Transfected stem cells and T cells are promising in personalized cell therapy and immunotherapy against various diseases. However, existing transfection techniques face a fundamental trade-off between transfection efficiency and cell viability; achieving both simultaneously remains a substantial challenge. This study presents an acoustothermal transfection method that leverages acoustic and thermal effects on cells to enhance the permeability of both the cell membrane and nuclear envelope to achieve safe, efficient, and high-throughput transfection of primary T cells and stem cells. A rare earth metal, gadolinium has unique magnetic properties and the largest neutron absorption cross section making it highly sought after in emerging technology. However, unlike most metals, it is difficult to work with and lacks a damascene process. When incorporated into alloys, gadolinium’s properties are muted and the film thickness must increase to account for the low concentration. In recent years, significant attention has been directed toward evaluating the reactivity of diverse polymers to gauge their catalytic activity against hazardous chemicals. Among these polymers, pim-1 has emerged as an exceptional candidate due to its noteworthy attributes including high surface area, excellent solubility, and the capability to fine-tune the nitrile functional groups along its polymeric backbone. Building off this information, these ALC particles will now be tested with an actual lung cancer drug. In this project ionizable lipids were analyzed for their efficacy in delivery of mRNA in multiple metastatic lung cancer cells. Different ionizable lipids with their molar ratio in making the LNPs were analyzed in order to enhance the nucleic acid delivery to cytoplasm. Phospholipids also play a role in targeting efficacy of the LNPs to the lung cells. Different combination of ionizable lipids with multiple phospholipids were studied for physical characterization and in-vitro transcription efficiency. Transcription efficiency was then quantified using flow cytometry and optimal particles for transcription were noted.

Asphalt deterioration involves many physical and chemical changes caused by environmental factors such as automotive strain, extreme heat, and UV exposure. Asphalt binder assessment in the industry is currently based on structurally destructive testing methods and tests of physical performance without accounting for chemical structure. An emerging evaluation of asphalt binder uses relaxometry, a field of nuclear magnetic resonance (NMR) spectroscopy, to directly observe how asphalt binder is changed by causes of deterioration such as UV exposure. In 2022, ~288,000 American women were diagnosed with metastatic breast cancer which was the leading cause of patient mortality with ~43,000 American women succumbing to the disease. Women can be diagnosed with four different molecular subtypes of breast cancer: luminal A, luminal B, human epithelial growth receptor type 2 enriched (HER2+), and triple-negative breast cancer (TNBC). TNBC accounts for ~12% of all breast cancers, yet it has the worst outcomes. TNBC is highly invasive, with ~46% of TNBC patients developing metastatic disease. Chapter 2 discusses how to optimize cure-on-demand, thin layer coatings, using frontal polymerization. The purpose of this work is the chemical modification of cotton fabrics with natural globular protein. Modification was carried out in the process of finishing cotton fabrics, after decoction and bleaching in a continuous mode. The effect of modification on the strength, protein content and functional groups of cotton fabric has been studied. According to the results obtained using the AUTOGRAPH AGS-N machine, it was found that the modification of cotton fabrics with a solution of globular protein increases the strength of the treated fabrics, depending on the concentration of the modifier and the temperature regime.
