Early-stage breast cancers resistant to neoadjuvant therapy (NAT), characterized by high residual cancer burden (RCB) after treatment, have an increased risk of metastatic recurrence. Here, we show that circulating tumor DNA (ctDNA) detected using a tumor-informed test (1) can improve risk stratification of patients with NAT-resistant tumors (RCB-II/RCB-III) and (2) predict response to NAT. Stratification using ctDNA status at pretreatment or post-NAT and ctDNA dynamics identified NAT-resistant tumors with a significantly decreased risk of metastatic recurrence. ctDNA clearance as early as week 3 across receptor subtypes predicted favorable responses to NAT, including immunotherapies. The human brain contains a milieu of extracellular matrix (ECM) components that promote normal development and physiology. ECM signaling pathways are often dysregulated in brain pathologies including the malignant cancer glioblastoma (GBM). Here, we used single-cell spatial transcriptomic platforms to map the expression of nearly 400 ECM genes in matching non-cancerous brain and GBM samples. At least four different GBM cell populations have been identified that show unique ECM expression profiles and spatial enrichment in distinct intratumor regions. Spatial mapping demonstrates largely non-overlapping expression signatures of ECM components in GBM stromal cell types, particularly in vascular endothelial cells and microglia/macrophages. …

Sappanwood (Caesalpinia sappan L.) is a shrub or small tree that thrives in tropical regions and has been widely distributed across various areas. It has long been used as a natural dye in textiles, cosmetics, and herbal beverages due to its content of various bioactive compounds. This literature review discusses the secondary metabolites in sappanwood, their health benefits, and strategies to enhance product quality through pre-harvest and post-harvest treatments. This literature review was conducted by searching for relevant journals on Google Scholar using relevant keywords. This thesis develops a comprehensive framework for fair sequential resource allocation in multi-agent systems where a centralized allocator coordinates actions under global feasibility constraints, while satisfying preferences of different agents. Ranging from ridesharing platforms and homelessness intervention programs to power grid management, such systems play a critical role in shaping access to essential resources. Yet, existing approaches to resource allocation often prioritize aggregate utility, leading to systematic inequities across individuals and groups, particularly in sequential settings where decisions unfold over time. Straw mushrooms are among the most sought-after edible mushrooms, but their harvest is highly perishable due to their high respiration and transpiration rates, leading to a decline in quality during storage. Shallots are a major horticultural commodity with high economic value. The quality of shallots is determined not only by the quantity of the harvest but also by physical characteristics such as bulb color, texture, and water content. This study aims to evaluate the differences in these physical parameters in three popular local cultivars: Brebes, Sumenep, and Tuktuk, harvested at 56, 63, and 70 days after planting (DAP), respectively. Outcomes in classic Hodgkin lymphoma (cHL) have steadily improved; however, additional therapies are needed for patients who relapse or do not respond to novel agents. Here, we report the efficacy and safety of camidanlumab tesirine (Cami), an anti-CD25 antibody-drug conjugate, in patients with relapsed/refractory cHL after brentuximab vedotin/programmed cell death protein 1 inhibitor therapies from the phase 2 ADCT-301-201 study. Eligible patients were adults with cHL who had received ≥3 previous lines of systemic therapy (or ≥2 if ineligible for hematopoietic stem cell transplant). 98% of T cells reside in tissues, yet nearly all human T cell analyses are performed on peripheral blood. We performed single-cell sequencing of 5.7 million T cells from autologous blood and tonsils of ten donors. We identified distinct patterns of clonal expansion associated with tonsil-restricted phenotypes. Clonal sharing between blood and tonsils was lower than previous estimates and increased with age. Identical T cell receptor (TCR) sequences exhibited limited concordance in their phenotypes across compartments. Furthermore, location dictated the frequencies, clonal dominance, and phenotypes of antigen-specific T cells. Using immune organoids, we showed that antigen exposure drives functionally distinct …

Evidence is emerging that perturbed postnatal gut microbiota development is causally related to childhood undernutrition. In the phase 3 ECHELON-2 trial, brentuximab vedotin, cyclophosphamide, doxorubicin, and prednisone (BV-CHP) significantly improved progression-free survival (PFS) and overall survival (OS) compared with cyclophosphamide, doxorubicin, vincristine, and prednisone (CHOP) in patients with CD30+ peripheral T-cell lymphoma, benefits that were maintained at 5 years. Interim positron emission tomography (PET) scan can be used to assess prognosis and risk-stratify patients. The prognostic value of interim PET was assessed in this post hoc exploratory analysis from ECHELON-2, evaluating interim 18F-fluorodeoxyglucose PET scans after cycle 4 (PET4) and end-of-treatment-based response, and correlated with PFS per investigator and OS. Heart failure remains a pressing global health challenge, affecting approximately 64 million people and incurring annual healthcare costs of $108 billion, driven by high readmission rates and delayed interventions. Current risk prediction models predominantly rely on static machine learning approaches, utilizing fixed datasets that fail to adapt to the evolving nature of patient health. This limitation results in outdated risk assessments, hindering timely clinical responses and worsening patient outcomes and resource efficiencies. Kaffir lime (Citrus hystrix DC.) is a leafy spice commodity widely used as a culinary ingredient, in bio-pharmaceuticals, and as a raw material for essential oil production. Shoulder fatigue is a key contributor to performance decline and injury risk in swimmers, highlighting the need for objective monitoring tools; yet, despite their widespread use in land-based sports, wearable sensors remain underutilized in aquatic environments. PURPOSE: To examine how wearable muscle oxygenation (SmO2%) sensors can quantify local fatigue during performance, predict fatigue-induced kinematic breakdowns, and inform evidence-based training strategies. Postprandial hyperglycemia contributes to the development of chronic diseases such as diabetes mellitus. While post-meal physical activity (PA) is known to reduce blood glucose (BG), it remains unclear whether certain modes of PA are more effective than others when matched for energy expenditure. This study aims to compare the BG-attenuating effects of two energetically equivalent postprandial modes of PA.

Frailty and pre-frailty affect over half of patients with heart failure (HF) and are linked to less favorable outcomes, independent of age and New York Heart Association (NYHA) functional class. Pre-frailty, a transitional and potentially reversible state, is more prevalent than overt frailty but remains less studied. CASE HISTORY: A 21-year-old female with hypermobile Ehlers-Danlos Syndrome (hEDS) sustained a right ankle inversion injury while stepping off a curb, resulting in acute pain, swelling, and inability to weightbear. Initial conservative treatment including rest, ice, and traditional physical therapy (PT) was initiated but failed to relieve symptoms. Magnetic Resonance Imaging (MRI) revealed a Grade-II ATFL tear with chronic instability. After three months of unsuccessful conservative management and persistent pain and instability, surgical intervention was pursued.
