Alternative teacher certification programs are used nationwide to address the critical teacher shortage in special education. There is a gap in knowledge regarding the challenges that early childhood special education teachers experience, which make them leave the profession at twice the rate of general education teachers. The purpose of this basic qualitative study was to explore alternatively certified early childhood special education teachers’ perspectives on remaining in their teaching positions and the support they need to continue teaching. The rapid evolution of computer science (CS) as an integral part of K-12 public education in the United States has highlighted significant equity and diversity issues. Guided by critical race theory and community cultural wealth, this basic qualitative study explored high school CS teachers’ perceptions of the challenges and opportunities related to recruiting, retaining, and ensuring the success of students of color. Thematic analysis of data collected using semistructured interviews with 12 high school CS teachers revealed key challenges, including curriculum rigidity, funding inequities, and lack of administrative support. This staff education project aimed to enhance nurses’ knowledge on medication administration error prevention through targeted education interventions. Medication administration errors are a significant concern in health care, leading to patient harm and reduced quality of care. This project addressed the gap in nursing knowledge regarding strategies to prevent these errors, with the goal of improving patient safety and outcomes. IMPORTANCE: Therapeutic responses in acute myeloid leukemia (AML) demonstrate considerable variability both across and within established risk stratifications and age groups. Moreover, significant racial disparities persist, with Black patients experiencing inferior survival outcomes compared with their White counterparts.
OBJECTIVE: To validate the association of the previously reported 10 single nucleotide variant (SNV)-based ara-C pharmacogenomics score (ACS10) with survival outcomes in a large cohort of pediatric AML patients; to evaluate whether ACS10 remains relevant in an adolescent and young adult (AYA) population of patients with AML treated with similar intensive induction chemotherapy protocols; and to assess the association of ACS10 with …

The court affirmed dismissal, holding that Article 78 proceedings provide sufficient pre-compliance review for record demands, and the low risk of coercion mitigates Fourth Amendment concerns. Due process was satisfied by opportunities for landlords to contest vacancy calculations through public hearings and subsequent Article 78 review, ensuring constitutional muster.

Research on Generative Artificial Intelligence (GAI) in higher education primarily focuses on faculty use and experiences, with limited attention given to why some abstain from using it. Drawing from Innovation Resistance Theory, this study aims to address this gap by exploring the perceptions of both faculty users and non-users of GAI, identifying the reasons and concerns why they avoid GAI. A survey of 294 full-time higher education faculty from two mid-size U.S. public universities was conducted. Using qualitative and quantitative analysis, results show that over one-third of the faculty members opted out of using GAI for five primary reasons: not …

In this Outlook, Huang et al. discuss a study in this issue of Genes & Development by Fraire et al. that shows that a deficiency in miRNA processors Drosha and Dicer and consequent cell cycle gene derepression promote pineoblastoma development. The authors highlight the heterogeneity of pineoblastoma's pathogenic mechanisms and its implications for therapeutic interventions in the clinic.

Cell plasticity is a hallmark of cancer, enabling tumor cells to acquire multiple phenotypes responsible for tumor progression, metastasis, and therapy resistance. In this issue of the JCI, Kawai and colleagues leveraged genetically engineered mouse models (GEMM) of pancreatic ductal adenocarcinoma (PDAC) to demonstrate that loss of Pbrm1, a member of the SWI/SNF complex, drives dedifferentiation and aggressive tumor features. Pbrm1 loss activated a program of epithelial-to-mesenchymal transition (EMT) and allowed the emergence of poorly differentiated histologies that are commonly associated with high recurrence rate and dismal prognosis. Hyaluronan (HA) in the extracellular matrix promotes epithelial-mesenchymal transition (EMT) and metastasis; however, the mechanism by which the HA network constructed by cancer cells regulates cancer progression and metastasis in the tumor microenvironment (TME) remains largely unknown. In this study, inter-α-trypsin inhibitor heavy chain 2 (ITIH2), an HA-binding protein, was confirmed to be secreted from mesenchymal-like lung cancer cells when cocultured with cancer-associated fibroblasts. ITIH2 expression is transcriptionally upregulated by the EMT-inducing transcription factor ZEB1, along with HA synthase 2 (HAS2), which positively correlates with ZEB1 expression. Background: Melanoma brain metastases (MBM) have a unique molecular profile compared to extracranial metastases (ECM). Description of the biological features and clinical outcomes of MBM will facilitate the design of rational therapies.
Methods: We examined the mutational landscape and gene expression profiles of MBM (74 patients) and ECM (34 patients) in paired patient samples from a previously published dataset with whole-exome sequencing (WES) and RNA sequencing (RNAseq) data from MD Anderson Cancer Center (MDACC). We also present findings from MBM from a new cohort of 14 patients from Duke University to strengthen investigation of somatic mutations and gene expression profiles. …

Manipulating biology through chemistry is older than the discipline of chemistry itself. Traditionally, selectivity of oral or intravenous drugs relied on preferential drug-receptor binding. A novel approach exploiting image-guided techniques to impart spatial selectivity opens up a wide range of new possibilities for study and manipulation of biology. Motivated initially by the poor prognosis for solid tumors such as liver cancer, we demonstrate the use of an extreme form of in vivo chemistry for targeted delivery of 2-propylpentanoyl chloride in a swine model. When a jury awarded one million dollars in punitive damages to climatologist Michael Mann, in his defamation suit against writer Mark Steyn (who accused Mann of scientific fraud), it was newsworthy, even surprising, because opinions regarding scientific matters are rarely actionable. And yet it was not seen as a groundbreaking event in terms of legal doctrine, since the outcome was easily justifiable— Steyn’s accusation was provably false. IMPORTANCE: Atopic dermatitis (AD) is the most common inflammatory disease in childhood, and children with AD are more likely to develop other allergic diseases, including food allergy, allergic rhinitis, and asthma.
OBJECTIVE: To determine the phenotypes of AD expression across 12 US birth cohorts and identify factors associated with phenotype and development of allergic diseases.

The first several months of President Donald Trump’s second administration have been marred by the chaotic and often unlawful use, or threatened use, of wartime and terrorism-related authorities for what are essentially domestic immigration or criminal law enforcement purposes. This is not the first time a president has sought to push the boundaries of his wartime powers, or outright claimed those he did not have, but it may well be the most extreme. The global utility of acceptance and commitment therapy highlights the need for adapting measures that can effectively capture the richness of psychological flexibility. One such instrument is the Comprehensive Assessment of Acceptance and Commitment Therapy Processes (CompACT). We translated the CompACT into Luganda and adapted it for use in Uganda. The original CompACT was translated into the Luganda language and reviewed through a series of evaluations. Nine mental health professionals participated in one-on-one interviews, while a focus group of eight culturally competent laypersons provided further insights. Their feedback resulted in revisions to enhance the instrument’s clarity, relevance, acceptability and completeness. …

Understanding how poultry perceive and interpret their environment is essential to enhancing their welfare. Animal welfare science relies on measures of the behavioral and physiological components of affective states (positive and negative) as welfare indicators. Purpose: Analyze phenotypic data from knockout mice with late-adult retinal pathologic phenotypes to identify genes associated with development of adult-onset retinal diseases.
Methods: The International Mouse Phenotyping Consortium (IMPC) database was queried for genes associated with abnormal retinal phenotypes in the late-adult knockout mouse pipeline (49-80 weeks postnatal age). Importance: Although brain iron is necessary for neurogenesis, myelination, and neurotransmitter synthesis, iron deficiency (ID) is defined solely based on hematological outcomes.
Objective: To examine the association of ID without anemia with basal ganglia (BG) iron content and its structural and functional sequelae in adolescents.

Importance: While epinephrine is commonly administered in children with out-of-hospital cardiac arrest (OHCA) via an intraosseous (IO) or intravenous (IV) route, the optimal route of epinephrine delivery is unclear.
Objective: To evaluate the association between the route of epinephrine administration (IO or IV) and patient outcomes after pediatric OHCA.
Design, setting, and participants: Retrospective cohort study of pediatric patients (aged < 18 years) with nontraumatic OHCA treated by emergency medical services who received prehospital epinephrine either via an IO or IV route. Patients were included in the Resuscitation Outcomes Consortium Epidemiologic Registry, a prospective OHCA registry at 10 sites in the US and Canada from April 2011 to June 2015. Data analysis was performed from May 2024 to April 2025.
Exposure: Epinephrine administration route: IO or IV route.

IMPORTANCE: Accurate estimation of surgical transfusion risk is important for perioperative planning and effective resource allocation. Most machine learning models in health care are not validated or perform poorly in external settings.
OBJECTIVE: To externally validate a publicly available machine learning algorithm (Surgical Personalized Anticipation of Transfusion Hazard [S-PATH]) to estimate red cell transfusion during surgery within a national sample of hospitals.
