Surveillance Of A Novel Vesivirus Discovered In An Urban Mephitis Mephitis Population, Amanda E. Diggins

Honors Theses and Capstones

Caliciviridae is a family of non-enveloped, positive-sense, single stranded RNA viruses, including eleven defined genera that contain important pathogens of humans and animals. Within the Caliciviridae family, the genus Vesiviridae includes feline calicivirus, a leading cause of upper respiratory disease in cats, and vesicular exanthema of swine virus, a historical disease of domestic swine that has now been implicated in several marine mammal vesicular diseases, one of which has been confirmed zoonotic. Urban landscapes create a unique environment for rapid intra- and interspecies transmission of viruses. Clustered resources attract high-density populations of synanthropic wildlife species, increasing spatial overlap and heightening …

Proteomic Comparison Of Canine Gallbladder Versus Hepatic Neuroendocrine Neoplasms, Nadia Richmond Apr 2025

Proteomic Comparison Of Canine Gallbladder Versus Hepatic Neuroendocrine Neoplasms, Nadia Richmond

LSU Master's Theses

Hepatobiliary neuroendocrine neoplasms (HBNENs) are uncommon tumors in people and dogs, and the limited understanding of their biologic behavior often poses a diagnostic and prognostic challenge in the clinical setting. Though only a low number of cases have been described in dogs, there is evidence to indicate that primary gallbladder NENs may carry a more favorable prognosis compared to primary hepatic NENs. The emergence of liquid chromatography-mass spectrometry (LC-MS) in proteomic analysis has permitted large-scale analysis of neoplastic proteomes, allowing for better understanding of tumor pathophysiology while simultaneously enabling the rapid identification of protein biomarkers. Previous work in our laboratory …

Anthelmintic Resistance In Sheep Across Ireland And The Uk: A Literature Review Of The In Vivo Versus In Vitro Methods, Megan Tumulty Dec 2022

Anthelmintic Resistance In Sheep Across Ireland And The Uk: A Literature Review Of The In Vivo Versus In Vitro Methods, Megan Tumulty

SURE Journal: Science Undergraduate Research Experience Journal

Background: The animal welfare and production consequences of gastrointestinal nematode infections, namely Nematodirus battus, on sheep are compelling. The control of gastrointestinal infections has been heavily reliant on the administration of anthelmintics since their introduction into mainstream use in 1960. However, their frequent and often excessive use has resulted in anthelmintic resistance being reported extensively in several gastrointestinal nematode species. The prevalence of anthelmintic resistance as well as the increase in the number of cases of multiple-drug resistance in nematode populations now poses substantial hindrance on the viability of sheep production systems. However, despite the threat N. battus poses …

Occurrence Of Pulmonary Arteritis With Lungworm Infections In Gray Seals From New England, 2013-2020, Jillian R. Broadhurst, Inga Sidor Dvm, Ms, Dacvp

Honors Theses and Capstones

Gray seals (Halichoerus grypus) are a common pinniped species in New England coastal waters. Parasitic infection is an important cause of mortality for this species, and lungworm species are frequently implicated in cases of respiratory infection, especially in pups and weanling seals. The most common of these is Otostrongylus circumlitus, a large nematode found in the bronchi and bronchioles of affected seals. The full life cycle is unknown but its presence in lungs and sometimes pulmonary arteries suggests that migration via arterial circulation occurs. Signs and microscopic findings are widely variable; our cases included dyspnea, bloody mucoid …

Evaluation Of Oncolytic And Immunomodulatory Potential Of The Hsv-1 Live-Attenuated Vaccine Strain Vc2 In An Immunocompetent Murine Melanoma Model, Natalie Wall Fowlkes

LSU Doctoral Dissertations

Melanoma accounts for 90% of skin cancer-related deaths in humans. Treatment options for metastatic melanoma in people is very limited. Melanoma is considered to be an immunogenic tumor, spurring interest in development of immunotherapies for the treatment of metastatic melanoma. Oncolytic virotherapy has been widely investigated. The first ever oncolytic virotherapy to receive FDA-approval is an HSV-1-based virus (Talimogene Laherperavec (T-Vec) or Imlygic) containing a transgene for human GM-CSF to enhance anti-tumor immune responses after injection. Durable response rate in human patients was only 16% despite impressive efficacy in anti-tumor effects in vitro and in murine tumor models. Novel viruses …

In Vitro Assay Development As An Alternative To The Use Of Laboratory Animals For Measuring Clostridium Perfingens Type C Toxoid, Lauren M. Wilmes

School of Veterinary and Biomedical Sciences: Dissertations, Theses, and Student Research

Clostridium perfringens type C produces beta toxin, which is a primary virulence determinate that can cause necrotic enteritis in neonatal animals. Vaccines directed against C. perfringens toxins have shown to be efficacious in preventing disease. However, the production of a commercial vaccine requires not only an efficacious antigen, but also testing methods for quantifying antigens and potency on final product. This research was undertaken to fulfill a need for the reduction in small animal usage to produce and deliver vaccines. The current testing methods are performed in vivo according to the Code of Federal Regulations number 9 (9CFR) United …

Cellular And Molecular Basis Of Equine Arteritis Virus Persistent Infection In The Stallion Reproductive Tract: Characterization Of Local Host-Pathogen Interactions Mediating Long-Term Viral Persistence, Mariano Carossino Jan 2018

Cellular And Molecular Basis Of Equine Arteritis Virus Persistent Infection In The Stallion Reproductive Tract: Characterization Of Local Host-Pathogen Interactions Mediating Long-Term Viral Persistence, Mariano Carossino

Theses and Dissertations--Veterinary Science

Equine arteritis virus (EAV) has a global impact on the equine industry being the causative agent of equine viral arteritis (EVA), a reproductive, respiratory, and systemic disease of equids. A distinctive feature of EAV infection is that it establishes long-term persistent infection in the reproductive tract of stallions and is continuously shed in the semen (carrier state). Recent studies showed that long-term persistence is associated with a specific allele of the CXCL16 gene (CXCL16S). However, the cellular and molecular mechanisms underlying the establishment and maintenance of persistent infection are yet to be determined. The studies were undertaken herein …

Dnmt3a Haploinsufficiency Provokes Hematologic Malignancy Of B-Lymphoid, T-Lymphoid, And Myeloid Lineage In Mice, Garland Michael Upchurch

Theses & Dissertations

DNA methyltransferase 3A (DNMT3A) is a master epigenetic regulator of benign and malignant hematopoiesis. To dissect the biological consequences of homozygous and heterozygous Dnmt3a inactivation in malignant hematopoiesis, we generated Dnmt3a homozygous null (Dnmt3aΔ/Δ) and Dnmt3a heterozygous (Dnmt3a+/) mice and compared the presentations of hematologic malignancies between cohorts. Bi-allelic inactivation of Dnmt3a results in the presentation of mature lymphoid neoplasms resembling chronic lymphocytic leukemia (CLL; B220+CD19+CD5+; 88% penetrance (37/42)) and CD8+ peripheral T-cell lymphoma (PTCL; TCRβ+CD3+CD8+CD4; 40% penetrance (17/42)). …

Carbon metabolism and virulence determinant production are often linked in pathogenic bacteria, and several regulatory elements have been reported to mediate this linkage in Staphylococcus aureus. Previously, we described a novel protein, catabolite control protein E (CcpE) that functions as a regulator of the tricarboxylic acid cycle. Here we demonstrate that CcpE also regulates virulence determinant biosynthesis and pathogenesis. Specifically, deletion of ccpE in S. aureus strain Newman revealed that CcpE affects transcription of virulence factors such as capA, the first gene in the capsule biosynthetic operon; hla, encoding α-toxin; and psmα, encoding the phenol-soluble modulin cluster α. Electrophoretic mobility …

Of the Orthomyxoviridae family of viruses, only influenza A viruses are thought to exist as multiple subtypes and has nonhuman maintenance hosts. In April 2011, nasal swabs were collected for virus isolation from pigs exhibiting influenza-like illness. Subsequent electron microscopic, biochemical, and genetic studies identified an orthomyxovirus with seven RNA segments exhibiting approximately 50% overall amino acid identity to human influenza C virus. Based on its genetic organizational similarities to influenza C viruses this virus has been provisionally designated C/Oklahoma/1334/2011 (C/OK). Phylogenetic analysis of the predicted viral proteins found that the divergence between C/OK and human influenza C viruses was …

Development Of Repressible Systems To Control Gene Expression In Vaccinia Virus, Allison Titong Aug 2012

Development Of Repressible Systems To Control Gene Expression In Vaccinia Virus, Allison Titong

Master's Theses

Two vaccinia virus (VACV) expression systems that contain elements from the lactose (lac) and the tetracycline (tet) operons of E. coli were developed to repress the expression of a reporter gene, enhanced green fluorescent protein (EGFP), in the presence of tet operon inducers. In the first system, lac and tet operon elements were arranged in a gene circuit, and in the presence of increasing concentrations of a lac operon inducer (isopropyl-β-D-thiogalactoside, IPTG), EGFP expression increased in a dose dependent manner and at high IPTG concentrations, expression reached the same levels as a positive control virus. Importantly, in the …

Female B10.S mice are highly resistant to proteolipid protein (PLP) 139–151-induced experimental autoimmune encephalomyelitis (EAE) and depletion of PLP 139–151- reactive CD4+CD25+regulatory T (Treg) cells can slightly increase their EAE susceptibility. Although male B10.S mice are moderately susceptible to EAE, we report that depletion of Treg cells in male B10.S mice before immunization with PLP 139–151 renders them highly susceptible to severe EAE with more CNS neutrophil infiltrates than nondepleted controls. Increased susceptibility is associated with an enhanced PLP 139–151-specific T cell response and greater production of IFN-γ, IL-6, and IL-17. Male CD4+CD25+ effector cells depleted of Treg cells proliferate …

Future management of sheep flocks to minimise fly strike may require recognition of the differences between blowfly species. This article gives a description of the primary species - those which can initiate strikes.

The Pathophysiology Of Acute Venous Occlusion, Lewis H. Curd Jr. Jul 1976

The Pathophysiology Of Acute Venous Occlusion, Lewis H. Curd Jr.

Biological Sciences Theses & Dissertations

Large artery, pedal small vein, femoral vein and central venous pressures and extremity volume changes were recorded in mongrel dogs in which the hindlimb was perfused at a constant flow. Arterial, pedal venous and femoral venous blood gases were drawn and analyzed for measurements of arterio-venous O2 and CO2 differences. Paw and thigh resistances were calculated. Measurements were repeated following ligation of the iliac and femoral veins, alpha receptor blockade (dibenzyline), and dilation with Vasodilan. Results indicate that ligation of extremity veins results in an active ·increase in total limb resistance below the ligation, an increase in the …
