Unassisted, the lifetime of many proteins outside their natural environment is very short. Scientists generally need to extract the sample, often in remote locations, and then transport, isolate, and store such biospecimens relatively far from the point of origin in order to study them. Currently, the most commonly used methods for storage of biospecimens are cryopreservation or refrigeration which, though effective, have their fair share of flaws. Cryopreservation and refrigeration-based storage of biospecimens requires a great deal of space and other resources to be effective. Continued population growth and the decrease of existing energy platforms demands long-term solutions for development and implementation of scalable plasmonic metamaterials for energy and agricultural applications. Self-assembled nanoantenna into random and ordered arrangements are advanced herein for optical and thermal enhancements in scalable thin film. An analytical approach to estimating the thermal dynamics of random arrangements of nanoantenna resulted in estimates within 30% across a range of geometric parameters, nanoantenna-containing media, and thermal parameters. Cystic fibrosis (CF) is the most common genetic disease resulting in the morbidity and mortality of Caucasian children and adults worldwide. Due to a genetic mutation resulting in malfunction of the Cystic Fibrosis Transmembrane Conductance Regulator (CFTR) protein, CF patients produce highly viscous mucus in their respiratory tract. This leads to impairment of the mucociliary clearance of inhaled microbes. In addition to reduced microbial clearance, anoxic environmental conditions in the lungs promote biofilm-mode growth of the pathogenic bacterial species Pseudomonas aeruginosa. Chronic infections of P. aeruginosa begin in early childhood and typically persist until respiratory failure and death result. The …

Glycosaminoglycans (GAGs) Play Vital Roles in Many Biological Processes and Are Naturally Present as Complex Mixtures of Polysaccharides with Tremendous Structural Heterogeneity, Including Many Structural Isomers. Mass Spectrometric Analysis of GAG Isomers, in Particular Highly Sulfated Heparin (Hep) and Heparan Sulfate (HS), is Challenging Because of their Structural Similarity and Facile Sulfo Losses during Analysis. Mesoporous bioactive glasses (MBGs) are designed to have high specific surface area. They are formulated by a sol–gel process to formulate the glass followed by calcination. This study evaluates how calcination heating rate influences the porous architecture, and thereby the specific surface area, of MBGs. MBGs of molar ratio 80:15:5 for SiO2 :CaO:P2 O 5 were calcined using both low (1 °C/min) and high (20 °C/min) heating rates, termed as L-MBG and H-MBG, respectively. Pharmacological therapy of osteoporosis reduces bone loss and risk of fracture in patients. Modulation of bone mineral density cannot explain all effects. Other aspects of bone quality affecting fragility and ways to monitor them need to be better understood. Keratinous tissue acts as surrogate marker for bone protein deterioration caused by oestrogen deficiency in rats. Ovariectomised rats were treated with alendronate (ALN), parathyroid hormone (PTH) or estrogen (E2). MicroCT assessed macro structural changes. Raman spectroscopy assessed biochemical changes. Micro CT confirmed that all treatments prevented ovariectomy-induced macro structural bone loss in rats. PTH induced macro structural changes unrelated to ovariectomy. …

Immediate control of uncontrolled bleeding and infection are essential for saving lives in both combat and civilian arenas. Inorganic well-ordered mesoporous silica and bioactive glasses have recently shown great promise for accelerating hemostasis and infection control. However, to date, there has been no comprehensive report assessing their specific mechanism of action in accelerating the hemostasis process and exerting an antibacterial effect. The House Observations of Microbial and Environmental Chemistry (HOMEChem) study is a collaborative field investigation designed to probe how everyday activities influence the emissions, chemical transformations and removal of trace gases and particles in indoor air. Sequential and layered experiments in a research house included cooking, cleaning, variable occupancy, and window-opening. This paper describes the overall design of HOMEChem and presents preliminary case studies investigating the concentrations of reactive trace gases, aerosol particles, and surface films. Gamma-Secretase (γ-secretase) is a transmembrane protease of increasing interest, which has been shown to have significant connections to both cancer and Alzheimer’s disease. γ-secretase cleaves both Notch-1, a transmembrane signaling protein, and Amyloid precursor protein (APP), a transmembrane protein whose cleavage may result in the formation of β-amyloid plaques in the brain. Notch-1 and APP are widely studied proteins that have substantial impacts on the development and proliferation of cancer and Alzheimer’s disease, respectively. Di-2-ethylhexyl phthalate (DEHP) is a plasticizer used in consumer products and building materials, including polyvinyl chloride flooring material. DEHP adsorbs from material and leaches into soil, water, or dust and presents an exposure risk to building occupants by inhalation, ingestion, or absorption. A number of bacterial isolates are demonstrated to degrade DEHP in culture, but bacteria may be susceptible to it as well, thus this study examined the relation of DEHP to bacterial communities in dust. The tested hypothesis was that features on polydimethylsiloxane (PDMS) surfaces coated with a poly(N-isopropylacrylamide)/aminopropyltriethoxysilane or pNIPAAM/APTES thin film would accelerate cell detachment than the film coated on a unfeatured surface. This project tested samples with features generated by molds, wrinkling, and sandpaper roughened substrates. Surface feature generation methods were limited to mechanical means, and characterized by microscopy and strain rates. 50/50 mixtures of 1.5 wt.% pNIPAAM/ APTES were used to coat thin films (30-40 nm) on PDMS membranes by spin-coating, and the coated membranes were thermally annealed to chemically graft pNIPAAm/APTES on the membrane and their thermo-responsive property was assessed …

Using contact angle to determine surface morphology and chronoamperometry testing to identify ideal sensor conditions, tests optimized parameters like weight percent of nylon or other polymers, carbon nanotube (CNT) isomer, and solution concentration to determine reproducibility of functional sensors. The main objective of this honor’s project is to encapsulate drugs with different molecular weights inside of chitosan-heparin based nanoparticles (Cs-Hep NPs). Three different drugs with low (NNC), medium (IFN-γ), and large (BSA) molecular weights were chosen in order to compare all types of drugs and how they react with the nanoparticles. Analysis of particle size, zeta potential, and a release study was performed. The release took place over a 168-hour time period and liquid samples were taken throughout. Due to the demand for in-situ electrolyte sodium sensors, prototype sensors have been produced to create a product for athletes and marathoners to monitor hydration levels. These senors address a desire in the marketplace for athletic monitoring for overall athlete wellness and performance. Sweat sensor samples were created using Multi-walled Carbon Nanotubes functionalized on a Nylon-6 fiber mat using cyclo-oligomeric Calixarene. This production process produces sensors capable of detecting sodium ion concentration present in human sweat in presence of an electric current. It was hypothesized that through the use of Hofmeister Ions, the lower critical solution temperature (LCST) of poly(vinyl methyl ether) (PVME) and poly(ethylene glycol)- poly(N-isopropylacrylamide) (PEG-b-pNIPAAm) block-copolymer could be controlled. Through literature searches and small lab experiments, our team found that there may be a connection between Hofmeister effects and phase transition of a thermo-responsive polymer.
