternative processing of scrambled genes. Objective. To assess course instructors’ and students’ perceptions of the Educating Pharmacy Students and Pharmacists to Improve Quality (EPIQ) curriculum.

Methods. Seven colleges and schools of pharmacy that were using the EPIQ program in their curricula agreed to participate in the study. Five of the 7 collected student retrospective pre- and post-intervention questionnaires. Changes in students’ perceptions were evaluated to assess their relationships with demographics and course variables. Instructors who implemented the EPIQ program at each of the 7 colleges and schools were also asked to complete a questionnaire.

Quorum sensing in bacteria is a process of cell-to-cell communication facilitated by small molecules called autoinducers. Interspecies quorum sensing is facilitated by the autoinducer AI-2. The enzyme LuxS catalyzes the formation of AI-2 from S-ribosyl homocysteine (SRH) in a wide variety of bacterial species. Inhibition of LuxS would therefore inhibit interspecies quorum sensing. The goal of this project is to establish biochemical assays for the evaluation of small molecules as potential LuxS inhibitors. The first assay is a conventional colorimetric assay that utilizes Ellman’s reagent to quantify the homocysteine byproduct of DPD production by LuxS. For this assay purified …

The objectives of this study were to develop a transcriptomic reference resource and to characterize polymorphism between isolates of Erysiphe necator (syn. Uncinula necator), grape powdery mildew. The wine and fresh fruit markets are economically vital to many countries worldwide, and E. necator infection can cause severe crop damage and subsequent financial loss. Most of the publicly available sequence data for Erysiphales are from research done on Blumeria graminis f. sp. hordei, barley powdery mildew, which occupies a distinct clade within the Erysiphales. We obtained 641,601 sequencing reads from a Roche 454–FLX next–generation sequencer (RNA–Seq) and performed multiple assemblies using …

This paper explores the relationship between tuition differentials and low-income students in Engineering fields at two public, research intensive universities. Although current reports indicate the need for increased participation within the Science, Technology, Engineering, and Mathematics (STEM) fields, rising tuition prices at the university and program levels may deter low-income students to enroll and persist within STEM, specifically Engineering. The findings reveal that increased costs due to tuition differentials policies are initially offset by financial aid, but over time costs increase, particularly for low-income students. The results highlight the need for comprehensive, time-sensitive financial aid packages that provide students opportunities …

BACKGROUND

The zinc importer LIV-1 is widely distributed, mainly in hormonally controlled tissues. Attention has focused on its role in breast cancer, especially its regulation by estrogen and epidermal growth factor (EGF) and its link to the Epithelial–Mesenchymal Transition (EMT) marker, E-cadherin (CDH1). EMT is important for tumor progression and metastasis. CDH1 expression is under complex control, including by two transcriptional repressors, Snail and Slug. Contrast to CDH1, elevated MMP-9 expression has been linked to increased metastasis and tumor stage.

This thesis presents a structural dynamic modification of vibrating structures by using topology optimization method in order to find the most effective design of a structure supporting rotating machinery. Modal testing and operating deflection shape (ODS) measurement were applied to identify the dynamic characteristics of a pedestal structure. The dynamic characteristics of the structure were ascertained to validate the finite element model. Then, topology optimization method was used to determine the optimum design of the structure. The chemical constituents of Ochrosia oppositifolia have been studied. The compounds were extracted from the bark and leaves of this plant by using n-hexane,dichloromethane and methanol as solvents. The crudes were subjected to extensive chromatographic techniques like preparative thin layer chromatography, column chromatography and high performance liquid chromatography (HPLC). A total of six compounds were obtained. Structural elucidation was established through several spectroscopic methods, such as 1D-NMR (1H, 13C, DEPT, NOE), 2D-NMR (COSY,NOESY, HMQC, and HMBC), UV, IR, and MS (GCMS, LCMS and HRMS) and comparison with the published data. Objective. To assess the impact of a graduate student mentoring program on student interest in research and postgraduate education and on graduate student confidence in mentoring.

Methods. Undergraduate and pharmacy students (mentees) and graduate students (mentors) were matched and participated in the study, which required them to engage in at least 2 discussions regarding research and careers. Mentees completed a pre- and post-assessment of their perceptions of research, postgraduate training plans, and perceptions about mentors. Mentors completed a pre- and post-assessment of their perceptions about themselves as mentors and their confidence in mentoring.

This dissertation is consists of three essays. In the first essay, I analyze how the information contained in the disaggregate components of aggregate inflation helps improve the forecasts of the aggregate series using inflation data from Ghana. Direct univariate forecasting of the aggregate inflation data by an autoregressive (AR) model is used as the benchmark with which all autoregressive (AR), moving average (MA) and vector autoregressive (VAR) models of the disaggregates are compared. The results show that directly forecasting the aggregate series from the benchmark model is generally superior to aggregating forecasts from the disaggregate components. Here we report the isolation of 6 temperate bacteriophages (phages) that are prevented from replicating within the laboratory strain Pseudomonas aeruginosa PA14 by the endogenous CRISPR/Cas system of this microbe. These phages are only the second identified group of naturally occurring phages demonstrated to be blocked for replication by a nonengineered CRISPR/Cas system, and our results provide the first evidence that the P. aeruginosa type I-F CRISPR/Cas system can function in phage resistance. Previous studies have highlighted the importance of the protospacer adjacent motif (PAM) and a proximal 8-nucleotide seed sequence in mediating CRISPR/Cas-based immunity. The purpose of this study was to develop and provide initial validation of an instrument to measure writing attributional style among college students, the Attributional Style Questionnaire for Writing (ASQ-W). A sample of 133 college students from a Midwestern university participated in the current study. A qualitative and quantitative mixed method study was conducted to report the perceived causes for writing success and failure and examine the internal consistency, discriminant validity, and predictive validity of the measure. Microorganisms numerically dominate terrestrial biodiversity, and play important biochemical and geochemical roles in the environments they inhabit. To understand structure and function of complex ecosystems, it is essential to identify primary drivers of microbial diversity and community structure. Historically, the study of microbial ecology was reductive, limited to microbes able to be cultured and enumerated. Microbes meeting this criterion were thought to comprise the dominating members of the environments they were isolated from, however, estimates suggesting up to 99% of the endogenous species are uncultivable with existing methodologies; a concept that reflects experimental failure, rather than a verifiable conclusion. Native American* postsecondary education students encounter several barriers to academic persistence including cultural assimilation issues, limited access to career information services, and an individual sense of duty and responsibility to remain tied to traditional spiritual values and beliefs systems, joined with family pressure to stay home. The Effect Of Presumed Media Influence On College Athletes, Justin E. Anderson

In this article, I examine the notion that perceptions of media have a perceived effect on performance and morale on both the athlete and the team. I test this idea on a sample of college athletes at Nicholls State University (N=94), at the end of the 2011-2012 school year. Findings show that the presumed media influence is displayed in the context of athletics and that there are some indirect effects from one's perceptions of how their teammates were portrayed in the media. Research found that positive media had a perceived effect on the team but not on ones' self. Findings …

The behavior of the macro or mass-production segment of the U.S. brewing industry appears to be paradoxical. Since the end of Prohibition in 1934, the number of independent brewer has continuously declined while the major national brewers, such as Anheuser-Busch, Miller, and Coors, have gained market share. In spite of this decline in the number of competitors, profits and market power have remained low in brewing. Iwasaki et al. (2008) explain this result by providing evidence that changes in marketing and production technologies favored larger brewers and forced the industry into a war of attrition, in which only a handful …

American Orientalism And Cosmopolitan Mixed Race: Early Asian American Mixed Race In The American Literary Imagination, Melissa Eriko Poulsen

Asian American Literature: Discourses & Pedagogies

This paper offers and initial examination of representations of Asian American mixed race during the long period of Asian exclusion and enforced anti-miscegenation when racial mixing was legally proscribed. I argue that this literature must be understood through two categories – Orientalist texts that produce and reproduce dominant discourses about Asian American mixed race and cosmopolitan texts written by mixed-race figures circling in vanguard cliques of art and literature. After tracking the representations of early Asian American mixed race in U.S. literature, this paper offers an analysis of the writing of Asian American mixed-race authors themselves in order to expand …
