Retinal dopaminergic amacrine neurons (DA neurons) play a central role in reconfiguring retinal function according to prevailing illumination conditions, yet the mechanisms by which light regulates their activity are poorly understood. We investigated the means by which sustained light responses are evoked in DA neurons. Sustained light responses were driven by cationic currents and persisted in vitro and in vivo in the presence of L-AP4, a blocker of retinal ON-bipolar cells. Several characteristics of these L-AP4-resistant light responses suggested that they were driven by melanopsin-expressing intrinsically photosensitive retinal ganglion cells (ipRGCs), including long latencies, marked poststimulus persistence, and a peak …

Thioredoxin (TRx) is known to control redox homeostasis in cells. In recent years, a specific TRx binding protein called thioredoxin binding protein-2 (TBP-2) was found in other cell types and it appeared to negatively regulate TRx bioavailability and thereby control TRx biological function. In view of the sensitivity of lens transparency to redox status, proper regulation of TRx bioavailability is of the utmost importance. This study was conducted to examine the presence and function of TBP-2 in human lens epithelial cells (HLE B3). Porcine reproductive and respiratory syndrome virus (PRRSV) is an economically significant agent for which there currently are no effective treatments. Development of antiviral agents for PRRSV as well as many other viruses has been limited by toxicity of known antiviral compounds. In contrast, antibiotics for non-virus microbial infections have been widely useful, in part because of their acceptable toxicity in animals. We report here the discovery that the quinolone containing compound Plasmocin™, as well as the quinolones nalidixic acid and ciprofl oxacin, have potent anti-PRRSV activity in vitro. West Nile virus (WNV) was first isolated in 1937 from a febrile Ugandan woman. WNV now has a worldwide distribution from Australia and India in the east to Europe and the United States in the west. The first human cases of WNV in the United States were detected in New York in 1999. The North American isolates were also found to be highly neuroinvasive and neurovirulent and in many cases leading to mortality or permanent CNS sequelae among humans. Heme oxygenase-1 (HO-1) catalyzes the initial and rate-limiting reaction in heme catabolism (yielding equimolar quantities of iron, carbon monoxide, and biliverdin), which is further metabolized to bilirubin by biliverdin reductase. HO-1, an important enzyme with antioxidant and cytoprotective properties, functions against oxidative stress generated in response to heme and to various stress stimuli, such as zinc. The cytoprotective function of HO-1 is dependent on both the activities of the reaction products and on the inducibility of the heme oxygenase gene (hmox-1), which is regulated primarily at the level of gene transcription by two distal enhancers, E1 and E2. HSV-1 acquires its final envelope by budding into cytoplasmic vesicles thought to be derived from Trans-Golgi Network (TGN) membranes. This process is facilitated by interactions among the carboxyl termini of viral glycoproteins and tegument proteins. To investigate the relative importance of different viral glycoproteins in cytoplasmic virion morphogenesis, a set of recombinant viruses were constructed silencing expression of the glycoprotein E (ΔgE), the carboxyl terminus of glycoprotein D (gDΔcp), and the membrane protein UL20 (ΔUL20). In addition, recombinant viruses were constructed having the ΔgE+gDΔcp, and the ΔgE+ΔgM (glycoprotein M) deletions. A conditional silencing system using anti-gB siRNAs was devised to investigate the structure and function of the KSHV gB. Transient co-transfection of plasmids constitutively expressing gB and anti-gB siRNAs in 293 cells substantially inhibited gB mRNA levels and protein production. Similarly, transient expression of siRNAs into the basal cavity-based lymphoma cells (BCBL-1) caused substantial reduction of gB transcription and protein synthesis. TaqMan real-time PCR and infectivity assays showed that gB was essential for virion egress and infectivity. Transfection of a codon-optimized gB not recognized by the anti-gB siRNAs, efficiently rescued virion egress and infectivity in BCBL-1 cells in the presence …

Oncolytic herpes simplex virus type-1 (HSV-1) has shown great potential as an effective agent for cancer therapy ¨C oncolytic virotherapy. To enhance oncolytic capabilities of previously reported agents, new oncolytic and fusogenic HSV-1 OncSyn and OncdSyn viruses were constructed based on wild type HSV-1 (F) strain. To provide for safety and tumor selectivity, the viruses carried a large deletion including one of the two ¦Ã134.5 genes. The ¦Ã134.5 gene, a major neurovirulence factor, was replaced by a gene cassette constitutively expressing the red fluorescent protein.
