As sea levels rise and climate disasters become more prevalent, this will displace residents. The importance of a scalable adaptable emergency shelter solution is vital. By proposing an renovation of existing big-box warehouses, such as Walmart, into refuge centers that can accommodate large number of displaced individuals. These refuge centers with their elevated platforms, strategic geographic distribution, and proximity to their flood zones, were well suited for elevated, climate resilient shelters. Gdgse: An Algorithm To Evaluate Pathway Enrichment By Discretizing Gene Expression Values, Jiangti Luo, Qiqi Lu, Mengjiao He, Xiaobo Zhang, Xiang Yang, Xiaosheng Wang

We proposed gdGSE, a novel computational framework for gene set enrichment analysis. Unlike conventional methods that rely on continuous gene expression values, gdGSE employs discretized gene expression profiles to assess pathway activity. This approach effectively mitigates discrepancies caused by data distributions. This algorithm consists of two steps: (1) applying statistical thresholds binarizing gene expression matrix, and (2) converting the binarized gene expression matrix into a gene set enrichment matrix. Our results demonstrated that gdGSE could robustly extract biological insights from a diverse array of simulated and real bulk or single-cell gene expression datasets. Background: Posterior circulation strokes comprise one-fourth of ischemic strokes and can arise from many causes. One rare etiology is transient occlusion of the vertebral artery (VA) with head rotation due to an osteophyte, termed bow hunter's syndrome. The authors present the case of recurrent thromboembolic posterior strokes secondary to transient reopening of an osteophyte-occluded VA.

Observations: A 71-year-old man presented with dizziness and visual changes with a history of prior cervical fusion and myocardial ischemia, controlled with Plavix and aspirin. CT angiography of the neck revealed osteophyte occlusion of the right VA at C4, and MRI revealed a right posterior …

Background: Signed language integrates complex sensorimotor processes involving visual and motor networks. The contribution of language regions such as Broca's and Wernicke's areas during signing is not well established. Here, the authors report the case of a 45-year-old right-handed male with congenital deafness, who communicates using American Sign Language (ASL) and Signed English, presenting with focal seizures. Imaging showed a diffuse infiltrative glioma in the right cerebral hemisphere.

Efficiency is essential to support responsiveness w.r.t. ever-growing datasets, especially for Deep Learning (DL) systems. DL frameworks have traditionally embraced deferred execution-style DL code—supporting symbolic, graph-based Deep Neural Network (DNN) computation. While scalable, such development is error-prone, non-intuitive, and difficult to debug. Consequently, more natural, imperative DL frameworks encouraging eager execution have emerged but at the expense of run-time performance. Though hybrid approaches aim for the "best of both worlds," using them effectively requires subtle considerations to make code amenable to safe, accurate, and efficient graph execution—avoiding performance bottlenecks and semantically inequivalent results. Ultrahigh-energy cosmic rays are known to be mainly of extragalactic origin, and their propagation is limited by energy losses, so their arrival directions are expected to correlate with the large-scale structure of the local Universe. In this work, we investigate the possible presence of intermediate-scale excesses in the flux of the most energetic cosmic rays from the direction of the supergalactic plane region using events with energies above 20 EeV recorded with the surface detector array of the Pierre Auger Observatory up to 2022 December 31, with a total exposure of 135,000 km2 sr yr. This research aims to investigate and explain three-dimensional (3D) learning through the phenomenon-based science education approach. The study employed documentary review research methodology to analyze sources that introduce phenomenon-based science education (including research articles, new science education standards, and Iran's national science curriculum guide). The findings indicate that three-dimensional learning, with its emphasis on disciplinary core ideas, crosscutting concepts, and science and engineering practices, provides a comprehensive framework for understanding and explaining natural phenomena. This master's thesis explores how oral histories from Vietnam veterans provide vital counter-narratives to traditional, often impersonal, military histories. Inspired by the author's grandfather, a Vietnam veteran, the project collects and analyzes ten interviews conducted as part of the Library of Congress’ Veterans History Project. Background: This study examined the relationship between hearing device price and sound quality. Method: A novel consumer-centric metric of sound quality (“SoundScore”) was used to assess hearing devices’ audio performance. Each hearing device is tested with two fittings. The “Initial Fit” is designed to approximate the most likely fitting for an individual with a mild-to-moderate sloping sensorineural hearing loss. The “Tuned Fit” includes adjusting parameters optimized to hit prescriptive fitting targets (NAL NL2) on an acoustic manikin. Each fitting is evaluated across five dimensions. The primary objective of this scoping review (ScR) was to assess the breadth and type of evidence related to time-restricted eating (TRE) as an intervention to modify metabolic health outcomes in individuals with diagnosed metabolic syndrome (MetS), a major health challenge due to increasing prevalence and association with other chronic diseases. MetS comprises three or more of hypertension, hypercholesterolaemia, dyslipidaemia, dysregulated glucose homeostasis, and abdominal obesity. TRE, also known as time-restricted feeding (TRF), restricts food intake to specific time windows within a day, for example, a 10-h eating period between 10:00 and 20:00. Genome-wide association studies (GWAS) have been key in expanding our understanding of the genetic contributions to common diseases. However, these genetic associations frequently fail to clarify causal disease mechanisms, as they often fall in non-coding regions and are difficult to interpret. One solution is to perform a GWAS for the levels of a cellular trait, known as quantitative trait locus (QTL) mapping. Through methods such as colocalization, Mendelian Randomization, and transcriptome/proteome-wide association studies, the QTL variants can be compared to disease GWAS, identifying shared variation between cellular and disease traits. Patients with common variable immunodeficiency (CVID) who develop noninfectious complications (NIC) have worse clinical outcomes than those with infections only (INF). While gut microbiome aberrations have been linked to NIC, reductionist animal models that accurately recapitulate CVID are lacking. Our aim in this study was to uncover potential microbiome roles in the development of NIC in CVID. We performed whole-genome shotgun sequencing on fecal samples from CVID patients with NIC, INF, and their household controls. Oligodendrocytes are the myelinating cells of the central nervous system, known for modulating signal transmission, refining neural circuits, and providing metabolic support to axons. Oligodendrocytes are generated throughout life from oligodendrocyte precursor cells (OPCs) and are damaged or lost in demyelinating and neurodegenerative diseases and age-related pathologies. Thus, understanding the cellular checkpoints that occur during the generation and degeneration of oligodendrocytes is crucial for maintaining their population in health and recovering it in disease and aging.

The complement system is an ancient pathway that serves dual roles as a key regulator of neuroinflammation and a critical regulator of neural networks. Few studies have addressed how these two pathways may interact during infection of the central nervous system. While some components of complement are expressed in the healthy developing brain, complement component 3a receptor (C3aR) is a G protein coupled receptor that is expressed by astrocytes, microglia and infiltrating immune cells during inflammatory states. Density Functional Theory (DFT) is widely used as a powerful tool for studying the electronic structure of materials. However, due to the local or semilocal nature of exchange-correlation functionals, such as the local density approximation and generalized gradient approximation functional, DFT often underestimates the electronic band gap. Many-body perturbation theory (MBPT) within the GW approximation provides more accurate quasiparticle energies calculation by incorporating many-electron screening effects. Additionally, solving the Bethe-Salpeter Equation (BSE) allows for a detailed analysis of excitonic effects in optical spectra. Black Americans disproportionately experience higher rates of health challenges and mortality compared to White Americans, yet the mechanisms underlying these disparities remain inadequately understood. Prominent theoretical models highlight stress and resulting allostatic load as putative mechanisms through which these Black-White racial disparities emerge; however, empirical data supporting such models with regard to mortality remains sparse. The current study examined the potential role of cumulative stress exposure across the life span and elevated levels of C-Reactive Protein, a biomarker of inflammation, in contributing to the longstanding increased mortality risk among Black relative to White Americans. Breast cancer (BC) is the most common non-keratinocyte cancer diagnosed in women in the United States with approximately 300,000 new cases diagnosed each year. Despite a myriad of treatment options, BC remains the second-most deadly cancer. Estrogen receptor-positive (ER+) BC comprises 60-70% of BC diagnoses and is treated with endocrine therapies that limit ER signaling. Despite endocrine therapy options, ~1/3 of patients experience recurrence within 10-20 years of diagnosis. Novel therapeutic strategies are required to limit BC recurrence-related morbidity and mortality.

Inborn errors of immunity are monogenic disorders of the immune system that lead to immune deficiency and/or dysregulation in patients. Identification of precise genetic causes of disease aids diagnosis and advances our understanding of the human immune system; however, a significant portion of patients lack a molecular diagnosis. Somatic mosaicism, genetic changes in a subset of cells, is emerging as an important mechanism of immune disease in both young and older patients. Here, we review the current landscape of somatic genetic errors of immunity and methods for the detection and validation of somatic variants.

Aligning a large language model (LLM) to individual preferences is difficult to perform at scale. This disclosure describes techniques that leverage direct preference optimization (DPO) and low-rank adaptation (LoRA) to enable scalable alignment of artificial intelligence (AI) models. With user permission, user’s edits to suggestions from the model are obtained. The context of the user’s written data or interaction with the LLM is obtained. This disclosure describes techniques that leverage remotely provisioned attestation certificates (RPAC) to actively deter device theft. Devices that are reported as lost are identified, and a robust access restriction mechanism is activated that denies both device-local and backend services to the device. To deny service, a bipartite mechanism is deployed that includes enabling a user to declare that their device is stolen and conveying to various services that the device is stolen.
