Essays in Developmental Psychology
Systematic observation is the process of explicitly defining desired target behaviors and observing them using a systematic set of guidelines. Systematic observation assists in reducing or eliminating bias in experimental designs. The reduction allows for increased reliability and validity therefore, an integral part in experimental designs. This process is widely used in school, home, and clinical settings. The process allows preset rules and guidelines to be to be followed prior to beginning the experiment. The rules and guidelines maintain the design within reasonable boundaries. All the preset rules are inferred prior to starting the experiment.

John Locke, an English philosopher, stated that all babies come to the world with a blank slate or tabula rasa, ready to absorb everything that their outside world has to offer. Behaviorists like John B. Watson similarly advocated the power of nurture (i.e., learning) and disregarded the contribution of nature (i.e., biology, genetics) to human development. Researchers today agree that babies are born with unique characteristics called temperament and that these individual differences in behavior, affect, and attention are believed to have genetic and biological bases. However, researchers also agree that temperament is not immune to environmental influences. In other …

Starting at middle age, adults often suffer from visceral adiposity and associated adverse metabolic disorders. Lineage tracing in mice revealed that adipose progenitor cells (APCs) in visceral fat undergo extensive adipogenesis during middle age. Thus, despite the low turnover rate of adipocytes in young adults, adipogenesis is unlocked during middle age. Transplantations quantitatively showed that APCs in middle-aged mice exhibited high adipogenic capacity cell-autonomously. Single-cell RNA sequencing identified a distinct APC population, the committed preadipocyte, age-enriched (CP-A), emerging at this age. CP-As demonstrated elevated proliferation and adipogenesis activity. Pharmacological and genetic manipulations indicated that leukemia inhibitory factor receptor signaling was …

Hepatoblastoma is a rare pediatric liver malignancy usually treated with surgery and chemotherapy. To explore new treatment options for hepatoblastoma, drug screening was performed using six cell models established from aggressive hepatoblastoma tumors and healthy pediatric primary hepatocytes. Of the 527 screened compounds, 98 demonstrated cancer-selective activity in at least one hepatoblastoma model. The kinesin spindle protein (KSP) inhibitor filanesib was effective in all models and was further evaluated. Filanesib induced G2/M arrest and apoptosis in hepatoblastoma cells at concentrations tolerable to primary hepatocytes. Prominent nuclear fragmentation was observed in filanesib-treated hepatoblastoma cells. Genes participating in cell cycle regulation were …

Frontotemporal lobar degeneration with neuronal inclusions of the TAR DNA-binding protein 43 (FTLD-TDP) is a fatal neurodegenerative disorder with only a limited number of risk loci identified. We report our comprehensive genome-wide association study as part of the International FTLD-TDP Whole-Genome Sequencing Consortium, including 985 patients and 3,153 controls compiled from 26 institutions/brain banks in North America, Europe and Australia, and meta-analysis with the Dementia-seq cohort. We confirm UNC13A as the strongest overall FTLD-TDP risk factor and identify TNIP1 as a novel FTLD-TDP risk factor. In subgroup analyzes, we further identify genome-wide significant loci specific to each of the three …

Motor rehabilitation is a therapeutic process to facilitate functional recovery in people with spinal cord injury (SCI). However, its efficacy is limited to areas with remaining sensorimotor function. Spinal cord stimulation (SCS) creates a temporary prosthetic effect that may allow further rehabilitation-induced recovery in individuals without remaining sensorimotor function, thereby extending the therapeutic reach of motor rehabilitation to individuals with more severe injuries. In this work, we report our first steps in developing a non-invasive brain-spine interface (BSI) based on electroencephalography (EEG) and transcutaneous spinal cord stimulation (tSCS). The objective of this study was to identify EEG-based neural correlates of …

BACKGROUND: Eating disorders (EDs) are severe psychiatric conditions, with prevalence rates ranging from 5.5 to 17.9% in women and 0.6 to 2.4% in men. EDs carry a high risk of chronicity and mortality, highlighting the need for effective prevention strategies. Primary prevention can target the entire population (universal), high-risk groups (selective), or individuals with early signs (indicated). Despite substantial research, prior reviews often show limitations, such as single-author data extraction, lack of quality assessment, reliance on endpoint data, exclusion of obesity prevention programs, or outdated findings. No review has yet evaluated the comparative effectiveness of multiple interventions across risk groups. …

Widowhood, Kirsten Horne, Lisa K. Lashley, Charles J. Golden
Widowhood, Kirsten Horne, Lisa K. Lashley, Charles J. Golden
Essays in Developmental Psychology
Widowhood is defined as the state or period of being a widow or widower. A widow is a female whose spouse has died, while a widower is a male whose spouse has died. Although both men and women are likely to experience widowhood, research indicates that women are more likely to be widowed than men. Research suggests there are two main reasons why women are more likely to be widowed. First, women in average live longer than men. The second reason consists of the idea that women tend to marry older men. (Now that the USA and many other countries …

Writing Skills are an essential component of communicating and develop over time. It does not matter the intention of writing, it is a skill used daily, and we must prepare our students to have the adequate skills they will need to function as writers. Writing skills taught in school are focused on academic writing, which should transfer and generalize across settings. Writing skills take on different forms with a variety of levels of importance. The necessary writing skills needed include; the writing process, which is supported by, mechanics, writing for a variety of purposes, and finally, building technological skills.
Work-Family Conflict, Alexandra M. Daro, Samantha K. Ammons
Work-Family Conflict, Alexandra M. Daro, Samantha K. Ammons
Essays in Developmental Psychology
Individuals will take on many different roles throughout their lives. Three of the largest roles an individual can take on – employee, spouse, parent – often come into conflict with each other. Work-family conflict occurs when demands at work and demands in the family are incompatible. The term “family” can mean different things to different people. Although we often think of family members as individuals tied to us through blood or marriage, families can also be comprised of other loved ones, close friends, and even pets.

Language acquisition is the way that humans learn and attain their spoken language. Infants are biologically equipped to attain four aspects of language. These include phonology, semantics, grammar, and pragmatics. The earliest acquired language begins with phonology, attained as early as three months and make rapid progressions to conversational language by four years of age.

Word Learning Rules In Children, Caitlin F. Herman, Lisa K. Lashley, Charles J. Golden
Essays in Developmental Psychology
The ability to learn new words requires the use of numerous cognitive functions. It is expected that from the time a child begins speaking around 12 months of age to about six years old, children learn approximately 14,000 words (Templin, 1957 as cited in Brady & Goodman, 2014). From an early age, children are exposed to the language used around them and slowly develop their own use of language through recognition of objects, combination of phonemes, and memorization of visual and auditory stimuli. In order to successfully master language and learn new words, a child must integrate these processes to …

Patients with high-grade serous ovarian carcinoma (HGSC) are usually diagnosed with advanced-stage disease, and the tumors often have immunosuppressive characteristics. Together, these factors are important for disease progression, drug resistance, and mortality. In this study, we used a combination of single-cell sequencing and spatial transcriptomics to identify the molecular mechanisms that lead to immunosuppression in HGSC. Primary tumors consistently showed a more active immune microenvironment than did omental tumors. In addition, we found that untreated primary tumors were mostly populated by dysfunctional CD4 and CD8 T cells in later stages of differentiation; this, in turn, was correlated with expression changes …

Ovarian cancer is associated with high rates of patient mortality and morbidity. Laparoscopic assessment of tumor localization can be used for treatment planning in newly diagnosed high-grade serous ovarian carcinoma (HGSOC). While spread to multiple intra-abdominal areas is correlated with worse outcomes, whether other morphological tumor differences are also associated with patient outcomes is unknown. Given the large volume of visual information in laparoscopic videos, we investigated whether deep-learning models can capture implicit features and predict treatment outcomes. We developed a novel deep-learning framework using pre-treatment laparoscopic images to assess clinical outcomes following upfront standard treatment, defined as short progression-free …

Current treatments for advanced prostate cancer (PCa) primarily target the androgen receptor (AR) pathway. However, the emergence of castration-resistant prostate cancer (CRPC) and resistance to AR pathway inhibitors (APPIs) remains ongoing challenges. Here, we present BSJ-5-63, a proteolysis-targeting chimera (PROTAC) targeting cyclin-dependent kinases (CDKs) CDK12, CDK7, and CDK9, offering a multipronged approach to CRPC therapy. BSJ-5-63 degrades CDK12, diminishing BRCA1 and BRCA2 expression and inducing a sustained "BRCAness" state. This sensitizes cancer cells to PARP inhibitors (PARPis) regardless of their homologous recombination repair (HRR) status. Furthermore, CDK7 and CDK9 degradation attenuates AR signaling, enhancing its therapeutic efficacy. Preclinical studies, including …

Background/Objectives: The fusion of the TFCP2 gene with either EWSR1 or FUS typically results in a spindle cell and/or epithelioid variant of rhabdomyosarcoma. This is an ultra-rare type of sarcoma, with most of our knowledge about these coming from case reports and small case series. Herein, we describe the clinical characteristics and treatment course of 10 patients with TFCP2 fusion sarcomas.
Methods: We identified 10 patients in our hospital system with TFCP2 fusion sarcomas and 43 previously reported cases in the literature. We assessed primary tumor characteristics, treatment regimens, and survival rates among all cases.
Results: We find that …

Obesity is a risk factor for osteoarthritis (OA), and leptin is among the adipokines implicated in obesity-induced OA. However, the specific role of leptin in OA severity and pain is not known. Using lipodystrophic (LD) mice, we show that fat-secreted factors are required for knee OA development, implicating a fat-cartilage cross-talk. Fat pad implantation or systemic leptin restoration in LD mice reintroduced structural OA and pain, whereas implantation of leptin-deficient fat pad did not change OA susceptibility. Isochronic parabiosis and spatial transcriptomics confirmed that a fat-joint cross-talk likely occurred via soluble mediators. Global unsupervised multiomics of conditioned media from fat …

The challenge of attracting qualified behavioral health professionals to positions in rural areas has been widely recognized. Underserved population segments, issues unique to clinicians practicing in rural areas, and a lack of effective recruitment incentives impact not only an organization but also the community it serves. The subject of this qualitative case study was a private behavioral health office located in a rural community. Semistructured interviews were held with five behavioral health leaders of this organization in addition to a review of organizational records. The Baldridge Performance Excellence Program was the conceptual framework that grounded this study. Qualitative analysis, including …

Organization Change Process Initiative Recurrent Emergency Room Visits By Mental Health And Substance Use Patients, Julie Marie Tucker
Walden Dissertations and Doctoral Studies
The rising rates of patients diagnosed with mental health and substance use disorders (MHSUD) signify a critical national crisis, worsened by the COVID-19 pandemic, as reported by the United States Department of Health and Human Services. This increase has reduced access to essential resources and treatment options, resulting in overcrowded emergency rooms (ER) and extended wait times. Addressing these issues is vital for nursing practice, as they significantly influence patient care and health outcomes, especially for vulnerable populations. This doctoral project explores whether an organizational change designed to raise awareness of updated community resources impacts MHSUD patient visits to the …

Field failures of products are frequently modeled using a constant failure rate assumption along with biased device usage times. This methodology provides convenience but may be inaccurate due to the failure rate assumptions and unavailability of precise device usage data. Techniques described herein provide for computing the precise device usage time along with assuming varying failure rates to better estimate field failures.
