The antiprogestin, mifepristone, has previously been shown to noncompetitively inhibit estrogen-induced endometrial proliferation in nonhuman primates (van Uem et al., 1989; Wolf et al., 1989b; Neulen et al., 1990; Neulen et al., 1996). For both economical and ethical reasons, we are encouraged to identify comparative laboratory rodent models which can substitute the need to use primate models. In the following study, we compared capabilities of the rat uterine weight bioassay versus a primate uterine bioassay, to identify the noncompetitive antiestrogenic/antiproliferative effects of mifepristone.

Apoptosis, a form of physiological cell death, has been found to occur during regression of the corpus luteum (Juengel etal, 1993; Dharmarajan etal, 1994). The pathways involved in this process, however, have yet to be specified. One possible mediator of corpus luteum regression is the Fas (or AP0-1 or CD95) receptor, a transmembrane protein which induces apoptosis in the cell when ligated. In order to further confirm this hypolhesis, the present study establishes and quantitates the presence and regulation of Fas receptor and Fas ligand (Fasl) in the rat corpus luteum during pregnancy and post-partum. The regional distribution of ChAT activity in human spermatozoa is altered during in vitro capacitation and it correlates with the fertilizing potential of sperm.
Regional immunoreactivity in human spermatozoa as assessed by fluorescent immunocytochemistry was compared with ChAT and CaAT activity determined by enzymatic methodologies. Increasing proportions of sperm exhibited ChAT immunoreactivity along the equatorial region with a concomitant decrease in ChAT reactivity in the midpiece. Also, competitive studies with unlabeled ChAT blocked the equatorial region labeling; the unlabeled CaAT blocked staining along the midpiece region of the tail, suggesting some cross-reactivity of the ChAT antiserum with the CaAT enzyme. …

These studies were designed to characterize the relative roles of angiotensin II (ANG II) and pressure in the structural alterations that occur in experimental hypertension. Two separate studies were performed in order to differentiate these two mechanisms. First, male Wistar rats were subjected to one-kidney, one-clip (1K1C) hypertension or sham operation. Losartan, an AT1 receptor antagonist, was administered chronically to half of each surgical group beginning one day before the operation and continued until the end of the study. Autoimmune diseases are a worldwide healthcare concern. Research has focused mainly on the role of the immune system and genetic susceptibility to autoimmune disease. However, this study investigates altered target tissue properties and their contributions to the pathogenesis of induced autoimmune thyroiditis in Lewis rats. Our hypothesis was; inflammation-induced lymphocytic infiltration and subsequent fibrosis results in physical disruption of gap junctional communication in the target tissue contributing to the disease process. The purpose of this study was to precisely locate, in living humans, a myofascial trigger point associated with the upper portion of the trapezius muscle (TrP1) that refers pain to the head and neck and to determine if this point is associated with anatomical structures. This study is descriptive and utilizes data from measurements of the location of TrP1 in relation to anatomical landmarks, of pressure sensitivity overlying the trigger point and electromyography recordings in localizing the trigger point. The gastrointestinal tracts, lungs, and urinary bladders from 77 Bufo microscaphus, 61 Bufo woodhousii, and 8 of their hybrids were examined for helminths. One species of trematode (Glypthelmins quieta), 1 species of cestode (Distoichometra bufonis), and 5 species of nematodes (Aplectana incerta, A. itzocanensis, Rhabdias americanus, Physaloptera sp., and Physocephalus sp.) were found. The greatest prevalence (41%) and mean intensity (231.7) were recorded for Aplectana incerta in Bufo woodhousii. It appears hybrids harbor fewer parasites than either parent species.

An endogenous circadian biological clock controls the temporal aspects of life in most organisms, including rhythmic control of genes involved in clock output pathways. In the fungus Neurospora crassa, one pathway known to be under control of the clock is asexual spore (conidia) development. To understand more fully the processes that are regulated by the N. crassa circadian clock, systematic screens were carried out for genes that oscillate at the transcriptional level. We studied myocardial contractility in fetal sheep exposed to ~112 days of hypoxia at high-altitude (3,820 m). We recorded left and right ventricular wet weight, and measured the inotropic responses to extracellular calcium [Ca2+]o (0.2-10 mM), ryanodine (10-10-10-4 M), isoproterenol (10-10-10-6 M), and forskolin (0.01-10 uM) in isometrically contracting papillary muscles. In addition, we quantified dihydropyridine (DHPR), ryanodine (RyR), and β-adrenergic receptor densities, and measured basal and stimulated intracellular cAMP levels. Endothelial dysfunction has been found prior to any signs of atherosclerotic plaque formation in patients with a variety of risk factors for atherosclerosis. Elevated serum cholesterol has repeatedly been associated with endothelial dysfunction as demonstrated by impaired vasoreactivity to vasodilating stimuli.
The aim of this research was to determine whether short term lowering of serum cholesterol levels by life style changes or by cholesterol lowering medication could reverse or reduce the endothelial dysfunction. The inhibitory effects of the two naturally occuning isothiocyanates, benzyl isothiocyanate (BITC) and phenethyl isothiocyanate (PEITC) on the metabolism and mutagenicity of 4-(methylnitrosamino)-l-(3-pyridyl)-lbutanone (NNK) and benzo[a]pyrene (BaP) by F344 rat and male Syrian golden hamster liver microsomes were investigated. This article describes a new angle-independent method suitable for three-dimensional (3-D) blood flow velocity measurement that tracks features of the ultrasonic speckle produced by a pulse echo system. In this method, a feature is identified and followed over time to detect motion. Other blood flow velocity measurement methods typically estimate velocity using one- (1-D) or two-dimensional (2-D) spatial and time information. Speckle decorrelation due to motion in the elevation dimension may hinder this estimate of the true 3-D blood flow velocity vector. The western spruce budworm (Choristoneura occidentalis Freeman) prefers to feed on flushing buds and current-year needles of Douglas-fir (Pseudotsuga menziesii [Mirb.] Franco). Budworm larvae will not typically consume older age classes of needles unless all current-year foliage is depleted. We tested the following null hypotheses: (1) budworm larvae can feed on foliage with a wide range of qualities (i.e., current 1-, 2-, or 3-year-old needles) without measurable effects on fitness; and (2) budworm adults do not show any oviposition preference linked to the age of the foliage they fed on as larvae. Congress is currently evaluating the wilderness status of Bureau of Land Management (BLM) public lands in Utah. Wilderness areas play many important roles, and one critical role is the conservation of biological diversity. We propose that objectives for conserving biodiversity on BLM lands in Utah be to (1) ensure the long-term population viability of native animal and plant species, (2) maintain the critical ecological and evolutionary processes upon which these species depend, and (3) preserve the full range of communities, successional stages, and environmental gradients. Pressure distribution under the foot has been considered a useful indicator in the treatment of foot problems. The purpose of this study was to determine the relationship between foot types and pressure patterns under the forefoot. Fifty-four healthy subjects (19 males, 35 females, mean age=27 years, S0=4, range=21-39) participated in this study. A BTE pedobarograph was used to record pressure distribution under the foot during barefoot walking. Foot types were classified as neutral, pronated, and supinated. Pressure distribution under the forefoot was classified as medial, central, and lateral patterns. The medial pattern had the highest peak pressure under the first …

The osteoinductive potential (osteoinductivity) is a very important aspect of demineralized bone matrix (DBM) as a biomaterial. This study is directed at establishing qualitative and quantitative assays of the osteoinductivity of human DBM. Further aspects of this study include an assessment of the effects of the production process, matrix characterization, and donor related factors on osteoinductivity of the DBM.
Athymic mice were chosen as the animal model for use in the in vivo bioassay. Implantation of DBM in muscle pouches provided for much higher osteoinductivity (remineralization to 10% calcium) than subcutaneous site implantation (remineralization to 1.6% calcium). The purpose of this investigation was to determine if gender played a factor in perception during physical exertion, and whether those perceptions were influenced by conditioning level. Sixteen male and sixteen female volunteer subjects, ranging in age from 21 to 35, constituted the sample for this study. The Bruce protocol for a Symptom-Limited Graded Exercise test was the instrument utilized to evaluate heart rate during the maximal physical exercise program. A ventilatory analyzer measured each subject's maximal oxygen consumption. Borg's Rated Perceived Exertion Scale was employed for subjective evaluation of an individual's perception of effort. Muscle atrophy and decreased strength are an accepted part of aging. While physical inactivity may account for some of these losses, physically active individuals still experience age related reduction in muscle mass and strength. The purpose of this study is to examine the effects that aging has on skeletal muscle regeneration. It is hypothesized that the recovery of muscle mass and force following muscular injury will decrease with age. Planktotrophy is a relatively common developmental mode among bathyal and abyssal echinoderms, but the sources of food used by deep-sea planktotrophic larvae remain generally unknown. Very few deep-sea echinoderm larvae have been collected in plankton samples, so we do not know whether larvae migrate to the euphotic zone to feed or if they rely on bacteria or detritus at greater depths. We approached this question indirectly by investigating whether larvae of bathyal echinoids can tolerate the temperatures they would encounter in the euphotic zone and whether they possess sufficient energy stores to migrate to the euphotic zone without feeding. The catabolism of amino acids derived from dietary protein results in the production of ammonia and bicarbonate ion, waste products that an animal must eliminate. Often, animals excrete a nitrogenous waste product that is correlated with water availability: Freshwater fish, living where water is abundant, usually excrete ammonia while terrestrial animals excrete urea or uric acid. Recent studies report that fish inhabiting alkaline waters (pH 10) characterized by high bicarbonate concentrations do not excrete ammonia, instead synthesize urea, an energetically expensive process. The purpose of this study was to determine the effect of supramaximal exercise intensity on the maximal accumulated oxygen deficit during uphill treadmill running. The accumulated oxygen deficit is the difference between the predicted oxygen demand and the actual oxygen utilization, and was determined during three different supramaximal exercises. A group of eight well-trained female athletes (age= 19.6 ± 1.1 year; body weight= 55.5 ± 5.0 kg; peak oxygen uptake= 56.26 ± 5.55 ml/kg/min) performed three exercises at intensities that corresponded to 110%, 125%, and 140% of peak oxygen uptake (V02peak). Transport And Metabolism Of Alanine And Palmitic Acid By Field-Collected Larvae Of Tedania Ignis (Porifera, Demospongiae): Estimated Consequences Of Limited Label Translocation, William B. Jaeckle

The epidermis of larvae of Tedania ignis(Porifera, Demospongiae) is uniformly ciliated except for the posterior pole. The epidermal cells are long, columnar, and monociliate; each cilium arises from an epidermal crypt; symbiotic bacteria were not observed in larval cells. These lecithotrophic ("nonfeeding") larvae can feed by assimilating dissolved organic materials (DOM) from seawater. Larvae transported both the amino acid alanine (mean = 2.73 pmol larva-1 h-1; [S] = 1 µM) and the fatty acid palmitic acid (mean = 16.27 pmol larva-1 h-1; [S] = 1 µM) from seawater. Hypoxic and hypoxic/ischemic episodes contribute to various neurological aberrations, learning disabilities, et cetera in surviving infants. Despite major advances in understanding the pathophysiology of hypoxia, the cellular basis for hypoxic-mediated injury remains poorly understood. In addition, the role of developmentally essential enzymes vis á vis their contributions, or lack thereof, to hypoxic-mediated injury has not been adequately investigated. This study evaluated candidate carriers for recombinant human bone morphogenic protein-2 (rhBMP-2) in periodontal reconstructive surgery.
Canine demineralized bone matrix (DBM), bovine inorganic crystallene bone matrix (Bio-Oss), bovine derived microfibrillar collagen matrix (Helistat), poly (D,L-lactide-coglycolide) micropaiticles (PLGA), and polylactic acid granules (Drilac) were used with rhBMP-2 (20 (µg/100 µl implant volume) in routine critical size canine supraalveolar periodontal defects. Contralateral defects in six beagle dogs were randomly assigned to receive: DBM/rhBMP-2, DBM-control, Bio-Oss/rhBMP-2, Helistat/rhBMP-2, PLGA/rhBMP-2, or Drilac/rhBMP-2, all with autologous blood except for Helistat and PLGA. Muscle contraction is thought to be accomplished by sliding of myosin filaments along actin filaments. Although actomyosin naturally uses ATP as an energy source, it can also use other nucleoside triphosphates (NTP) as substrates. In this work, the dependence of rate and equilibrium constants of the various steps in the muscle contraction mechanism upon nucleotide structure was investigated to unravel the dependence of the mechanical properties of the muscle upon changes in different biochemical steps of the mechanism.
