Toxicological Effects And Treatment Of Accidental Ingestion Of Simparica Trio® In Felis Catus: An Investigative Review, Aussia M. Stander

Simparica TRIO®, a canine endectocide manufactured by Zoetis, contains sarolaner, moxidectin, and pyrantel pamoate salt in an orally administered chewable tablet. These compounds are all toxic to both cats and dogs in sufficient amounts. As Simparica TRIO® rises in popularity among pet owners, Felis catus (felines, cats) in multi-pet households (households with both cats and dogs) are at increased risk of overdosage and toxicosis from accidental ingestion of Simparica TRIO® due to their smaller weight.

Current testing methodologies within equine forensic toxicology focus on arbitrary thresholds and zero-tolerance policy. Modern analytical chemistry’s limits of detection are low enough that oftentimes femtogram-per-milliliter amounts of a substance can readily be identified in both blood and urine of a horse. For most pharmacologically relevant compounds, these concentrations have no relevance to pharmacological effect. It is therefore crucial that testing methodologies to determine appropriate thresholds and cut-offs be developed that are driven by biological activity rather than arbitrary limits of detection. Kisspeptin is a neuropeptide that governs the reproductive axis upstream to GnRH. We wanted to study whether kisspeptin modulates plasma LH and FSH levels and ovarian follicular dynamics in buffaloes and whether kisspeptin can be used for fixed time artificial insemination (FTAI). We carried out these studies in comparison with buserelin, a potent GnRH agonist. Kisspeptin dose-dependently increased plasma LH levels. However, the kisspeptin-induced increase in LH was short-lived as the peak reached in 15–30 min returned to basal values by 1–2 h. There are no controlled studies of acetaminophen toxicity in horses. The objective was to test the hypotheses that oral acetaminophen administered at a dosage 25% higher than that sometimes used in horses would result in measurable hepatic toxicity as seen in humans and other species. Six healthy adult horses were administered 25 mg/kg acetaminophen powder in corn syrup twice daily for 30 days. Three other horses served as negative controls receiving only corn syrup. Many antibiotics and medications used in the treatment of animals have a withdrawal time; residues are a concern for the meat industry. The most recently published 2009 USDA-FSIS Residue Program Data Report listed 135,389 Inspector-in-Charge-Generated (IICG) residue samples from 43,142,500 beef and dairy cattle inspected that year at harvest. Of these samples, 1,306 contained violative antibiotic residues and 327 violative flunixin meglumine residues. Two classes of antimicrobials comprised over half of the documented violative antimicrobial residues (beta-lactams that include ceftiofur and sulfa drugs that include sulfadimethoxine). It has long been recognised that a means of killing rabbits selectively in the presence of stock would be a major advance in rabbit control. It would enable a whole farm to be treated for rabbit control at one time, saving operational costs and cutting down reinfestation of rabbits into treated areas from untreated areas.

A literature search and some pilot trials indicated that indandione derivatives were more toxic to rabbits than to ruminants.

Pindone (2- pivalyl-l,3-indandione) was selected for evaluation.
