Despite the increasing reliance of rainwater harvesting in semi-urban areas, there is a lack of comprehensive localized data on the safety of harvested rainwater, particularly in areas such as Wazo Hill where communities predominantly depend on it. This study evaluates the quality of harvested rooftop rainwater intended for human consumption and assesses its compliance with the World health Organization (WHO) and Draft East Africa Standards (DEAS) for drinking water. This paper presents the simulation-based design of a synchronous reluctance motor (SynRM) coupled electromechanical system suitable for the hybrid electric vehicle. The study considers SynRM with a 75kW power rating with the capability to produce up to 125kW maximum power for the electric vehicle. The focus is mainly on rotor and stator design; the angles of the flux-barrier ends are considered to reduce the torque ripple due to the slot harmonics. Reluctance machines are indeed competitive for applications requiring high efficiency at low cost. Since the first approval of tisagenlecleucel in 2017, pediatric and young adult patients with relapsed/refractory B-cell acute lymphoblastic leukemia (B-ALL) may receive this CD19-directed chimeric antigen receptor T-cell therapy. We report real-world data from the Center for International Blood and Marrow Transplant Research (>2.5 years of follow-up). As of 4 May 2022, 768 patients with B-ALL had received tisagenlecleucel. Patients aged ≥18 and < 18 years of age (including those < 3 years) were treated during first relapse (26.6% and 26.7% [< 3 years, 44.8%], respectively) or primary refractory disease (12.4% and 12.1% [< 3 years, 15.5%], respectively) with 17.6% and 11.6% (< 3 years, 13.8%), respectively, having high disease burden (≥50% bone marrow [BM] blasts) and 20.2% and 20.2% (< 3 years, 13.8%), respectively, having low disease burden (>0 to < 5% BM blasts). Among patients with ≥12 months postinfusion follow-up (n = 578; median follow-up, 32.1 months), the best overall response of complete remission/complete remission with incomplete blood count recovery was 86.0%. The 12-month relapse-free survival (RFS) and overall survival (OS) were 61.8% and 79.4%, respectively, whereas the 24-month RFS and OS were 50.3% and 63.8%, respectively. Age (< 18 years) and disease burden (< 50% BM blasts) were associated with better outcomes. Previous inotuzumab therapy and KMT2A rearrangement were associated with worse outcomes. Older patients (≥18 years) experienced a higher rate of any-grade cytokine release syndrome (CRS) associated with higher disease burden before infusion. Any-grade CRS and neurotoxicity were lower in patients aged < 3 years. Extended follow-up continues to demonstrate high rates of RFS and favorable safety in this population.

Metastatic disease is the primary driver of cancer-related mortality. In prostate cancer (PCa), patient survival decreases dramatically once the disease spreads to distant organs, with over 80% of metastases occurring in the bone. This dissemination is often followed by a clinically silent period of tumor dormancy that can last for years, creating a critical reservoir of cells poised for lethal relapse. A significant barrier to understanding and targeting this dormant phase has been the lack of robust experimental models. Poly(ADP-ribosyl)ation (PARylation) is a post-translational modification mediated by ADP-ribosyltransferases, known as PARPs, which attach ADP-ribose units onto proteins, forming negatively charged multimeric chains. This modification relaxes chromatin at DNA damage sites, facilitating repair machinery access. Additionally, PAR polymers serve as docking platforms for effector proteins, termed PAR "readers", commonly involved in DNA repair. The recruitment of these proteins is mediated through conserved protein domains, including RNA recognition motifs (RRMs). Using an array of hundreds of recombinant RNA-binding domains, we systematically examined RRM interactions with PAR chains of varying lengths. Kidneys are central in maintaining acid-base homeostasis by recovering filtered bicarbonate (HCO3-) in the proximal tubule and by secreting H+ in the collecting duct. Here, we demonstrate a critical role of the exchange protein directly activated by cAMP (Epac) signaling, and particularly the Epac2, in governing renal adaptation to dietary acid load. RNAseq analysis of the renal cortical area revealed that Epac1&2 deficiency was associated with changes in gene profile seen in acidosis. In the phase 3 AGILE study, after a 12.4-month median follow-up, ivosidenib, a mutant isocitrate dehydrogenase 1 (IDH1) inhibitor, combined with azacitidine significantly improved event-free survival, overall survival (OS), and complete remission rates compared with placebo-azacitidine in patients with newly diagnosed IDH1-mutated acute myeloid leukemia (AML), who were unfit for intensive chemotherapy. This post hoc analysis reports long-term follow-up results from AGILE after a median follow-up of 28.6 months. Overall, 148 patients were randomized to receive ivosidenib-azacitidine (n = 73) or placebo-azacitidine (n = 75). Background/Objectives: Gene rearrangements involving oncogenes are major drivers in acute leukemia, influencing disease classification, prognosis, and therapeutic decision-making. Targeted RNA sequencing (RNA-Seq) panels capable of detecting intergenic and intragenic fusions across multiple genes are increasingly used in diagnostic settings. However, comparative evaluation with orthogonal technologies remains limited.

Material and Methods: We compared the performance of a 108-gene anchored multiplex PCR (AMP)-based RNA-Seq panel with that of Optical Genome Mapping (OGM) in 467 acute leukemia cases. Importance: Extremely preterm infants are at high risk for bronchopulmonary dysplasia (BPD) and death. Multiple small randomized clinical trials showed that a combination of budesonide with surfactant compared with surfactant alone reduced BPD or death.

Objective: To determine if early intratracheal administration of a combination of budesonide (0.25 mg/kg) mixed with surfactant, compared with surfactant alone, reduces physiologic BPD or death by 36 weeks' postmenstrual age in extremely preterm infants.

[Objective] To improve the functional characteristics of egg white protein (EWP) by tagatose glycosylation. [Methods] Firstly, the differences in antioxidant activity, emulsifying property, emulsion stability, foaming ability, and foaming stability are compared among glycosylated EWP (G-EWP), heat-treated EWP (H-EWP), and EWP. Then, the mechanism underlying the structural changes of G-EWP is investigated by sodium dodecyl sulfate-polyacrylamide gel electrophoresis, Fourier transform infrared spectroscopy, circular dichroism, scanning electron microscopy, and particle size analysis. [Results] Compared with EWP and H-EWP, G-EWP exhibits significantly improved antioxidant activity, emulsifying property, emulsion stability, foaming ability, and foaming stability (P<0.05). After glycosylation, the molecular weight increases. Tagatose is covalently grafted to egg white protein, with the content of α-helix, β-sheet, and β-turn changing and the morphology changing from spherical to large block-like complexes. The average particle size decreases from 153.53 nm to 129.33 nm (P<0.05), and the zeta potential value decreases from-22.68 mV to-31.83 mV (P<0.05). [Conclusion] Tagatose glycosylation can significantly improve the functional characteristics of EWP.

Then, the freezing curve, freezing time, and microstructures of apple samples are observed to analyze the effect of HPEFs on the freezing process and cell structure of apple samples. [Objective] To investigate the effects of structural changes in Cyperus esculentus starch on its apparent viscosity, water absorption index, and stability, providing theoretical support for its development and application in blended powders. [Methods] C.esculentus powder was modified by three methods, i.e., ultrafine grinding, microwave treatment, and enzymatic treatment. The effects of starch structure changes caused by these modifications on powder dispersibility were analyzed. [Results] Compared with native C.esculentus powder, enzymatic treatment reduced the relative crystallinity from 43.49% to 20.69%, causing the most significant starch structure disruption and notably improving dispersibility. [Objective] To address the limitation of spectral overlap interference in the rapid detection of 3, 4-dihydroxyphenylethanol (DHPET) within the 4-hydroxyphenethyl alcohol (HPET) catalytic system using traditional ultraviolet spectrophotometry. [Methods] To overcome the insufficient specificity of existing methods, a rapid spectrophotometric detection method based on Fe3+ coordination modulation was proposed. Through bidentate chelation between Fe3+ and the catechol group of DHPET, the characteristic absorption spectrum was induced to undergo a bathochromic shift to 760 nm, significantly distinguishing it from the weak coordination response of monophenolic compounds. [Objective] To analyze the preservation effect of ginger essential oil nanoemulsion as a natural preservative for pork. [Methods] Pork tenderloin was treated with 1.5% (w/w) ginger essential oil nanoemulsion. Changes in sensory, physicochemical, and microbiological indexes of the pork were analyzed during storage at 4 ℃.High-throughput sequencing technology was applied to analyze changes in the pork microflora during storage. [Results] The pH value, volatile basic nitrogen, and malondialdehyde content of pork treated with ginger essential oil nanoemulsion were significantly lower than those of the control group treated with sterile water at different time points (P<0.05), showing good preservation effects and extending the shelf life of pork tenderloin by 6 days. The nanoemulsion effectively reduced microbial abundance and diversity and inhibited spoilage bacteria such as Macrococcus and Brochothrix, thereby delaying pork spoilage. [Conclusion] Ginger essential oil nanoemulsion can serve as a natural preservative that effectively inhibits spoilage bacteria growth, reduces protein degradation and lipid oxidation in pork, improves sensory quality, and extends the shelf life of pork products.

Effect Of Ultrasonic Thawing On The Quality And Protein Oxidation Of Litopenaeus Vannamei, Peng Panpan

[Objective] To improve the texture quality of frozen Litopenaeus vannamei. [Methods] Using Litopenaeus vannamei as the research object, the study systematically evaluates the effects of ultrasonic thawing (300, 400, 500 W) on the texture properties, thawing loss, cooking loss, TVB-N, TBARS values, protein oxidation (content of sulphydryl and carbonyl groups), amino acid release, pH value, and sensory evaluation of frozen shrimp. [Results] Ultrasonic thawing significantly increases the elasticity, hardness, and recovery of the frozen shrimp (P<0.05), with the 400 and 500 W ultrasonic groups showing significantly higher hardness and elasticity than the 300 W group. Ultrasonic thawing significantly reduces thawing loss, cooking loss, and TVB-N content in the frozen shrimp (P<0.05), with the 500 W ultrasonic group showing the most significant reduction. Amino acid analysis reveals that ultrasonic thawing significantly increases the total amino acid (TAA) content and the proportion of essential amino acids (EAA), with the release of essential amino acids such as leucine and glutamic acid being highest in the 300 and 400 W ultrasonic groups. The protein oxidation analysis shows that the sulphydryl content in the ultrasonic-thawed group was significantly higher than that in the room-temperature thawed group, while the carbonyl content was significantly lower. Sensory evaluation results indicated that the overall score of the ultrasonic-thawed group is significantly higher than that |of the room-temperature thawed group, with the 400 W ultrasonic group achieving the highest sensory score. Pearson correlation analysis indicates that the thawing loss of frozen shrimp is negatively correlated with elasticity and hardness (P<0.05), and positively correlated with TVB-N, TBARS values, and carbonyl content (P<0.05). TVB-N is negatively correlated with hardness, elasticity, and recovery (P<0.05), and positively correlated with TBARS values and carbonyl content (P<0.05). [Conclusion] Ultrasonic thawing improves the texture quality of frozen shrimp compared to room-temperature thawing and effectively inhibits lipid and protein oxidation during the thawing process, with 400 W ultrasonic thawing being the most suitable.
