The SARS-Coronavirus (SARS-CoV) is the etiological agent of the severe acute respiratory syndrome (SARS). The SARS-CoV spike (S) glycoprotein mediates membrane fusion events during virus entry and virus-induced cell-to-cell fusion. Investigations, described herein, have focused on the genetic manipulation of the SARS-CoV S glycoprotein in order to delineate functional domains within the protein. This was accomplished by incorporating single point mutations, cluster-to-lysine and cluster-to-alanine mutations, as well as carboxyl terminal truncations into the protein and investigating these mutants in transient expression experiments. An Edwardsiella ictaluri strain with disruption of ureG was identified through the use of signature tagged insertion mutagenesis as being attenuated for virulence in the channel catfish host. Sequencing of the flanking regions surrounding the insert showed that the gene was part of a urease gene complex that included ureE, ureF, ureG, ureD, ureI, and an ammonium transporter homologue. The ureG gene encodes a GTP-binding accessory protein which is thought to function in energy-dependent urease assembly. The ureG mutant strain was found to be attenuated for mortality, persistence, and for the ability to establish infection in a competition challenge during …

Vaccines for pathogenic Strains of bovine herpesvirus 1 (BHV-1) which are based on attenuated BHV-1 having a mutation in the latency-related gene are provided. Live, attenuated vaccines are also provided which express anti gens from other viral or bacterial pathogens and thus form the basis of a variety of vaccines.

The phosphoprotein (P) of vesicular stomatitis virus (VSV) is an essential subunit of the viral RNAdependent RNA polymerase (RdRp) complex. It is phosphorylated at two different domains. Using defective interfering (DI) RNA or minigenomic RNA templates, we previously demonstrated that phosphorylation within the amino-terminal domain I is essential for transcription, whereas phosphorylation within the carboxyterminal domain II is necessary for replication. For the present study, we examined the role of the phosphorylation of residues in these domains in the life cycle of VSV. During the course of their disease, leprosy patients may experience two types of inflammatory reactions- erythema nodosum leprosum (ENL) or reversal reaction (RR). Thalidomide is effective treatment for ENL, but not for RR. Using concentrations of thalidomide similar to that achieved in the treatment of ENL, we investigated thalidomide’s effect on reactions, viability of M. leprae, and integrity of plasma membranes. Columnaris disease, caused by the bacterium Flavobacterium columnare, is an economically significant problem in many warmwater fish species. Difficulties encountered in the isolation and culture of F. columnare have been an impediment to research on the organism and the disease it causes. The goal of this study was to improve the methods for isolation, culture, identification and maintenance of F. columnare. Brucellosis and pseudorabies lead to abortion in pregnant sows and are perpetuated by feral swine reservoirs. A multivalent oral vaccine for these diseases would improve vaccination and eradication programs worldwide. Previous studies have shown that the rough attenuated Brucella strains RB51 and VTRS1, when administered subcutaneously to swine, stimulate host immune responses, transiently colonize tissues, and provide partial protection against virulent B. suis infection in pregnant sows. The leading viral killer of commercially produced channel catfish is Channel Catfish Virus (CCV). Studies conducted to evaluate DNA vaccination against CCV compared encoded gene, dose, multiple DNA vaccines, and immune response to vaccination. Genes were selected (ORFs 1 and 3 [immediate early genes], ORFs 6, 19, and 46 [membrane genes], and ORF59 [putative major envelope glycoprotein gene], cloned into a plasmid, and expressed in mammalian and fish cell culture to detect predicted molecular weight proteins. The very potent antibacterial activity of cecropin B makes it a likely candidate to prevent and/or treat Mannheimia haemolytica 1:A infection in the upper respiratory tract (URT) of cattle. Validation of diagnostic assay performance is hampered where no gold standard exists. Bayesian estimates of the sensitivity and specificity were applied to studies of two persistent bovine infections: bovine immunodeficiency virus (BIV), and bovine viral diarrhea virus (BVDV). BVDV pathogenesis is well described. Losses of $20 million in 1992 alone led the Danish government to mandate eradication. Two enzyme-linked immunosorbent assay (ELISA) tests were developed; one antibody-based to detect exposure, one antigen-based to detect persistent infection. Bayesian estimates of the sensitivity and specificity were 96.6% and 99.2%, and 98.2% and 99.8% for the antibody and antigen ELISA, respectively. Maximum Likelihood …

The host-parasite interaction during early filarial nematode migration is poorly understood. The objective of this study was to develop a model of early cutaneous filarid migration using Brugia pahangi in the jird (gerbil) host and measure the histologic and cytokine responses during this period. Male gerbils were intradermally inoculated in the left hindlimb with 100 B. pahangi L3 then necropsied at 3 hours, 24 hours, 3 days, 7 days, and 28 days post-infection. Larvae were recovered and tissues collected for histology and cytokine measurement. At 3 hours, most larvae (96.3%) were recovered from tissues associated with the infection site. Migration …

The overall goals of this research were to develop a reproducible method of detecting stable DNA insertion into Japanese quail and provide a method for gene location on avian chromosomes. The Herpes Simplex Virus life cycle contains a number of membrane fusion events that must function properly to ensure a productive infection, including: virus attachment and entry into susceptible cells, de-envelopment at the outer nuclear lamellae, and virus-induced cell-to-cell fusion. A virus-free cell fusion assay was recently developed in order to attempt to understand the underlying mechanisms that are responsible for viral fusion events and was utilized in order to investigate the effect of mutations targeted to the carboxyl terminus of gB. The first event in the infection process of Ostertagia ostertagi in cattle is the process of exsheathment. Before trichostrongylid nematodes can transition from a free-living infective stage larva (L3) on pasture to a parasitic existence within the ruminant host, it must first undergo exsheathment. Exsheathment is the process whereby the L2 cuticle retained from the previous molt is cast from the L3. Exsheathment enables the developmental transition from a free-living stage on pasture to a parasitic existence in the bovine host. The ability to regulate cell volume is an ancient conserved trait present in essentially all species through evolution. The maintenance of a constant cell volume is a homeostatic imperative in animal cells. Changes in cell water content affecting the concentration of intracellular messenger molecules impair the complex signaling network, crucial for cell functioning and intercellular communication. Although the renal homeostatic mechanisms exert a precise control of extracellular fluid osmolarity, this is challenged in a variety of pathological situations. To study the protective responses of cyathostome-infected ponies, two challenges were performed employing animals with different histories of exposure to these parasites. The hypothesis developed and to be tested in these experiments was that ponies that had longer exposure to cyathostome contaminated pastures would express acquired resistance to infection. The assumption behind this hypothesis was that helminth-naïve ponies infected with cyathostomes would eliminate the infection using only innate immune responses. Whereas previously exposed ponies would eliminate the infection with acquired immune responses, and these would be more effective in ponies with longer exposure to cyathostomes. Inflammation is a component of the innate immune response. However, severe or prolonged inflammation can be detrimental. Dietary n-3 fatty acid supplementation has proven clinical benefits in chronic inflammatory diseases, most likely due to reduced synthesis of inflammatory mediators and inhibition of lymphocyte proliferation and function. The purposes of this study were to characterize alterations in inflammatory mediator production, and lymphocyte proliferation, in dogs fed a diet consisting of 0.65% n-3 fatty acids (DMB) with an n-6:n-3 fatty acid ratio of 3.4:1. Among the gastrointestinal nematode parasites that cause the most problems to small ruminants, Haemonchus contortus is one of major concern. Currently, the control of H. contortus and others is almost entirely based on the use of anthelmintics. Consequently, anthelmintic resistance has developed worldwide and this has become a serious problem in small ruminant nematode parasite control programs. Helicobacter hepaticus is well established as an unwanted variable in laboratory rodent colonies. Historically, caesarean section and embryo transfer have been used to derive Helicobacter-free mouse colonies. Neonatal transfer of newborn mice onto Helicobacter-free foster dams was recently reported as an alternative method of deriving Helicobacter-free mice, but until now, the age by which pups must be fostered to remain Helicobacter-free was unknown. The current pathogenesis of Sudden Infant Death Syndrome (SIDS) is unknown. Elevated endogenous ethanol production by gastrointestinal yeast has been proposed as a possible mechanism for SIDS. To investigate the role of ethanol in SIDS, three studies were undertaken. In the first study, transcutaneous blood gas technology was evaluated for repeated monitoring of arterial blood gases. The second study identified the cardiorespiratory responses to inhaled CO2 challenge in juvenile rats, to determine the age corresponding to the age range of peak incidence of SIDS in human infants. Swelling subsequent to hyposmotic conditions activates a process of volume regulation present in most cell types. This volume adjustment is accomplished by osmolyte translocation toward the extracellular space to reach a new osmotic equilibrium. Molecules involved in this homeostatic mechanism have been broadly classified into two categories: organic and inorganic osmolytes. Inorganic osmolytes comprise mainly the intracellular ions K+ and Cl–. Cell swelling–induced activation of separate K+ and Cl– channels has been described in most preparations. The cytochrome P450 (CYP) 2J2 arachidonic acid epoxygenase gene was down-regulated at a pre-translational level in human hepatoma-derived HepG2 cells incubated in a hypoxic environment; under these conditions, the expression of c-Jun and c-Fos mRNA and protein was increased. The 5′-upstream region of the CYP2J2 gene was isolated by amplification of a 2341 bp fragment and putative regulatory elements that resembled activator protein-1 (AP-1)-like sequences were identified. From transient transfection analysis, c-Jun was found to strongly activate a CYP2J2–luciferase reporter construct, but co-transfection with plasmids encoding c-Fos or c-Fos-related antigens, Fra-1 and -2, abrogated reporter activity. Nearly all human immunodeficiency virus (HIV) infections are acquired mucosally, and the gut-associated lymphoid tissues are important sites for early virus replication. Thus, vaccine strategies designed to prime virus-specific cytotoxic T lymphocyte (CTL) responses that home to mucosal compartments may be particularly effective at preventing or containing HIV infection. The Salmonella type III secretion system has been shown to be an effective approach for stimulating mucosal CTL responses in mice. The present invention provides a method to detect the presence of a microorganism or agent in an animal. The method encompasses placement of devices at various locations where the animal resides so as to induce the animal to initiate contact with the device. As a result of this contact, the animal deposits various microorganisms and agents on the device. The device is then tested for the presence of the particular microorganism or agent of interest.

Complete sequencing and annotation of the M. leprae genome has provided new information related to proteins constituting its hypothetical proteome. Since M. leprae can not be grown in vitro, novel approaches are needed to determine which proteins are expressed during infection and whether these proteins are related to pathogenesis. Secreted proteins represent a distinct group of protein with respect to their structure and function, contribution to virulence and are of particular importance for vaccine development because they are often immunogenic and have the potential to be recognized early in infection. It is important to be able to predict the potential spread of water borne diseases when building dams or redirecting rivers. This study was designed to test whether the use of a growing degree day (GDD) climate model and remotely sensed data (RS) within a geographic information system (GIS), could be used to predict both the distribution and severity of Schistosoma haematobium. Growing degree days are defined as the number of degrees centigrade over the minimum temperature required for development. Vaccination programs are designed to protect an animal from infection, however, depending upon the age and health of the animal vaccination may not stimulate a protective humoral response. It is possible that, as in the human and mouse models, geriatric equines may be less responsive than their younger counterparts to current vaccination protocols. The purpose of this study was to identify an age related diminution in the primary and secondary immune responses of geriatric horses in response to vaccination. Two groups of horses were sampled.
