Current Therapeutic Approaches To Equine Protozoal Myeloencephalitis, L. Dirikolu, Jonathan H. Foreman, Thomas Tobin

Maxwell H. Gluck Equine Research Center Faculty Publications

Equine protozoal myeloencephalitis is the most important infectious neurologic disease of horses in the Western Hemisphere. Equine protozoal myeloencephalitis can interfere with a horse's ability to race, work, and perform; untreated, EPM can be lethal. Antemortem diagnosis of EPM is challenging, requiring careful evaluation of the animal's history, clinical signs, and laboratory data, with rigorous exclusion of other causes.

Therapeutic approaches to EPM are evolving. First-generation therapeutic approaches for EPM were based on the classic anti–Toxoplasma gondii pyrimethamine–sulfonamide combinations; treatment is prolonged and can be associated with a considerable relapse rate, which may be associated with the difficulty in …

Clinical Anesthesia And Analgesia In Fish, Lynne U. Sneddon Jan 2012

Clinical Anesthesia And Analgesia In Fish, Lynne U. Sneddon

Veterinary Science and Medicine Collection

Fish have become a popular experimental model and companion animal, and are also farmed and caught for food. Thus, surgical and invasive procedures in this animal group are common, and this review will focus on the anesthesia and analgesia of fish. A variety of anesthetic agents are commonly applied to fish via immersion. Correct dosing can result in effective anesthesia for acute procedures as well as loss of consciousness for surgical interventions. Dose and anesthetic agent vary between species of fish and are further confounded by a variety of physiological parameters (e.g., body weight, physiological stress) as well as environmental …

The Toxicity Of Acetaminophen, Caffeine And Carbendazim In Earthworms (Eisenia Fetida), Matthew M. Bulman

Masters Theses

There has been a need for the assessment of ground water contamination risk on terrestrial organisms. One terrestrial organism, Eisenia fetida, was chosen as a test species and exposed to three common pharmaceutical and/or pesticide related contaminants: acetaminophen (CAS#103-90-2), caffeine (CAS#58-08-2) and carbendazim (CAS#10605-21-7), respectively. Levels of contamination varied from grams per liter to micrograms per liter (ppb) using distilled water as the solvent. Eisenia fetida was examined through a 28 day range/index toxicity test. A Benchmark Dose (BMD) dichotomous probit analysis, using a 95% confidence interval, was calculated for all three contaminants. All three contaminants were found to have …

The Efficacy Of Three Types Of Analgesic Drugs In Reducing Pain In The Rainbow Trout, Oncorhynchus Mykiss, Jessica J. Mettam, Lois J. Oulton, Catherine R. Mccrohan, Lynne U. Sneddon Sep 2011

The Efficacy Of Three Types Of Analgesic Drugs In Reducing Pain In The Rainbow Trout, Oncorhynchus Mykiss, Jessica J. Mettam, Lois J. Oulton, Catherine R. Mccrohan, Lynne U. Sneddon

Veterinary Science and Medicine Collection

Recent research has shown the possibility of pain perception in fish; therefore, the use of analgesia or “painkillers” should be considered for invasive procedures. However, there is relatively little information on the effectiveness of analgesic drugs nor on the appropriate dose for fish. This study assessed the efficacy of three types of drug: an opioid, buprenorphine, a non-steroidal anti-inflammatory drug (NSAID), carprofen and a local anaesthetic, lidocaine. Each drug was tested at three doses on rainbow trout that were noxiously stimulated and the most effective dose was also given to fish experiencing no pain to investigate side-effects. Ventilation rate and …

Mechanisms Of The Extreme Sensitivity Of Turkeys To Aflatoxin B1, Sumit Rawal May 2010

Mechanisms Of The Extreme Sensitivity Of Turkeys To Aflatoxin B1, Sumit Rawal

All Graduate Theses and Dissertations, Spring 1920 to Summer 2023

The pathogenesis of hepatotoxic and hepatocarcinogenic actions of the mycotoxin aflatoxin B1 (AFB1) involves initial bioactivation by microsomal cytochrome P450s (P450) to a reactive and electrophilic intermediate, exo-aflatoxin B1-8,9-epoxide (exo-AFBO). Poultry, especially turkeys, are extremely sensitive to AFB1, a condition associated with efficient epoxidation by P450s. The purpose of this research was to 1) discover and characterize the P450s in turkey liver responsible for AFB1 bioactivation, and 2) determine the relative importance of these P450s in turkey liver.

Initial investigations led to the discovery of CYP1A5. We then identified CYP3A37, a human CYP3A4 homologue from turkey liver, …

The Effects Of The Acetic Acid “Pain” Test On Feeding, Swimming, And Respiratory Responses Of Rainbow Trout (Oncorhynchus Mykiss): A Critique On Newby And Stevens (2008), Lynne U. Sneddon

Experimentation Collection

Newby and Stevens’ (2008) paper ‘‘The effects of the acetic acid ‘pain’ test on feeding, swimming, and respiratory responses of rainbow trout (Oncorhynchus mykiss)’’ examines the effects of a noxious stimulus on the behaviour of trout in an attempt to replicate research conducted in my laboratory (Sneddon, 2003a; Sneddon, 2007; Sneddon et al., 2003a,b; Reilly et al., 2008). However, the authors used a different protocol to the one already published and I would like to respond to some of their points of discussion to provide scientific explanations for their results using data from my laboratory. The authors show that swimming …

Medicina Veterinaria

The Pet Food Recall Puzzle: Who, What, Why, And How Much, Roxanne Greitz Miller

Education Faculty Articles and Research

Last spring, North America was gripped in the largest pet food recall in history. News outlets reported tens of thousands of dogs and cats becoming ill, and many dying, as a result of ingesting contaminated pet food. Several pet owners have filed lawsuits against the manufacturers and distributors of the pet food products. I think this is an excellent real-life science story to use with students to show science investigation in action, and to discuss the intersections between the role of regulatory agencies and the public.

In order to understand the case, you first have to know the facts. We'll …

The Evidence For Pain In Fish: The Use Of Morphine As An Analgesic, Lynne U. Sneddon Sep 2003

The Evidence For Pain In Fish: The Use Of Morphine As An Analgesic, Lynne U. Sneddon

Veterinary Science and Medicine Collection

This paper discusses the evidence for pain perception in fish and presents new data on morphine analgesia in fish. Recent anatomical and electrophysiological studies have demonstrated that fish are capable of nociception, the simple detection of a noxious, potentially painful stimulus and the reflex response to this. To prove pain perception, it must be demonstrated that an animal’s behaviour is adversely affected by a potentially painful event and this must not be a reflex response. The present study examined the acute effects of administering a noxious chemical to the lips of rainbow trout (Oncorhynchus mykiss) to assess what changes occurred …

An Overview Of The Methylxanthines And Their Regulation In The Horse, J Daniel Harkins, W. Allen Rees, G. D. Mundy, Scott D. Stanley, Thomas Tobin

Maxwell H. Gluck Equine Research Center Faculty Publications

Caffiene, theophylline and theobromine are naturally occurring members of the methylxanthine family;pentoxfylline, dyphylline and enprofylline are structurally related synthetic pharmaceuticals. Caffiene has predominantly central nervous system effects, theophylline, dyphylline and enprofylline have predominantly bronchodilator effects, while theobromine is associated with diuretic responses. Pentoxfylline is thought to increase red cell deformability and facillitate blood flow through capillary beds. The methylxanthines are not highly potent agents; they are typically administered in gram doses and they tend to have relatively long half-lives. They remain detectable in plasma and urine for relatively long periods. Similarly, traces of the naturally occurring members of this family …

Isoxsuprine is reported to be a peripheral vasodilator used in human and veterinary medicine to treat ischaemic vascular disease. In horses, it is generally administered orally to treat navicular disease and other lower limb problems. To deflne the scope and duration of its pharmacological responses after oral administration, 6 horses were dosed with isoxsuprine HCI (1.2 mg/kg bwt) q. 12 h for 8 days and then tested to assess the duration and extent of pharmacological actions. There was no significant difference between isoxsuprine and control treatment values for heart rate, spontaneous activity, sweat production, anal muscle tone, core and skin …

The Pharmacologic Effects Of Isoxsuprine, J. Daniel Harkins, Thomas Tobin

Maxwell H. Gluck Equine Research Center Faculty Publications

Isoxsuprine is a therapeutic medication used to treat navicular disease and other lower limb problems in horses and is one of the more frequently detected therapeutic agents in racing horses. In crossover studies, horses were administered intravenous and oral isoxsuprine to determine the character and duration of pharmacological effects. Following intravenous administration, isoxsuprine significantly increased heart rate, spontaneous activity, and sweat production. There was an apparent, although statistically insignificant, increase in cutaneous blood flow. Skin temperature decreased below control values, and there was a significant decrease in core temperature. Isoxsuprine also reduced smooth muscle tone. In contrast, after oral dosing, …
