There are no controlled studies of acetaminophen toxicity in horses. The objective was to test the hypotheses that oral acetaminophen administered at a dosage 25% higher than that sometimes used in horses would result in measurable hepatic toxicity as seen in humans and other species. Six healthy adult horses were administered 25 mg/kg acetaminophen powder in corn syrup twice daily for 30 days. Three other horses served as negative controls receiving only corn syrup. Alfaxalone is a neuroactive synthetic steroid (Brewster & Bodor, 1990) that produces anesthetic induction with dose- and speed-dependent cardiorespiratory depression in dogs and cats (Chiu et al. 2016; Warne et al., 2015). At clinical doses in unpremedicated dogs (2 mg kg^-1) and cats (5 mg kg^-1), alfaxalone induces a mild decrease in systemic vascular resistance, systemic blood pressure (Muir et al., 2009; Muir et al., 2008), apnea, hypoventilation, and hypoxemia (Muir et al. 2009; Muir et al., 2008). Research has recently demonstrated that larval zebrafish show similar molecular responses to nociception to those of adults. Our study explored whether unprotected larval zebrafish exhibited altered behaviour after exposure to noxious chemicals and screened a range of analgesic drugs to determine their efficacy to reduce these responses. This approach aimed to validate larval zebrafish as a reliable replacement for adults as well as providing a high-throughput means of analysing behavioural responses. Anaesthesia causes temporary absence of consciousness and may reduce the stress and/or pain associated with handling and certain invasive procedures. The rainbow trout (Oncorhynchus mykiss) is a widely-used model species with relevance to both aquaculture and natural ecosystems. This study sought to establish the relative acute impact of commonly used anaesthetics on rainbow trout when used for anaesthesia or euthanasia by exploring their effects on aversion behaviour and stress physiology. Kentucky 31 tall fescue (KY31) infected with the common toxic endophyte strains of Epichloё coenophiala produces toxic alkaloids that improve plant vigor, but cause numerous adverse effects in grazing animals. Researchers developed a variety of KY31 containing an alternative strain of E. coenophiala, termed novel endophyte (NE). Adverse health effects in mares have not been evaluated.

Second-hand smoke (SHS) has been associated with respiratory cancers in canines, with the predisposed locations determined by the cephalic ratio (head length / head width). Exposure to SHS is frequently measured by analyzing urine or blood samples for the nicotine by-product cotinine. We investigated the suitability of saliva as an alternative source for cotinine analyses in dogs because obtaining saliva is less invasive and cotinine concentrations from SHS exposure are instantaneously measurable in saliva compared to other collection methods. Endocrine-disrupting chemicals (EDCs), such as those from plastics and pesticides, have been hypothesized to affect wildlife populations. According to the U.S. National Institute of Environmental Health Sciences, EDCs ‘are chemicals that may interfere with the body’s endocrine system and produce adverse developmental, reproductive, neurological, and immune effects in both humans and wildlife.’ A growing number of studies suggest wildlife are being exposed to EDCs, but how EDCs affect wildlife health and wildlife conservation is less well-understood. Reversible Downregulation Of Hypothalamic-Pituitary-Gonadal Axis In The Stallion With A Third-Generation Gnrh Antagonist, Gabriel Monteiro Davolli

The objectives of this thesis were: (1) to evaluate the downregulation of the stallion hypothalamic-pituitary-gonadal (HPG) axis by a GnRH antagonist (acyline) based upon endocrine, seminal, testicular and behavioral effects, and (2) to assess recovery after treatment. Stallions were treated for 50 days (n=4; 330µg/kg acyline q 5d) and controls (n=4) received vehicle alone. Stallions were assessed pre-treatment and for 72 days after last treatment. Treatment induced declines (p<0.05) in FSH, LH, testosterone (to castrate levels) and estrone sulfate. Gonadotropins and testosterone returned to control values within nine days and estrone sulfate by 14 days after treatment discontinuation. Acyline-treated stallions failed to respond with FSH, LH and testosterone increase after exogenous GnRH stimulation (25µg gonadorelin, IV) compared to pre-treatment and control stimulation. Total sperm numbers and motility were reduced in acyline-treated stallions, as well as total seminal plasma protein and testicular volume (p<0.05). Time to ejaculation was increased in acyline group (p<0.5). Testicular, sexual behavior and most seminal parameters regained normal levels within 72 days after treatment ceased. Sperm output of acyline-treated stallions was regained within seven months after ending treatment. Acyline reversibly suppressed the stallion HPG axis, thus has potential for treating the androgen-dependent Equine-Arteritis-Virus carrier state and as behavior modulator.

Intra-articular glucocorticoid injections are commonly used to treat synovitis and osteoarthritis in horses. These agents are highly effective at relieving pain, swelling, and other symptoms of joint inflammation. The drugs also have therapeutic benefits by down regulating the expression of cytokines and protease enzymes that participate in the degradation of articular cartilage. However, detrimental effects on chondrocyte function and cell viability that is independent of osteoarthritis pathogenesis have been described and linked to glucocorticoid use. Many antibiotics and medications used in the treatment of animals have a withdrawal time; residues are a concern for the meat industry. The most recently published 2009 USDA-FSIS Residue Program Data Report listed 135,389 Inspector-in-Charge-Generated (IICG) residue samples from 43,142,500 beef and dairy cattle inspected that year at harvest. Of these samples, 1,306 contained violative antibiotic residues and 327 violative flunixin meglumine residues. Two classes of antimicrobials comprised over half of the documented violative antimicrobial residues (beta-lactams that include ceftiofur and sulfa drugs that include sulfadimethoxine). While the violative residue rate seems small, violative residues are unacceptable. Management of carcasses …

Equine protozoal myeloencephalitis is the most important infectious neurologic disease of horses in the Western Hemisphere. Equine protozoal myeloencephalitis can interfere with a horse's ability to race, work, and perform; untreated, EPM can be lethal. Antemortem diagnosis of EPM is challenging, requiring careful evaluation of the animal's history, clinical signs, and laboratory data, with rigorous exclusion of other causes.

Therapeutic approaches to EPM are evolving. The aim of this study was to describe cases of epistaxis associated with post-race Exercise Induced Pulmonary Hemorrhage (EIPH) in Thoroughbred horses at the Hippodrome "La Rinconada", Caracas, Venezuela, through a clinic pathological study. We studied a total of 29 cases of epistaxis post-race in Thoroughbred horses at the Hippodrome La Rinconada, Caracas, Venezuela, which is 2,950 meters above sea level. The study included horses between the ages of 2-5 years, 16 stallions and 13 mares, weighing between 450-510 kg. There has been a need for the assessment of ground water contamination risk on terrestrial organisms. One terrestrial organism, Eisenia fetida, was chosen as a test species and exposed to three common pharmaceutical and/or pesticide related contaminants: acetaminophen (CAS#103-90-2), caffeine (CAS#58-08-2) and carbendazim (CAS#10605-21-7), respectively. Levels of contamination varied from grams per liter to micrograms per liter (ppb) using distilled water as the solvent. Eisenia fetida was examined through a 28 day range/index toxicity test. A Benchmark Dose (BMD) dichotomous probit analysis, using a 95% confidence interval, was calculated for all three contaminants. All three contaminants were found to have …

Recent research has shown the possibility of pain perception in fish; therefore, the use of analgesia or “painkillers” should be considered for invasive procedures. However, there is relatively little information on the effectiveness of analgesic drugs nor on the appropriate dose for fish. This study assessed the efficacy of three types of drug: an opioid, buprenorphine, a non-steroidal anti-inflammatory drug (NSAID), carprofen and a local anaesthetic, lidocaine. Bovine viral diarrhea (BVD) is an economically important disease of cattle. Calves persistently infected (PI) with the bovine viral diarrhea virus (BVDV) are a powerful agent for spread of the virus. A total of 24,423 southeast origin beef cattle comingled at three Warren County, Kentucky locations were tested from November 2007 to June 2010 for PI BVDV. A total of 97 head tested positive for PI BVDV, giving an average overall prevalence of 0.397%.

The pathogenesis of hepatotoxic and hepatocarcinogenic actions of the mycotoxin aflatoxin B1 (AFB1) involves initial bioactivation by microsomal cytochrome P450s (P450) to a reactive and electrophilic intermediate, exo-aflatoxin B1-8,9-epoxide (exo-AFBO). Poultry, especially turkeys, are extremely sensitive to AFB1, a condition associated with efficient epoxidation by P450s. The purpose of this research was to 1) discover and characterize the P450s in turkey liver responsible for AFB1 bioactivation, and 2) determine the relative importance of these P450s in turkey liver.

Newby and Stevens’ (2008) paper ‘‘The effects of the acetic acid ‘pain’ test on feeding, swimming, and respiratory responses of rainbow trout (Oncorhynchus mykiss)’’ examines the effects of a noxious stimulus on the behaviour of trout in an attempt to replicate research conducted in my laboratory (Sneddon, 2003a; Sneddon, 2007; Sneddon et al., 2003a,b; Reilly et al., 2008). Last spring, North America was gripped in the largest pet food recall in history. News outlets reported tens of thousands of dogs and cats becoming ill, and many dying, as a result of ingesting contaminated pet food. Several pet owners have filed lawsuits against the manufacturers and distributors of the pet food products. I think this is an excellent real-life science story to use with students to show science investigation in action, and to discuss the intersections between the role of regulatory agencies and the public.
