The RNA exosome is an evolutionarily conserved, multiprotein complex that is the major RNase in 3' processing and degradation of a wide range of RNAs in eukaryotes. Single amino acid changes in RNA exosome subunits cause rare genetic diseases collectively called exosomopathies. However, distinguishing disease-causing variants from nonpathogenic ones remains challenging, and the mechanism by which these variants cause disease is largely unknown. Previous studies have employed a budding yeast model of RNA exosome-linked diseases that relies on mutating the orthologous yeast genes. This paper proposes the development of a European Open Science Cloud (EOSC) Federation national node for Ireland (EOSC Ireland). EOSC Ireland will be a digital platform that connects researchers and research infrastructures across Ireland to the European-wide EOSC Federation. It allows researchers to share and access data, tools, and applications, making national and international collaboration easier. By engaging with EOSC Ireland, researchers can find and contribute new datasets, share analysis, and provide analytical tool and code that others can use, helping to advance scientific discovery. Climate change has been altering ecosystems across the world at a substantial rate, disrupting the delicate ecophysiological relationships between ecosystems and the organisms that live in them. Ectotherms, such as amphibians, rely on external temperatures to behaviorally thermoregulate, making it especially imperative to study their thermal biology. The clinical utility of liquid biopsy (LB) for pancreatic ductal adenocarcinoma (PDAC) remain understudied. Our single-institution cohort of 311 PDAC patients with non-tumor tissues informed LB found 81.2% positivity (N = 186) in metastatic cases and in 52.4% (N = 43) of localized disease. KRAS mutations were detected in 64.6% (N = 148) of metastatic cases and 16% (N = 13) for localized disease. Positive LB, especially KRAS mutation detection, is associated with worse overall survival (OS) in metastatic PDAC (median 14.5 vs. 31.3 months, HR = 2.7, 95%CI = 1.7-4.3, P <  0.0001). The positive concordance rates of KRAS and TP53 mutations were 63% and 68% in metastatic disease but only 7% (KRAS) and 33% (TP53) in localized disease, respectively. Among the 41 patients who underwent serial liquid biopsy testing, 25% tested positive after an initial negative result. LB detects therapeutically targetable mutations in 58.5% of PDAC patients and is associated with OS.

BCL11A, a transcription factor, is vital for hematopoiesis, including B and T cell maturation and the fetal-to-adult hemoglobin switch. Mutations in BCL11A are linked to neurodevelopmental disorders. BCL11A contains two DNA-binding zinc-finger arrays, low-affinity ZF2-3 and high-affinity ZF4-6, separated by a 300-amino-acid linker. ZF2-3 and ZF4-5 share 73% identity, including five out of six DNA base-interacting residues. These arrays bind similar short sequence motifs in clusters, with the linker enabling a broader binding span. Crystallographic structures of ZF4-6, in complex with oligonucleotides from the β-globin locus region, reveal DNA sequence recognition by residues Asn756 (ZF4), Lys784 and Arg787 (ZF5). A …

Background/aim: Epithelial ovarian cancer is the second most prevalent type of gynecological cancer. We aimed to investigate prognostic factors related to survival in patient with 2014 FIGO stage IIIC high-grade serous ovarian cancer (HGSOC).

Background/aim: Patients experience moderate-to-severe pain, especially in the first days after lumbar disc surgery. Poorly controlled pain in the postoperative period negatively affects patient outcomes. Dexmedetomidine is a highly selective α2 adrenoceptor agonist with demonstrated analgesic efficacy. However, conflicting results have been reported in the current literature regarding the efficacy of dexmedetomidine in this surgery. In this study, we tested the hypothesis that dexmedetomidine safely improves pain scores and reduces opioid consumption in lumbar microdiscectomy.

Background/aim: This retrospective study aimed to compare the clinical outcomes, including reoperation rates, mortality, and valve size measurements, in patients undergoing sternotomy and thoracotomy for cardiac surgery, particularly focusing on tricuspid valve replacements. Lateralized auditory processing is essential for specialized functions such as speech processing, typically dominated by the Left Auditory Cortex (ACx) in humans. Hemispheric specializations also occur in the adult mouse ACx, but their developmental origins are unclear. Our study finds that the Left and Right ACx in mice reach developmental milestones at different ages. Thalamocortical responses and maturation of synaptic dynamics develop earlier in the Right ACx than the Left. We show that this timing offset predicts hemisphere-dependent differences in sensory-driven plasticity. The flagellar calcium channel CatSper is essential for male fertility, as it regulates calcium influx to trigger the hyperactive motility required for sperm to fertilize the egg. Precise activation of CatSper is critical, as premature activation can impair sperm function. While optimal temperature is known to influence fertilization, its effect on CatSper remains unknown.
